    Serological Investigations of Brucellosis in Cattle, Farmers and Veterinarians in the Kars District of Turkey

    The prevalence of brucellosis was investigated in cattle, farmers and veterinarians in the Kars district of Turkey between 2004 - 2006. In order to achieve this, a total of 407 serum samples of cattle from 27 herds having history of abortions were examined for Brucella antibodies by RBPT and SAT. In addition, the sera collected from 246 farmers (130 males and 116 females) and 28 veterinarians in the same district were analysed serologically by RBPT, SAT and ELISA. Of the cattle sera analysed, 134 (32.92%) and 141 (34.64%) were determined as positive by RBPT and SAT, respectively. Thirty-two (13%), 35 (14.22%) and 44 (17.88%) of the farmers' sera were found positive for brucellosis by RBPT, SAT and ELISA, respectively. There was no significant difference between sexes for Brucella seropositivity. Of the 28 sera from veterinarians, 13 (46.42%) were positive by the three serological tests. The high prevalence of brucellosis both in cattle and humans suggests that brucellosis is common in this area. Preventive and control measures should be implemented and pursued more strictly to reduce and/or eradicate brucellosis from the area

    Quantum singularities in a model of f(R) Gravity

    The formation of a naked singularity in a model of f(R) gravity having as source a linear electromagnetic field is considered in view of quantum mechanics. Quantum test fields obeying the Klein-Gordon, Dirac and Maxwell equations are used to probe the classical timelike naked singularity developed at r=0. We prove that the spatial derivative operator of the fields fails to be essentially self-adjoint. As a result, the classical timelike naked singularity remains quantum mechanically singular when it is probed with quantum fields having different spin structures.Comment: 12 pages, final version.     Boron Stress Responsive MicroRNAs and Their Targets in Barley

    Boron stress is an environmental factor affecting plant development and production. Recently, microRNAs (miRNAs) have been found to be involved in several plant processes such as growth regulation and stress responses. In this study, miRNAs associated with boron stress were identified and characterized in barley. miRNA profiles were also comparatively analyzed between root and leave samples. A total of 31 known and 3 new miRNAs were identified in barley; 25 of them were found to respond to boron treatment. Several miRNAs were expressed in a tissue specific manner; for example, miR156d, miR171a, miR397, and miR444a were only detected in leaves. Additionally, a total of 934 barley transcripts were found to be specifically targeted and degraded by miRNAs. In silico analysis of miRNA target genes demonstrated that many miRNA targets are conserved transcription factors such as Squamosa promoter-binding protein, Auxin response factor (ARF), and the MYB transcription factor family. A majority of these targets were responsible for plant growth and response to environmental changes. We also propose that some of the miRNAs in barley such as miRNA408 might play critical roles against boron exposure.     Key and Smart Actions to Alleviate Hunger and Poverty Through Irrigation and Drainage

    In the pursuit of information to support policies and actions to alleviate hunger and poverty through irrigation and drainage, this paper attempts to provide correlations between water scarcity, communities and poverty. Many reviews have found strong direct and indirect relationships between irrigation and poverty. One of the main goals of the international community is to eliminate hunger and poverty and in this perspective, through the Millennium Development Goals, much progress has been achieved and evidence obtained. Sustainable Development Goals and various other United Nations initiatives intend to move forward this agenda by making it a part of broader development frameworks. In this paper, the important elements of irrigation and drainage that affect the alleviation of hunger and poverty are discussed. These elements are grouped into governance, rights-based developments, water rights and pricing, management, efficiency improvement, and the role of technology. Both the potential and the need for innovative technology and solutions in irrigation are underlined, which can be used to cater for the challenges in different subsectors. The main focus of these solutions is on maximizing productivity and efficiency, reducing water losses, achieving sustainable intensification and managing demands on water resources and the associated trade-offs

    A Comprehensive Approach to Identify Reliable Reference Gene Candidates to Investigate the Link between Alcoholism and Endocrinology in Sprague-Dawley Rats

    Gender and hormonal differences are often correlated with alcohol dependence and related complications like addiction and breast cancer. Estrogen (E2) is an important sex hormone because it serves as a key protein involved in organism level signaling pathways. Alcoholism has been reported to affect estrogen receptor signaling; however, identifying the players involved in such multi-faceted syndrome is complex and requires an interdisciplinary approach. In many situations, preliminary investigations included a straight forward, yet informative biotechniques such as gene expression analyses using quantitative real time PCR (qRT-PCR). The validity of qRT-PCR-based conclusions is affected by the choice of reliable internal controls. With this in mind, we compiled a list of 15 commonly used housekeeping genes (HKGs) as potential reference gene candidates in rat biological models. A comprehensive comparison among 5 statistical approaches (geNorm, dCt method, NormFinder, BestKeeper, and RefFinder) was performed to identify the minimal number as well the most stable reference genes required for reliable normalization in experimental rat groups that comprised sham operated (SO), ovariectomized rats in the absence (OVX) or presence of E2 (OVXE2). These rat groups were subdivided into subgroups that received alcohol in liquid diet or isocalroic control liquid diet for 12 weeks. Our results showed that U87, 5S rRNA, GAPDH, and U5a were the most reliable gene candidates for reference genes in heart and brain tissue. However, different gene stability ranking was specific for each tissue input combination. The present preliminary findings highlight the variability in reference gene rankings across different experimental conditions and analytic methods and constitute a fundamental step for gene expression assays

    A computational-based update on microRNAs and their targets in barley (Hordeum vulgare L.)

    <p>Abstract</p> <p>Background</p> <p>Many plant species have been investigated in the last years for the identification and characterization of the corresponding miRNAs, nevertheless extensive studies are not yet available on barley (at the time of this writing). To extend and to update information on miRNAs and their targets in barley and to identify candidate polymorphisms at miRNA target sites, the features of previously known plant miRNAs have been used to systematically search for barley miRNA homologues and targets in the publicly available ESTs database. Matching sequences have then been related to Unigene clusters on which most of this study was based.</p> <p>Results</p> <p>One hundred-fifty-six microRNA mature sequences belonging to 50 miRNA families have been found to significantly match at least one EST sequence in barley. As expected on the basis of phylogenetic relations, miRNAs putatively orthologous to those of <it>Triticum </it>are significantly over-represented inside the set of identified barley microRNA mature sequences. Many previously known and several putatively new miRNA/target pairs have been identified. When the predicted microRNA targets were grouped into functional categories, biological processes previously known to be regulated by miRNAs, such as development and response to biotic and abiotic stress, have been highlighted and most of the target molecular functions were related to transcription regulation. Candidate microRNA coding genes have been reported and genetic variation (SNPs/indels) both in functional regions of putative miRNAs (mature sequence) and at miRNA target sites has been found.</p> <p>Conclusions</p> <p>This study has provided an update of the information on barley miRNAs and their targets representing a foundation for future studies. Many of previously known plant microRNAs have homologues in barley with expected important roles during development, nutrient deprivation, biotic and abiotic stress response and other important physiological processes. Putative polymorphisms at miRNA target sites have been identified and they can represent an interesting source for the identification of functional genetic variability.</p
