    Infection with host-range mutant adenovirus 5 suppresses innate immunity and induces systemic CD4+ T cell activation in rhesus macaques.

    Ad5 is a common cause of respiratory disease and an occasional cause of gastroenteritis and conjunctivitis, and seroconversion before adolescence is common in humans. To gain some insight into how Ad5 infection affects the immune system of rhesus macaques (RM) 18 RM were infected with a host-range mutant Ad5 (Ad5hr) by 3 mucosal inoculations. There was a delay of 2 to 6 weeks after the first inoculation before plasmacytoid dendritic cell (pDC) frequency and function increased in peripheral blood. Primary Ad5hr infection suppressed IFN-γ mRNA expression, but the second Ad5hr exposure induced a rapid increase in IFN-gamma mRNA in peripheral blood mononuclear cells (PBMC). Primary Ad5hr infection suppressed CCL20, TNF and IL-1 mRNA expression in PBMC, and subsequent virus exposures further dampened expression of these pro-inflammatory cytokines. Primary, but not secondary, Ad5hr inoculation increased the frequency of CXCR3+ CD4+ T cells in blood, while secondary, but not primary, Ad5hr infection transiently increased the frequencies of Ki67+, HLADR+ and CD95+/CCR5+ CD4+ T cells in blood. Ad5hr infection induced polyfunctional CD4 and CD8+ T cells specific for the Ad5 hexon protein in all of the animals. Thus, infection with Ad5hr induced a complex pattern of innate and adaptive immunity in RM that included transient systemic CD4+ T cell activation and suppressed innate immunity on re-exposure to the virus. The complex effects of adenovirus infection on the immune system may help to explain the unexpected results of testing Ad5 vector expressing HIV antigens in Ad5 seropositive people

    Comparison of need and supply of syringes for therapeutic injections in Pakistan

    Objectives: To determine the extent of shortfall in syringes by measuring the need from nationally validated sources and comparing it with position on the supply side. This was done in order to contribute to the ongoing national discussions that have focused on increasing syringe supply to curtail syringe reuse.Methods: Using \u27Injections received\u27 data from the Pakistan Demographic Health Survey 2006-7 (N: 5429) and the National Hepatitis Survey (N: 47,043), the study estimated the number of injections received in Pakistan. We matched these against the local syringe production figures from the syringe manufacturers\u27 association and import figures from the Federal Board of Revenue (FBR).Results: Approximately 731+/-867 million injections (5.1 12 injections per head) are received annually in Pakistan and around 861+/-961 million syringes are supplied to the open market. Overall, 52-77% Pakistanis receive at least one injection in any given year.Conclusions: Injection need in Pakistan is among the highest worldwide but is completely matched by available syringe supplies, suggesting lower reuse than previously considered. However, highly prevalent Hepatitis B and C viruses suggest that even this reuse is a major public health concern and measures directed at both providers and recipients of injections are needed to curtail high injection need and syringe reuse

    Scientific Mapping of Industry 4.0 Research:A Bibliometric Analysis

    The fourth industrial revolution is progressing very rapidly. This research aims to investigate the research patterns and trends of industry 4.0 research with a focus on manufacturing. This bibliometric analysis is performed on data of the past five years (2016 to 2020) retrieved from the Scopus database. This research is conducted on 1426 articles in which the top productive countries, authors, institutions, and most cited articles were investigated. Findings demonstrated that Italy, the United States, and China are the most active countries in terms of research publications. South China University of Technology (China) has been identified as the most productive institution.  Wan, J., Li, D., Rauch, E. were found to be the most productive authors. Industry 4.0 is primarily focused on the fields of engineering and computer science and sustainability is the most prolific journal. Co-occurrence analysis of keywords, co-authorship analysis of authors and countries were carried out along with bibliographic coupling of documents using VoS viewer which is the most common information visualisation software. This article summarises the growth of Industry 4.0 in the past five years and gives a short overview of the related works and applications of Industry 4.0.</p

    Hepatitis B and hepatitis C in Pakistan: prevalence and risk factors

    SummaryBackgroundPakistan carries one of the world’s highest burdens of chronic hepatitis and mortality due to liver failure and hepatocellular carcinomas. However, national level estimates of the prevalence of and risk factors for hepatitis B and hepatitis C are currently not available.MethodsWe reviewed the medical and public health literature over a 13-year period (January 1994–September 2007) to estimate the prevalence of active hepatitis B and chronic hepatitis C in Pakistan, analyzing data separately for the general and high-risk populations and for each of the four provinces. We included 84 publications with 139 studies (42 studies had two or more sub-studies).ResultsMethodological differences in studies made it inappropriate to conduct a formal meta-analysis to determine accurate national prevalence estimates, but we estimated the likely range of prevalence in different population sub-groups. A weighted average of hepatitis B antigen prevalence in pediatric populations was 2.4% (range 1.7–5.5%) and for hepatitis C antibody was 2.1% (range 0.4–5.4%). A weighted average of hepatitis B antigen prevalence among healthy adults (blood donors and non-donors) was 2.4% (range 1.4–11.0%) and for hepatitis C antibody was 3.0% (range 0.3–31.9%). Rates in the high-risk subgroups were far higher.ConclusionsData suggest a moderate to high prevalence of hepatitis B and hepatitis C in different areas of Pakistan. The published literature on the modes of transmission of hepatitis B and hepatitis C in Pakistan implicate contaminated needle use in medical care and drug abuse and unsafe blood and blood product transfusion as the major causal factors
