    Measuring and Correcting Wind-Induced Pointing Errors of the Green Bank Telescope Using an Optical Quadrant Detector

    Wind-induced pointing errors are a serious concern for large-aperture high-frequency radio telescopes. In this paper, we describe the implementation of an optical quadrant detector instrument that can detect and provide a correction signal for wind-induced pointing errors on the 100m diameter Green Bank Telescope (GBT). The instrument was calibrated using a combination of astronomical measurements and metrology. We find that the main wind-induced pointing errors on time scales of minutes are caused by the feedarm being blown along the direction of the wind vector. We also find that wind-induced structural excitation is virtually non-existent. We have implemented offline software to apply pointing corrections to the data from imaging instruments such as the MUSTANG 3.3 mm bolometer array, which can recover ~70% of sensitivity lost due to wind-induced pointing errors. We have also performed preliminary tests that show great promise for correcting these pointing errors in real-time using the telescope's subreflector servo system in combination with the quadrant detector signal.Comment: 17 pages, 11 figures; accepted for publication in PAS

    Predictors of Cardiometabolic Disease Risk Factors in Professional Firefighters

    The leading cause of mortality (~50% of deaths) among on-duty firefighters is sudden cardiac death. Firefighters with adequate cardiorespiratory fitness levels (among other muscular fitness metrics) and lower body fat percentage (BF%) are likely to better combat on-duty cardiac events and cardiovascular disease (CVD) risk. Individuals with higher VO2max values may be at lower risk noted by lower oxidative stress and inflammatory biomarkers, which may be more ideal predictors of CVD risk compared to traditional biomarkers (i.e., blood glucose and lipids) among the fire community; however, more research is needed to elucidate. PURPOSE: This study examined the relationship between fitness metrics, body fat percentage, and traditional and non-traditional biomarkers among professional firefighters. METHODS: Ninety-eight professional, male firefighters (age = 36.26 ± 9.08 yrs; height = 179.53 ± 7.06 cm; waist circumference = 37.99 ± 4.13 cm; BF% = 24.39 ± 5.21; years of experience = 11.81 ± 8.00 yrs) from a local fire department were studied VO2max was estimated from total treadmill exercise time using the Foster equation.. Fitness metrics including muscular strength, endurance, and power variables were also measured. Fasted blood samples were analyzed for concentrations of lipids, glucose, insulin resistance (HOMA-IR), advanced oxidation protein products (AOPP), and ultra-sensitive C-reactive protein (CRP). To assess the relationship between fitness metrics and AOPP, CRP, and HOMA-IR, ordinary least square regression analyses were used. RESULTS: VO2max values were inversely (p \u3c 0.05) related to AOPP, HOMA-IR, and CRP. Waist circumference was positively correlated to AOPP concentrations and HOMA-IR, while increased BF% was significantly related to increased CRP concentrations. CONCLUSION: High VO2max values were significantly related to lower AOPP, CRP, and HOMA-IR. Additionally, high waist circumference and BF% values were related to increases in oxidative stress, inflammation, and insulin resistance. Firefighters are encouraged to maintain high cardiorespiratory fitness (VO2max) and lower BF% to reduce CVD and, ultimately, on-duty sudden cardiac death risk

    Feasibility of Prehospital Emergency Anesthesia in the Cabin of an AW169 Helicopter Wearing Personal Protective Equipment During Coronavirus Disease 2019

    OBJECTIVE: Pre-hospital emergency anaesthesia in the form of rapid sequence intubation (RSI) is a critical intervention delivered by advanced pre-hospital critical care teams. Our previous simulation study determined the feasibility of in-aircraft RSI. We now examine whether this feasibility is preserved in a simulated setting, when clinicians wear personal protective equipment (PPE) for aerosol-generating procedures (AGP) for in-aircraft, on-the-ground RSI. METHODS: Air Ambulance Kent Surrey Sussex is a Helicopter Emergency Medical Service (HEMS) which utilises an AW169 cabin simulator. Wearing full AGP PPE (eye protection, FFP3 mask, gown, gloves), 10 doctor-paramedic teams performed RSI in a standard “can intubate, can ventilate” scenario and a “can't intubate, can't oxygenate” (CICO) scenario. Pre-specified timings were reported, and participant feedback was sought by questionnaire. RESULTS: RSI was most commonly performed by direct laryngoscopy and was successfully achieved in all scenarios. Time to completed endotracheal intubation (ETI) was fastest (287s) in the standard scenario and slower (370s, p=.01) in the CICO scenario. Time to ETI was not significantly delayed by wearing PPE in the standard (p=.19) or CICO variant (p=.97). Communication challenges, equipment complications and PPE difficulties were reported, but ways to mitigate these also reported. CONCLUSION: In-aircraft RSI (aircraft on-the-ground) whilst wearing PPE for AGPs had no significant impact on time to successful completion of ETI in a simulated setting. Patient safety is paramount in civilian HEMS, but the adoption of in-aircraft RSI could confer significant patient benefit in terms of pre-hospital time saving and further research is warranted

    Comparative analysis of affinity-based 5-hydroxymethylation enrichment techniques

    The epigenetic modification of 5-hydroxymethylcytosine (5hmC) is receiving great attention due to its potential role in DNA methylation reprogramming and as a cell state identifier. Given this interest, it is important to identify reliable and cost-effective methods for the enrichment of 5hmC marked DNA for downstream analysis. We tested three commonly used affinity-based enrichment techniques; (i) antibody, (ii) chemical capture and (iii) protein affinity enrichment and assessed their ability to accurately and reproducibly report 5hmC profiles in mouse tissues containing high (brain) and lower (liver) levels of 5hmC. The protein-affinity technique is a poor reporter of 5hmC profiles, delivering 5hmC patterns that are incompatible with other methods. Both antibody and chemical capture-based techniques generate highly similar genome-wide patterns for 5hmC, which are independently validated by standard quantitative PCR (qPCR) and glucosyl-sensitive restriction enzyme digestion (gRES-qPCR). Both antibody and chemical capture generated profiles reproducibly link to unique chromatin modification profiles associated with 5hmC. However, there appears to be a slight bias of the antibody to bind to regions of DNA rich in simple repeats. Ultimately, the increased specificity observed with chemical capture-based approaches makes this an attractive method for the analysis of locus-specific or genome-wide patterns of 5hm
