    A Systematic Multidisciplinary Process for User Engagement and Sensor Evaluation: Development of a Digital Toolkit for Assessment of Movement in Children with Cerebral Palsy

    Objectives: To describe and critique a systematic multidisciplinary approach to user engagement, and selection and evaluation of sensor technologies for development of a sensor-based Digital Toolkit for assessment of movement in children with cerebral palsy (CP).Methods: A sequential process was employed comprising three steps: Step 1: define user requirements, by identifying domains of interest; Step 2: map domains of interest to potential sensor technologies; and Step 3: evaluate and select appropriate sensors to be incorporated into the Digital Toolkit. The process employed a combination of principles from frameworks based in either healthcare or technology design.Results: A broad range of domains were ranked as important by clinicians, patients and families, and industry users. These directly informed the device selection and evaluation process that resulted in three sensor-based technologies being agreed for inclusion in the Digital Toolkit, for use in a future research study.Conclusion: This report demonstrates a systematic approach to user engagement and device selection and evaluation during the development of a sensor-based solution to a healthcare problem. It also provides a narrative on the benefits of employing a multidisciplinary approach throughout the process. This work uses previous frameworks for evaluating sensor technologies and expands on the methods used for user engagement

    Dual G9A/EZH2 inhibition stimulates anti-tumour immune response in ovarian high-grade serous carcinoma

    Ovarian high-grade serous carcinoma (HGSC) prognosis correlates directly with presence of intratumoral lymphocytes. However, cancer immunotherapy has yet to achieve meaningful survival benefit in patients with HGSC. Epigenetic silencing of immunostimulatory genes is implicated in immune evasion in HGSC and re-expression of these genes could promote tumour immune clearance. We discovered that simultaneous inhibition of the histone methyltransferases G9A and EZH2 activates the CXCL10-CXCR3 axis and increases homing of intratumoral effector lymphocytes and natural killer cells whilst suppressing tumour-promoting FoxP3+ CD4 T cells. The dual G9A/EZH2 inhibitor HKMTI-1-005 induced chromatin changes that resulted in the transcriptional activation of immunostimulatory gene networks, including the re-expression of elements of the ERV-K endogenous retroviral family. Importantly, treatment with HKMTI-1-005 improved the survival of mice bearing Trp53-/- null ID8 ovarian tumours and resulted in tumour burden reduction. These results indicate that inhibiting G9A and EZH2 in ovarian cancer alters the immune microenvironment and reduces tumour growth and therefore positions dual inhibition of G9A/EZH2 as a strategy for clinical development

    Low-energy M1 and E3 excitations in the proton-rich Kr-Zr region

    Low-energy intrinsic KπK^\pi=1+^+, 0−0^-, 1−1^-, 2−2^-, and 3−3^- states in the even-even proton-rich Sr, Kr, and Zr nuclei are investigated using the quasiparticle random phase approximation. In the Z≃\simeqN nuclei the lowest-lying 1+^+ states are found to carry unusually large B(M1)B(M1) strength.     Planetary nebulae populations in the haloes of nearby massive early-type galaxies

    Planetary nebulae (PNe) are excellent tracers of the metal-poor haloes of nearby early-type galaxies. They are commonly used to trace spatial distribution and kinematics of the halo and intracluster light at distances of up to 100 Mpcs. The results on the early-type galaxy M105 in the Leo I group represent a benchmark for the quantitative analysis of halo and intragroup light. Since the Leo I group lies at just a 10 Mpc distance, it is at the ideal location to compare results from resolved stellar populations with the homogeneous constraints over a much larger field of view from the PN populations. In M105, we have -- for the first time -- established a direct link between the presence of a metal-poor halo as traced by resolved red-giant branch stars and a PN population with a high specific frequency (α\alpha-parameter).     Neurochemical changes in the developing rat hippocampus during prolonged hypoglycemia

    Hypoglycemia is common during development and is associated with the risk of neurodevelopmental deficits in human infants. The effects of hypoglycemia on the developing hippocampus are poorly understood. The sequential changes in energy substrates, amino acids and phosphocreatine were measured from the hippocampus during 180 min of insulin-induced hypoglycemia (blood glucose < 2.5 mmol/L) in 14-day-old rats using in vivo1H NMR spectroscopy. Hypoglycemia resulted in neuroglycopenia (brain glucose < 0.5 Όmol/g). However, the phosphocreatine/creatine (PCr/Cr) ratio was maintained in the physiological range until approximately 150 min of hypoglycemia, indicating that energy supply was sufficient to meet the energy demands. Lactate concentration decreased soon after the onset of neuroglycopenia. Beyond 60 min, glutamine and glutamate became the major energy substrates. A precipitous decrease in the PCr/Cr ratio, indicative of impending energy failure occurred only after significant depletion of these amino acids. Once glutamate and glutamine were significantly exhausted, aspartate became the final energy source. N-acetylaspartate concentration remained unaltered, suggesting preservation of neuronal/mitochondrial integrity during hypoglycemia. Correction of hypoglycemia normalized the PCr/Cr ratio and partially restored the amino acids to pre-hypoglycemia levels. Compensatory neurochemical changes maintain energy homeostasis during prolonged hypoglycemia in the developing hippocampus. © 2010 International Society for Neurochemistry

    NK cells augment oncolytic adenovirus cytotoxicity in ovarian cancer

    Oncolytic viruses (OVs) can trigger profound innate and adaptive immune responses, which have the potential both to potentiate and reduce the activity of OVs. Natural killer (NK) cells can mediate potent anti-viral and anti-tumoral responses, but there are no data on the role of NK cells in oncolytic adenovirus activity. Here, we have used two different oncolytic adenoviruses—the Ad5 E1A CR2-deletion mutant dl922-947 (group C) and the chimeric Ad3/Ad11p mutant enadenotucirev (group B)—to investigate the effect of NK cells on overall anti-cancer efficacy in ovarian cancer. Because human adenoviruses do not replicate in murine cells, we utilized primary human NK cells from peripheral blood and ovarian cancer ascites. Our results show that dl922-947 and enadenotucirev do not infect NK cells, but induce contact-dependent activation and anti-cancer cytotoxicity against adenovirus-infected ovarian cancer cells. Moreover, manipulation of NK receptors DNAM-1 (DNAX accessory molecule-1) and TIGIT (T cell immunoreceptor with Ig and ITIM domains) significantly influences NK cytotoxicity against adenovirus-infected cells. Together, these results indicate that NK cells act to increase the activity of oncolytic adenovirus in ovarian cancer and suggest that strategies to augment NK activity further via the blockade of inhibitory NK receptor TIGIT could enhance therapeutic potential of OVs
