Das Niederdeutsche im Rahmen einer Sprachgeschichte des Deutschen
Das Nd. spielt in der institutionalisierten Sprachgeschichtsforschung des Deutschen seit über 150 Jahren als Gegenstand und als Varietät, die bis ins östliche Baltikum als Verkehrssprache galt, eine marginale Rolle (vgl. dazu Sanders 1982; Debus 1996; Wirrer 1998; Stellmacher 2000 und 2004). Die Auseinandersetzung mit den Varietäten des Nordens hat zum Ziel, das Bewusstsein dafür zu entwickeln, dass man mit der Verkehrssprache zusammen bisher die historische Entwicklung eines ganzen Sprach- und Kulturraums vernachlässigte (vgl. Michelsen 1993; Debus 1996 und Stellmacher 2004). Auf der 5. Jahrestagung der Gesellschaft für Germanistische Sprachgeschichte wurden die verstreuten Studien zu diesem Bereich gebündelt. Die Fachtagung mit dem Titel „Deutsch im Norden. Varietäten des norddeutschen Raumes im Spiegel der germanistischen Sprachgeschichtsschreibung“, die vom 25. bis 27. September 2014 an der Christian-Albrechts-Universität zu Kiel durchgeführt wurde, bot für diejenigen, die sich in diesem Forschungsgebiet betätigen, ein anregendes Diskussionsforum. Blickt man auf die Forschungslandschaft zum Nd., so kann man feststellen, dass sich nur vergleichsweise wenige Arbeiten mit der Sprachgeschichte des Nd. befassen (vgl. Sanders 1982; Stellmacher 2000). Eher ist davon zu sprechen, dass „Aspekte“ einer Sprachgeschichte vorgelegt worden sind (vgl. Eickmanns 2003; Gessinger 2003; Mattheier 2003; Peters 2003a und b; Rösler 2003; Scheuermann 2003). Noch immer ist die Frage von Stellmacher (1998) aktuell, ob, und wenn ja warum, „für die Darstellung der niederdeutschen Sprache eigene Prinzipien“ gelten
PSGL-1 engagement by E-selectin signals through Src kinase Fgr and ITAM adapters DAP12 and FcRγ to induce slow leukocyte rolling
E-selectin binding to P-selectin glycoprotein ligand-1 (PSGL-1) can activate the β2 integrin lymphocyte function-associated antigen-1 by signaling through spleen tyrosine kinase (Syk). This signaling is independent of Gαi-protein–coupled receptors, results in slow rolling, and promotes neutrophil recruitment to sites of inflammation. However, the signaling pathways linking E-selectin engagement of PSGL-1 to Syk activation are unknown. To test the role of Src family kinases and immunoreceptor tyrosine-based activating motif (ITAM)–containing adaptor proteins, we used different gene-deficient mice in flow chamber, intravital microscopy, and peritonitis studies. E-selectin–mediated phosphorylation of Syk and slow rolling was abolished in neutrophils from fgr−/− or hck−/− lyn−/− fgr−/− mice. Neutrophils from Tyrobp−/− Fcrg−/− mice lacking both DAP12 and FcRγ were incapable of sustaining slow neutrophil rolling on E-selectin and intercellular adhesion molecule-1 and were unable to phosphorylate Syk and p38 MAPK. This defect was confirmed in vivo by using mixed chimeric mice. Gαi-independent neutrophil recruitment into the inflamed peritoneal cavity was sharply suppressed in Tyrobp−/− Fcrg−/− mice. Our data demonstrate that an ITAM-dependent pathway involving the Src-family kinase Fgr and the ITAM-containing adaptor proteins DAP12 and FcRγ is involved in the initial signaling events downstream of PSGL-1 that are required to initiate neutrophil slow rolling
Exciton spin relaxation in single semiconductor quantum dots
We study the relaxation of the exciton spin (longitudinal relaxation time
) in single asymmetrical quantum dots due to an interplay of the
short--range exchange interaction and acoustic phonon deformation. The
calculated relaxation rates are found to depend strongly on the dot size,
magnetic field and temperature. For typical quantum dots and temperatures below
100 K, the zero--magnetic field relaxation times are long compared to the
exciton lifetime, yet they are strongly reduced in high magnetic fields. We
discuss explicitly quantum dots based on (In,Ga)As and (Cd,Zn)Se semiconductor

In-Datacenter Performance Analysis of a Tensor Processing Unit
Many architects believe that major improvements in cost-energy-performance
must now come from domain-specific hardware. This paper evaluates a custom
ASIC---called a Tensor Processing Unit (TPU)---deployed in datacenters since
2015 that accelerates the inference phase of neural networks (NN). The heart of
the TPU is a 65,536 8-bit MAC matrix multiply unit that offers a peak
throughput of 92 TeraOps/second (TOPS) and a large (28 MiB) software-managed
on-chip memory. The TPU's deterministic execution model is a better match to
the 99th-percentile response-time requirement of our NN applications than are
the time-varying optimizations of CPUs and GPUs (caches, out-of-order
execution, multithreading, multiprocessing, prefetching, ...) that help average
throughput more than guaranteed latency. The lack of such features helps
explain why, despite having myriad MACs and a big memory, the TPU is relatively
small and low power. We compare the TPU to a server-class Intel Haswell CPU and
an Nvidia K80 GPU, which are contemporaries deployed in the same datacenters.
Our workload, written in the high-level TensorFlow framework, uses production
NN applications (MLPs, CNNs, and LSTMs) that represent 95% of our datacenters'
NN inference demand. Despite low utilization for some applications, the TPU is
on average about 15X - 30X faster than its contemporary GPU or CPU, with
TOPS/Watt about 30X - 80X higher. Moreover, using the GPU's GDDR5 memory in the
TPU would triple achieved TOPS and raise TOPS/Watt to nearly 70X the GPU and

Highly Pathogenic H5N1 Influenza Viruses Carry Virulence Determinants beyond the Polybasic Hemagglutinin Cleavage Site
Highly pathogenic avian influenza viruses (HPAIV) originate from avirulent precursors but differ from all other influenza viruses by the presence of a polybasic cleavage site in their hemagglutinins (HA) of subtype H5 or H7. In this study, we investigated the ability of a low-pathogenic avian H5N1 strain to transform into an HPAIV. Using reverse genetics, we replaced the monobasic HA cleavage site of the low-pathogenic strain A/Teal/Germany/Wv632/2005 (H5N1) (TG05) by a polybasic motif from an HPAIV (TG05poly). To elucidate the virulence potential of all viral genes of HPAIV, we generated two reassortants carrying the HA from the HPAIV A/Swan/Germany/R65/06 (H5N1) (R65) plus the remaining genes from TG05 (TG05-HAR65) or in reversed composition the mutated TG05 HA plus the R65 genes (R65-HATG05poly). In vitro, TG05poly and both reassortants were able to replicate without the addition of trypsin, which is characteristic for HPAIV. Moreover, in contrast to avirulent TG05, the variants TG05poly, TG05-HAR65, and R65-HATG05poly are pathogenic in chicken to an increasing degree. Whereas the HA cleavage site mutant TG05poly led to temporary non-lethal disease in all animals, the reassortant TG05-HAR65 caused death in 3 of 10 animals. Furthermore, the reassortant R65-HATG05poly displayed the highest lethality as 8 of 10 chickens died, resembling “natural” HPAIV strains. Taken together, acquisition of a polybasic HA cleavage site is only one necessary step for evolution of low-pathogenic H5N1 strains into HPAIV. However, these low-pathogenic strains may already have cryptic virulence potential. Moreover, besides the polybasic cleavage site, the additional virulence determinants of H5N1 HPAIV are located within the HA itself and in other viral proteins
Mutations in tropomyosin 4 underlie a rare form of human macrothrombocytopenia.
Platelets are anuclear cells that are essential for blood clotting. They are produced by large polyploid precursor cells called megakaryocytes. Previous genome-wide association studies in nearly 70,000 individuals indicated that single nucleotide variants (SNVs) in the gene encoding the actin cytoskeletal regulator tropomyosin 4 (TPM4) exert an effect on the count and volume of platelets. Platelet number and volume are independent risk factors for heart attack and stroke. Here, we have identified 2 unrelated families in the BRIDGE Bleeding and Platelet Disorders (BPD) collection who carry a TPM4 variant that causes truncation of the TPM4 protein and segregates with macrothrombocytopenia, a disorder characterized by low platelet count. N-Ethyl-N-nitrosourea-induced (ENU-induced) missense mutations in Tpm4 or targeted inactivation of the Tpm4 locus led to gene dosage-dependent macrothrombocytopenia in mice. All other blood cell counts in Tpm4-deficient mice were normal. Insufficient TPM4 expression in human and mouse megakaryocytes resulted in a defect in the terminal stages of platelet production and had a mild effect on platelet function. Together, our findings demonstrate a nonredundant role for TPM4 in platelet biogenesis in humans and mice and reveal that truncating variants in TPM4 cause a previously undescribed dominant Mendelian platelet disorder.The research participants were enrolled in the Biomedical Research Centres/Units Inherited Diseases Genetic Evaluation (BRIDGE) Bleeding and Platelet Disorders (BPD) study (UK REC10/H0304/66).
