    Polyharmonic approximation on the sphere

    The purpose of this article is to provide new error estimates for a popular type of SBF approximation on the sphere: approximating by linear combinations of Green's functions of polyharmonic differential operators. We show that the LpL_p approximation order for this kind of approximation is σ\sigma for functions having LpL_p smoothness σ\sigma (for σ\sigma up to the order of the underlying differential operator, just as in univariate spline theory). This is an improvement over previous error estimates, which penalized the approximation order when measuring error in LpL_p, p>2 and held only in a restrictive setting when measuring error in LpL_p, p<2.Comment: 16 pages; revised version; to appear in Constr.     Differences in Efficacy Between Gamithromycin, Tilmicosin, and Tulathromycin as Metaphylactic Treatments in High Risk Calves for Bovine Respiratory Disease

    The cost of Bovine Respiratory Disease to the beef industry due to death, poorer conversions, and therapy is estimated to cost more than $3 billion per year. Identifying and mitigating Bovine Respiratory Disease in cattle can be difficult due to the increased susceptibility for Bovine Respiratory Disease in high risk cattle. One management option to minimize an outbreak of respiratory disease is the use of metaphylaxis, the mass treatment of a group of calves to reduce the incidence and adverse effects of respiratory disease on high risk animals. Criteria used to determine the necessity of metaphylactic treatment against Bovine Respiratory Disease in feedlots can be based on several factors depending on feedlot preference; however, the primary criteria often considered are: a known history of no previous vaccinations, overall appearance of cattle, source of cattle, Bovine Respiratory Disease in calves received from same source previously, long shipping distance, season of the year, and light arrival weight.     Flake Density, Roll Diameter, and Flake Moisture All Influence Starch Availability of Steam-Flaked Corn

    Steam-flaked corn is commonly fed in feedlot finishing diets because steam-flaking improves starch availability and nutrient utilization, thus improving the overall feeding value of corn. In most operations which utilize steam-flaked corn, grain is processed to a pre-determined flake density by setting the rolls to a specific separation distance and using tension to hold rolls together. Flaked grain is most often produced to a bulk density between 24 and 32 lb/bu, with a common recommendation of 27 lb/bu for corn; however, flake density among steam-flakers within a single mill and among feedlots can vary greatly. Flaking to a similar density using 2 flakers does not ensure similar starch availability. The degree of starch gelatinization or starch availability of steam-flaked corn can be estimated using analytical procedures such as enzymatic hydrolysis, gas production, and steam-flaked corn gelatinization methods. Routinely evaluating starch availability is used as a quality control method to standardize the steam-flaking process to ensure within-day and day-to-day manufacturing consistency. The concentration of readily available starch in steam-flaked corn is indicative of the rate of starch fermentation in the rumen. When starch is too readily available and is fermented at an excessively rapid rate, acid can accumulate in the rumen, reducing ruminal pH, and ultimately resulting in increased prevalence of digestive disturbances. Factors that contribute to variation between feedlot operations with respect to steamflaked corn quality include type and dimensions of flaking equipment, grain type, grain variety and moisture content, roll wear, and steam-flaking procedures. Sampling and handling procedures contribute to precision of results; therefore, sampling procedures need special attention, and consistency must be evaluated when attempting to determine starch availability of steam-flaked corn. The objective of this study was to evaluate starch availability of steam-flaked corn comparing roll dimensions and steam-flaked corn flake densities among flaking systems and feedyards and to provide information on the equipment utilized, steam-flaked corn flaking procedures, and to define current manufacturing practices of steam-flaking in commercial feedlot operations

    Experimentation on Analogue Models

    Summary Analogue models are actual physical setups used to model something else. They are especially useful when what we wish to investigate is difficult to observe or experiment upon due to size or distance in space or time: for example, if the thing we wish to investigate is too large, too far away, takes place on a time scale that is too long, does not yet exist or has ceased to exist. The range and variety of analogue models is too extensive to attempt a survey. In this article, I describe and discuss several different analogue model experiments, the results of those model experiments, and the basis for constructing them and interpreting their results. Examples of analogue models for surface waves in lakes, for earthquakes and volcanoes in geophysics, and for black holes in general relativity, are described, with a focus on examining the bases for claims that these analogues are appropriate analogues of what they are used to investigate. A table showing three different kinds of bases for reasoning using analogue models is provided. Finally, it is shown how the examples in this article counter three common misconceptions about the use of analogue models in physics

    Service recovery strategies have been identified as a critical factor in the success of. service organizations. This study develops a conceptual frame work to investigate how specific service recovery strategies influence the emotional, cognitive and negative behavioural responses of . consumers., as well as how emotion and cognition influence negative behavior. Understanding the impact of specific service recovery strategies will allow service providers' to more deliberately and intentionally engage in strategies that result in positive organizational outcomes. This study was conducted using a 2 x 2 between-subjects quasi-experimental design. The results suggest that service recovery has a significant impact on emotion, cognition and negative behavior. Similarly, satisfaction, negative emotion and positive emotion all influence negative behavior but distributive justice has no effect

    Anaphase-promoting complex/cyclosome protein Cdc27 is a target for curcumin-induced cell cycle arrest and apoptosis

    <p>Abstract</p> <p>Background</p> <p>Curcumin (diferuloylmethane), the yellow pigment in the Asian spice turmeric, is a hydrophobic polyphenol from the rhizome of <it>Curcuma longa</it>. Because of its chemopreventive and chemotherapeutic potential with no discernable side effects, it has become one of the major natural agents being developed for cancer therapy. Accumulating evidence suggests that curcumin induces cell death through activation of apoptotic pathways and inhibition of cell growth and proliferation. The mitotic checkpoint, or spindle assembly checkpoint (SAC), is the major cell cycle control mechanism to delay the onset of anaphase during mitosis. One of the key regulators of the SAC is the anaphase promoting complex/cyclosome (APC/C) which ubiquitinates cyclin B and securin and targets them for proteolysis. Because APC/C not only ensures cell cycle arrest upon spindle disruption but also promotes cell death in response to prolonged mitotic arrest, it has become an attractive drug target in cancer therapy.</p> <p>Methods</p> <p>Cell cycle profiles were determined in control and curcumin-treated medulloblastoma and various other cancer cell lines. Pull-down assays were used to confirm curcumin binding. APC/C activity was determined using an <it>in vitro </it>APC activity assay.</p> <p>Results</p> <p>We identified Cdc27/APC3, a component of the APC/C, as a novel molecular target of curcumin and showed that curcumin binds to and crosslinks Cdc27 to affect APC/C function. We further provide evidence that curcumin preferably induces apoptosis in cells expressing phosphorylated Cdc27 usually found in highly proliferating cells.</p> <p>Conclusions</p> <p>We report that curcumin directly targets the SAC to induce apoptosis preferably in cells with high levels of phosphorylated Cdc27. Our studies provide a possible molecular mechanism why curcumin induces apoptosis preferentially in cancer cells and suggest that phosphorylation of Cdc27 could be used as a biomarker to predict the therapeutic response of cancer cells to curcumin.</p

    Arsenic Trioxide Exerts Antimyeloma Effects by Inhibiting Activity in the Cytoplasmic Substrates of Histone Deacetylase 6

    Arsenic trioxide (As2O3) has shown remarkable efficacy for the treatment of multiple myeloma (MM). Histone deacetylases (HDAC) play an important role in the control of gene expression, and their dysregulation has been linked to myeloma. Especially, HDAC6, a unique cytoplasmic member of class II, which mainly functions as α-tubulin deacetylase and Hsp90 deacetylase, has become a target for drug development to treat cancer due to its major contribution in oncogenic cell transformation. However, the mechanisms of action for As2O3 have not yet been defined. In this study, we investigated the effect of As2O3 on proliferation and apoptosis in human myeloma cell line and primary myeloma cells, and then we studied that As2O3 exerts antimyeloma effects by inhibiting activity in the α-tubulin and Hsp90 through western blot analysis and immunoprecipitation. We found that As2O3 acts directly on MM cells at relatively low concentrations of 0.5∼2.5 µM, which effects survival and apoptosis of MM cells. However, As2O3 inhibited HDAC activity at the relatively high concentration and dose-dependent manner (great than 4 µM). Subsequently, we found that As2O3 treatment in a dose- and time-dependent fashion markedly increased the level of acetylated α-tubulin and acetylated Hsp90, and inhibited the chaperone association with IKKα activities and increased degradation of IKKα. Importantly, the loss of IKKα-associated Hsp90 occurred prior to any detectable loss in the levels of IKKα, indicating a novel pathway by which As2O3 down-regulates HDAC6 to destabilize IKKα protein via Hsp90 chaperone function. Furthermore, we observed the effect of As2O3 on TNF-α-induced NF-κB signaling pathway was to significantly reduced phosphorylation of Ser-536 on NF-κB p65. Therefore, our studies provide an important insight into the molecular mechanism of anti-myeloma activity of As2O3 in HDAC6-Hsp90-IKKα-NFκB signaling axis and the rationale for As2O3 can be extended readily using all the HDAC associated diseases
