    The Protein Kinase C Inhibitor Aeb071 (Sotrastaurin) Modulates Migration and Superoxide Anion Production by Human Neutrophils In Vitro

    We examined the effect of the protein kinase C-selective inhibitor AEB071 (sotrastaurin) on neutrophil functions in vitro. Pre-incubation with AEB071 at concentrations similar to those reached during in vivo therapy significantly reduced cell capacity to migrate toward three different chemo-attractants and to produce superoxide anions (O2) in response to phorbol myristate acetate (PMA) or to iV-formyl-methionyl-leucyl-phenylalanine (fMLP). AEB071 also significantly inhibited the O−2 "overproduction induced by fMLP in neutrophils primed with tumor necrosis factor alpha (TNF-α) or granulocyte/macrophage-colony stimulating factor (GM-CSF). This inhibition was not linked to fMLP-receptor down-regulation since the drug had no effect on either fMLP-receptors or fMLP-induced CD11b membrane expression. When the activity of AEB071 was compared to that of the conventional protein kinase C (PKC) inhibitor Gö6850 (which, like sotrastaurin, inhibits classical and novel PKC isoforms), Gö6976 (an inhibitor of α and β PKC isoforms) and rottlerin (a prevailing δ PKC isoform inhibitor), AEB071 at an equimolar concentration of 3 μM (close to the maximum drug concentration reached in patients treated with AEB071) caused significantly more inhibition on both chemotactic response and superoxide production. These in vitro findings suggest that neutrophils may offer a cellular target for AEB071 activity in vivo

    The Globular Cluster System of M60 (NGC 4649). I. CFHT MOS Spectroscopy and Database

    We present the measurement of radial velocities for globular clusters in M60, giant elliptical galaxy in the Virgo cluster. Target globular cluster candidates were selected using the Washington photometry based on the deep 16\arcmin \times 16\arcmin images taken at the KPNO 4m and using the VIVI photometry derived from the HST/WFPC2 archive images. The spectra of the target objects were obtained using the Multi-Object Spectrograph (MOS) at the Canada-France-Hawaii Telescope (CFHT). We have measured the radial velocity for 111 objects in the field of M60: 93 globular clusters (72 blue globular clusters with 1.0(CT1)<1.71.0\le(C-T_1)<1.7 and 21 red globular clusters with 1.7(CT1)<2.41.7\le(C-T_1)<2.4), 11 foreground stars, 6 small galaxies, and the nucleus of M60. The measured velocities of the 93 globular clusters range from 500\sim 500 km s1^{-1} to 1600\sim 1600 km s1^{-1}, with a mean value of 107025+271070_{-25}^{+27} km s1^{-1}, which is in good agreement with the velocity of the nucleus of M60 (vgal=1056v_{\rm gal}=1056 km s1^{-1}). Combining our results with data in the literature, we present a master catalog of radial velocities for 121 globular clusters in M60. The velocity dispersion of the globular clusters in the master catalog is found to be 23414+13234_{-14}^{+13} km s1^{-1} for the entire sample, 22316+13223_{-16}^{+13} km s1^{-1} for 83 blue globular clusters, and 25831+21258_{-31}^{+21} km s1^{-1} for 38 red globular clusters.Comment: 29 pages, 8 figures.     Analysis of P1 Latency in Normal Hearing and Profound Sensorineural Hearing Loss

    ObjectivesP1 is a robust positivity at a latency of 50-150 msec in the auditory evoked potential of young children. It has been reported that over the first 2-3 years of life, there is a rapid decrease of the latency and the mean P1 latency in adults with normal hearing is approximately 60 msec. This study was designed to evaluate the change of the P1 latency in Koreans with normal hearing according to age and to compare this with the P1 latency of young patients with profound sensorineural hearing loss before and/or after cochlear implantation.MethodsAmong the patients who visited the Department of Otorhinolaryngology at Seoul National University Hospital from June 2007 to September 2009, the P1 response was recorded in 53 patients in the normal hearing group, in 13 patients in the pre-cochlear implantation (CI) group and in 10 patients in the post-CI group. A synthesized consonant-vowel syllable /ba/ was used to elicit the evoked responses. The evoked responses were collected using the center of the frontal head. For each subject, an individual grand average waveform was computed by averaging the ten recordings. The P1 latency was visually identified as a robust positivity in the waveform.ResultsFor the normal hearing group, the P1 latency showed the pattern of shortening as the age increased (coefficient, -0.758; P<0.001). For the pre-CI group, 10 cases showed delayed latencies and 3 cases did not show the P1 wave. For the post-CI group, the P1 latencies showed a less delayed tendency than those of the pre-CI group, but this was not statistically different.ConclusionThis report provides the standard value of the P1 latency at each age in Koreans for the first time and the findings support that the maturation of the central auditory pathways could be measured objectively using the P1 latency

    Immune reconstitution after allogeneic hematopoietic stem cell transplantation in children: a single institution study of 59 patients

    PurposeLymphocyte subset recovery is an important factor that determines the success of hematopoietic stem cell transplantation (HSCT). Temporal differences in the recovery of lymphocyte subsets and the factors influencing this recovery are important variables that affect a patient's post-transplant immune reconstitution, and therefore require investigation.MethodsThe time taken to achieve lymphocyte subset recovery and the factors influencing this recovery were investigated in 59 children who had undergone HSCT at the Department of Pediatrics, The Catholic University of Korea Seoul St. Mary's Hospital, and who had an uneventful follow-up period of at least 1 year. Analyses were carried out at 3 and 12 months post-transplant. An additional study was performed 1 month post-transplant to evaluate natural killer (NK) cell recovery. The impact of pre- and post-transplant variables, including diagnosis of Epstein-Barr virus (EBV) DNAemia posttransplant, on lymphocyte recovery was evaluated.ResultsThe lymphocyte subsets recovered in the following order: NK cells, cytotoxic T cells, B cells, and helper T cells. At 1 month post-transplant, acute graft-versus-host disease was found to contribute significantly to the delay of CD16+/56+ cell recovery. Younger patients showed delayed recovery of both CD3+/CD8+ and CD19+ cells. EBV DNAemia had a deleterious impact on the recovery of both CD3+ and CD3+/CD4+ lymphocytes at 1 year post-transplant.ConclusionIn our pediatric allogeneic HSCT cohort, helper T cells were the last subset to recover. Younger age and EBV DNAemia had a negative impact on the post-transplant recovery of T cells and B cells

    On Classical Equivalence Between Noncritical and Einstein Gravity : The AdS/CFT Perspectives

    We find that noncritical gravity, a special class of higher derivative gravity, is classically equivalent to Einstein gravity at the full nonlinear level. We obtain the viscosity-to-entropy ratio and the second order transport coefficients of the dual fluid of noncritical gravity to all orders in the coupling of higher derivative terms. We also compute the holographic entanglement entropy in the dual CFT of noncritical gravity. All these results confirm the nonlinear equivalence between noncritical gravity and Einstein gravity at the classical level.Comment: 19 pages, no figure

    The effect of the DLTIDDSYWYRI motif of the human laminin α2 chain on implant osseointegration

    Considerable effort has been directed towards replacing lost teeth using tissue-engineering methods such as titanium implants. A number of studies have tried to modify bioinert titanium surfaces by coating them with functionally bioactive molecules for faster and stronger osseointegration than pure titanium surfaces. Recently, peptides have been recognized as valuable scientific tools in the field of tissue-engineering. The DLTIDDSYWYRI motif of the human laminin-2 α2 chain has been previously reported to promote the attachment of various cell types; however, the in vivo effects of the DLTIDDSYWYRI motif on new bone formation have not yet been studied. To examine whether a laminin-2-derived peptide can promote osseointegration by accelerating new bone formation in vivo, we applied titanium implants coated with the DLTIDDSYWYRI motif in a rabbit tibia model. The application of the DLTIDDSYWYRI motif-treated implant to tibia wounds enhanced collagen deposition and alkaline phosphatase expression. It significantly promoted implant osseointegration compared with treatment with scrambled peptide-treated implants by increasing the bone-to-implant contact ratio and bone area.     Small anisotropy of the lower critical field and s±s_\pm-wave two-gap feature in single crystal LiFeAs

    The in- and out-of-plane lower critical fields and magnetic penetration depths for LiFeAs were examined. The anisotropy ratio γHc1(0)\gamma_{H_{c1}}(0) is smaller than the expected theoretical value, and increased slightly with increasing temperature from 0.6TcT_c to TcT_c. This small degree of anisotropy was numerically confirmed by considering electron correlation effect. The temperature dependence of the penetration depths followed a power law(\simTnT^n) below 0.3TcT_c, with nn>>3.5 for both λab\lambda_{ab} and λc\lambda_c. Based on theoretical studies of iron-based superconductors, these results suggest that the superconductivity of LiFeAs can be represented by an extended s±s_\pm-wave due to weak impurity scattering effect. And the magnitudes of the two gaps were also evaluted by fitting the superfluid density for both the in- and out-of-plane to the two-gap model. The estimated values for the two gaps are consistent with the results of angle resolved photoemission spectroscopy and specific heat experiments.Comment: 10 pages, 5 figure

    Solitary osteochondroma of the twelfth rib with intraspinal extension and cord compression in a middle-aged patient

    <p>Abstract</p> <p>Background</p> <p>Osteochondroma is a disease of growing bone and thus typically presents in younger patients. It has rarely been described in middle-aged and elderly patients. Data on the occurrence of osteochondroma show that the reported incidence of costal osteochondroma is very low. Moreover, costal osteochondroma arising at the costovertebral junction with neural foraminal extension and spinal cord compression is extremely rare.</p> <p>Case presentation</p> <p>This study reports the case of a 58-year-old patient with a solitary osteochondroma of the 12th rib with intraspinal extension and spinal cord compression. The clinical history, plain radiographs, computed tomography (CT), magnetic resonance imaging, and pathologic findings of the reported patient have been reviewed. The relevant medical literature has also been reviewed. The patient was treated with surgery for complete tumour excision to avoid tumour recurrence. After surgery, the patient's symptoms improved. An additional CT scan obtained at 1 year after surgery did not show any evidence of recurrence.</p> <p>Conclusions</p> <p>This patient is the oldest patient reported to have this rare form of costal osteochondroma. The age of the patient and the erosion of the adjacent bones raised clinical suspicion of malignancy; therefore, surgical management involved complete tumour excision with thoracolumbar fixation and fusion.</p

    Noncritical Einstein-Weyl Gravity and the AdS/CFT Correspondence

    We explore four-dimensional Einstein-Weyl gravity and supergravity on anti-de Sitter spacetime. For a specific range of the coupling with appropriate boundary conditions, we show the effective equivalence of the theory with Einstein gravity and AdS supergravity at the quadratic Lagrangian level. Furthermore we show that these equivalences can be promoted to the full nonlinear level. We also show that the similar behavior holds for the generalized Gibbons-Hawking terms. From this we find that the correlation functions in the dual conformal field theory of Einstein-Weyl gravity and supergravity can be readily read off from corresponding ones from Einstein gravity and AdS supergravity.
