    Pion properties at finite density

    In this talk, we report our recent work on the pion weak decay constant (F_pi) and pion mass (m_pi) using the nonlocal chiral quark model with the finite quark-number chemical potential (mu) taken into account. Considering the breakdown of Lorentz invariance at finite density, the time and space components are computed separately, and the corresponding results turn out to be: F^t_pi = 82.96 MeV and F^s_pi = 80.29 MeV at mu_c ~ 320 MeV, respectively.     Elevated intracellular cAMP exacerbates vulnerability to oxidative stress in optic nerve head astrocytes.

    Glaucoma is characterized by a progressive loss of retinal ganglion cells and their axons, but the underlying biological basis for the accompanying neurodegeneration is not known. Accumulating evidence indicates that structural and functional abnormalities of astrocytes within the optic nerve head (ONH) have a role. However, whether the activation of cyclic adenosine 3',5'-monophosphate (cAMP) signaling pathway is associated with astrocyte dysfunction in the ONH remains unknown. We report here that the cAMP/protein kinase A (PKA) pathway is critical to ONH astrocyte dysfunction, leading to caspase-3 activation and cell death via the AKT/Bim/Bax signaling pathway. Furthermore, elevated intracellular cAMP exacerbates vulnerability to oxidative stress in ONH astrocytes, and this may contribute to axonal damage in glaucomatous neurodegeneration. Inhibition of intracellular cAMP/PKA signaling activation protects ONH astrocytes by increasing AKT phosphorylation against oxidative stress. These results strongly indicate that activation of cAMP/PKA pathway has an important role in astrocyte dysfunction, and suggest that modulating cAMP/PKA pathway has therapeutic potential for glaucomatous ONH degeneration

    Topical ocular application of aggrelyte-2A reduces lens stiffness in mice

    Presbyopia is the progressive loss of the ability of the lens to focus on nearby objects due to its increased stiffness. It occurs in the mid-40s and continues to worsen until the mid-60s. The age-associated increase in protein cross-linking in the lens leads to protein aggregation and water insolubility, especially in the nuclear region, contributing to lens stiffness. This study reports the development of aggrelyte-2A (methyl S-acetyl-N-(3,3-dimethylbutanoyl) cysteinate, a derivative of our previously reported aggrelyte-2) for reversing the stiffness of aged lenses. Aggrelyte-2A showed minimal toxicity in cultured mouse lens epithelial cells (up to 2000 ĀµM) and human lens epithelial cells (up to 250 ĀµM). Lenses from aged mice (age: 24-25 months) treated with 1 mM aggrelyte-2A for 24Ā h, and human lenses (age: 47-67 years) treated with 250 ĀµM aggrelyte-2A for 48Ā h showed 11-14% reductions in stiffness, accompanied by an increase in acetyllysine in lens proteins, and free-thiols in the lens. Topical application of aggrelyte-2A (40 mM, 5 Āµl twice daily for 4 weeks) on mouse eyes significantly reduced lens stiffness. The topical application showed no toxicity to the lens, cornea, or retina, as revealed by morphological examination, H&E staining, and optical coherence tomography. These data suggest that aggrelyte-2A could be developed as a presbyopia-reversing therapeutic

    Comparison of Endothelial Progenitor Cells in Parkinson's Disease Patients Treated with Levodopa and Levodopa/COMT Inhibitor

    BACKGROUND: Levodopa treatment in Parkinson's disease (PD) increases in serum homocysteine levels due to its metabolism via catechol O-methyltransferase. Endothelial progenitor cells (EPCs) have the capacity to differentiate into mature endothelial cells and are markers for endothelial functions and cardiovascular risks. Along with traditional vascular risk factors, hyperhomocysteinemia is known to decrease the level of EPCs. In the present study, we hypothesized that that levodopa-induced hyperhomocysteinemia leads to a change in EPC levels. METHODOLOGY/PRINCIPAL FINDINGS: We prospectively enrolled PD patients who had been prescribed either levodopa/carbidopa (PD-L group, nā€Š=ā€Š28) or levodopa/carbidopa/COMT inhibitor (PD-LC group, nā€Š=ā€Š25) for more than 1 year. The number of circulating EPCs was measured by flow cytometry using dual staining of anti-CD34 and anti-KDR antibodies. The EPCs were divided into tertiles based on their distributions and a logistic regression analysis was used to estimate independent predictors of the highest tertile of EPCs. The number of endothelial progenitor cells was significantly decreased in PD-L patients (118Ā±99/mL) compared with either PD-LC patients (269Ā±258/mL, pā€Š=ā€Š0.007) or controls (206Ā±204/mL, pā€Š=ā€Š0.012). The level of homocysteine was significantly increased in PD-L patients (14.9Ā±5.3 Āµmol/L) compared with either PD-LC patients (11.9Ā±3.0 Āµmol/L, pā€Š=ā€Š0.028) or controls (11.1Ā±2.5 Āµmol/L, pā€Š=ā€Š0.012). The level of homocysteine was negatively correlated with endothelial progenitor cell levels (rā€Š=ā€Š-0.252, pā€Š=ā€Š0.028) and was an independent predictor of the highest tertile of endothelial progenitor cell levels (OR; 0.749 [95% CI: 0.584-0.961]). CONCLUSIONS/SIGNIFICANCE: These data indicate that a higher consumption of EPC for restoration of endothelial damage may be associated with chronic levodopa treatment in PD patients

    AdS/BCFT Correspondence for Higher Curvature Gravity: An Example

    We consider the effects of higher curvature terms on a holographic dual description of boundary conformal field theory. Specifically, we consider three-dimensional gravity with a specific combination of Ricci tensor square and curvature scalar square, so called, new massive gravity. We show that a boundary entropy and an entanglement entropy are given by similar expression with those of the Einstein gravity case when we introduce an {\it effective} Newton's constant and an {\it effective} cosmological constant. We also show that the holographic g-theorem still holds in this extension, and we give some comments about the central charge dependence of boundary entropy in the holographic construction. In the same way, we consider new type black holes and comment on the boundary profile. Moreover, we reproduce these results through auxiliary field formalism in this specific higher curvature gravity.Comment: 27pages, minor corrections, accepted in JHE

    Aspirin Inhibits TGFĪ²2-Induced Epithelial to Mesenchymal Transition of Lens Epithelial Cells:Selective acetylation of K56 and K122 in histone H3

    Posterior capsule opacification (PCO) is a complication after cataract surgery that can disrupt vision. The epithelial to mesenchymal transition (EMT) of lens epithelial cells (LECs) in response to transforming growth factor Ī²2 (TGFĪ²2) has been considered an obligatory mechanism for PCO. In this study, we tested the efficacy of aspirin in inhibiting the TGFĪ²2-mediated EMT of human LECs, LECs in human lens capsular bags, and lensectomized mice. In human LECs, the levels of the EMT markers Ī±-smooth muscle actin (Ī±-SMA) and fibronectin were drastically reduced by treatment with 2ā€…mM aspirin. Aspirin also halted the EMT response of TGFĪ²2 when introduced after EMT initiation. In human capsular bags, treatment with 2ā€…mM aspirin significantly suppressed posterior capsule wrinkling and the expression Ī±-SMA in capsule-adherent LECs. The inhibition of TGFĪ²2-mediated EMT in human LECs was not dependent on Smad phosphorylation or MAPK and AKT-mediated signaling. We found that aspirin significantly increased the acetylation of K56 and K122 in histone H3 of human LECs. Chromatin immunoprecipitation assays using acetyl-H3K56 or acetyl-H3K122 antibody revealed that aspirin blocked the TGFĪ²2-induced acetylation of H3K56 and H3K122 at the promoter regions of ACTA2 and COL1A1. After lensectomy in mice, we observed an increase in the proliferation and Ī±-SMA expression of the capsule-adherent LECs, which was ameliorated by aspirin administration through drinking water. Taken together, our results showed that aspirin inhibits TGFĪ²2-mediated EMT of LECs, possibly from epigenetic down-regulation of EMT-related genes

    Holographic Renormalization and Stress Tensors in New Massive Gravity

    We obtain holographically renormalized boundary stress tensors with the emphasis on a special point in the parameter space of three dimensional new massive gravity, using the so-called Fefferman-Graham coordinates with relevant counter terms. Through the linearized equations of motion with a standard prescription, we also obtain correlators among these stress tensors. We argue that the self-consistency of holographic renormalization determines counter terms up to unphysical ambiguities. Using these renormalized stress tensors in Fefferman-Graham coordinates, we obtain the central charges of dual CFT, and mass and angular momentum of some AdSAdS black hole solutions. These results are consistent with the previous ones obtained by other methods. In this study on the Fefferman-Graham expansion of new massive gravity, some aspects of higher curvature gravity are revealed.Comment: Version accepted for publication in JHEP, conclusion revised, references adde

    Host-mediated beneficial effects of phytochemicals for prevention of avian coccidiosis

    Both in vitro and in vivo studies were conducted to evaluate the beneficial effects of green tea extract (GT), cinnamon oil (CO), and pomegranate extract (PO) on avian coccidiosis. In experiment (EXP) 1, an in vitro culture system was used to investigate the individual effects of GT, CO, and PO on the proinflammatory cytokine response and integrity of tight junction (TJ) in chicken intestinal epithelial cells (IEC), on the differentiation of quail muscle cells and primary chicken embryonic muscle cells, and anticoccidial and antibacterial activities against Eimeria tenella sporozoites and Clostridium perfringens bacteria, respectively. In EXP 2 and 3, in vivo trials were carried out to study the dose-dependent effect of blended phytochemicals (GT, CO, PO) on coccidiosis in broiler chickens infected with E. maxima. For EXP 2, one hundred male broiler chickens (0-day-old) were allocated into the following five treatment groups: Control group for non-infected chickens (NC), Basal diet group for E. maxima-infected chickens (PC), PC group supplemented with phytochemicals at 50 (Phy 50), 100 (Phy 100), and 200 (Phy 200) mg/kg feed diets for E. maxima-infected chickens. For EXP 3, one hundred twenty male broiler chickens (0-day-old) were allocated into the following six treatment groups: NC, PC, PC supplemented with phytochemicals at 10 (Phy 10), 20 (Phy 20), 30 (Phy 30), and 100 (Phy 100) mg/kg feed for E. maxima-infected chickens. Body weights (BW) were measured on days 0, 7, 14, 20, and 22, and jejunum samples were used to measure cytokine, TJ protein, and antioxidant enzyme responses at 8 days post-infection (dpi). Fecal samples for oocyst enumeration were collected from 6 to 8 dpi. In vitro, CO and PO reduced LPS-induced IL-1Ī² and IL-8 in IEC, respectively, and GT enhanced the gene expression of occludin in IEC. PO at 1.0 and 5.0 mg/mL exerted antimicrobial effect against E. tenella sporozoites and C. perfringens bacteria, respectively. In vivo, chickens fed a diet supplemented with phytochemicals showed enhanced BW, reduced oocyst shedding, and decreased proinflammatory cytokines following E. maxima challenge. In conclusion, the combination of GT, CO, and PO in the diet of broiler chickens infected with E. maxima induced enhanced host disease resistance including innate immunity and gut health, which contributed to improved growth and reduced disease responses. These findings provide scientific support for the development of a novel phytogenic feed additive formula that enhances the growth and intestinal health of broiler chickens infected with coccidiosis
