    Reconstruction of the Elementary Life Education Curriculum From the Viewing Angle of AHP

    In recent years, more and more elementary school students die suddenly, and most of them are caused by the stampede or the jumping off unexpectedly. The Blue Book China Education Development Report (2014) announced by the Social Sciences Documentation Publishing House and Institute of 21st Century Education in May 2014, collecting 79 cases of suicide of primary and secondary school occurred in 2013. What’s more,there are many cases happened in 2014, such as the “September 26” murder case of Guangxi province, causing 3 deaths and one injury; The “September 26” stampede case of Yunnan, causing 6deaths and 26 injury; The “April 11” Elementary jump off event of Purple Tea House and “June 23” face-hitting to die event of Guangdong; The jumping event in Chongqing of a primary girl in May; And the “May 23” Nai xi Village pupils’ mass brawl event of Beijing. The above cases are all occurred in 2014. The frequent occurrence of the stampede and suicide, have beening causing the community’s attention. This paper attempts to analyze the elementary life education curriculum from the perspective of AHP, finding out and reconstructing a new elementary life education curriculum to reduce the mortality rate of primary school students

    Quantitative analysis of guided wave in dielectric logging through numerical simulation

    A good knowledge of the electromagnetic (EM) wave propagation behaviors in dielectric logging (DL) and borehole radar (BHR) surveying is critically important for the optimization of tool design and implementation, and interpretation of the acquired logging data, as well as understanding the influences of the dielectric permittivity and conductivity of the formation on the EM waves. This letter reported a novel method for the numerical simulation and analysis of the guided wave (GW) propagating along a metallic pipe in a typical DL configuration. A numerical simulation with the 3-D finite-difference time-domain (FDTD) method was applied to the broadband DL tool to obtain the wavefield and responses of the receiver. By monitoring the wave attenuation along the metallic drill collar, the intensity of the GW and loss factor can be determined. The coupling efficiency of the GW can be obtained when the total power emitted from the transmitting antenna is known. Simulation results revealed that the coupling efficiency of the GW changes with the water saturation of the formation and frequency. The simulation also suggest, by installing a slope structure adjacent to the transmitting antenna, the energy coupled into the GW could be reduced at different levels. Finally, the relationship between the received signals' amplitude and GW's coupling efficiency showed the quantified contribution of the GW to the received sign

    The Murine Reg3a Stimulated by Lactobacillus casei Promotes Intestinal Cell Proliferation and Inhibits the Multiplication of Porcine Diarrhea Causative Agent in vitro

    Lactobacillus casei (L. casei), a normal resident of the gastrointestinal tract of mammals, has been extensively studied over the past few decades for its probiotic properties in clinical and animal models. Some studies have shown that some bacterium of Lactobacillus stimulate the production of antimicrobial peptides in intestinal cells to clear enteric pathogens, however, which antimicrobial peptides are produced by L. casei stimulation and its function are still not completely understood. In this study, we investigated the changes of antimicrobial peptides’ expression after intragastric administration of L. casei to mice. The bioinformatics analysis revealed there were nine genes strongly associated with up-regulated DEGs. But, of these, only the antimicrobial peptide mReg3a gene was continuously up-regulated, which was also confirmed by qRT-PCR. We found out the mReg3a expressed in engineering E.coli promoted cell proliferation and wound healing proved by CCK-8 assay and wound healing assay. Moreover, the tight junction proteins ZO-1 and E-cadherin in mReg3a treatment group were significantly higher than that in the control group under the final concentration of 0.2 mg/ml both in Porcine intestinal epithelial cells (IPEC-J2) and Mouse intestinal epithelial cells (IEC-6) (p < 0.05). Surprisingly, the recombinant mReg3a not only inhibited Enterotoxigenic Escherichia coli (ETEC), but also reduced the copy number of the piglet diarrheal viruses, porcine epidemic diarrhea virus (PEDV), porcine transmissible gastroenteritis virus (TGEV), and porcine rotavirus (PoRV), indicating the antimicrobial peptides mReg3a may be feed additives to resist the potential of the intestinal bacterial and viral diarrhea disease

    Highly Anisotropic Glassy Polystyrenes Are Flexible

    We show that stretching polystyrene melts at a rate faster than the inverse Rouse time, followed by rapid quenching below the glass transition temperature, results in a material that is flexible and remains so for at least six months. Oriented micro/nanofibers are observed in the flexible samples after the mechanical tests. The fibers are probably related to the highly aligned molecules in melt stretching. At room temperature, a tensile strength over 300 MPa has been achieved for the flexible polystyrenes

    Orbital parameters for an ELM white dwarf with a white dwarf companion: LAMOST J033847.06+413424.2

    Double white dwarf systems are of great astrophysical importance in the field of gravitational wave and Type Ia supernova. While the binary fraction of CO core white dwarf is about a few percents, the extremely low mass white dwarfs are all thought to be within binary systems. In this work, we report the orbital solution of a double degenerate system: J033847.06+413424.24, an extremely low mass He core white dwarf orbiting a CO core white dwarf. With LAMOST and P200, time domain spectroscopic observations have been made and spectral atmosphere parameters are estimated to be Teff22500T_{\rm eff}\sim22500 K and log g5.6g\sim5.6 dex. Combining Gaia parallax, 3D extinction, and evolution tracks, we estimate a radius of 0.12\sim0.12 RR_{\odot} and a mass of 0.22\sim0.22 MM_{\odot}. With the 37 single exposure spectra, the radial velocities are measured and the orbital parameters are estimated to be P=0.1253132(1)P=0.1253132(1) days, K1=289±4K1=289\pm4 km/s and Vsys=41±3V_{sys}=-41\pm3 km/s. The radial velocity based system ephemeris is also provided. The light curves from several photometric surveys show no orbital modulation. The orbital solution suggests that the invisible companion has a minimum mass of about 0.60 MM_{\odot} and is 0.79\sim0.79 MM_{\odot} for an inclination of 60.060.0^{\circ}, indicating most probably a CO core white dwarf. The system is expected to merge in about 1 Gyr. With present period and distance (596\sim596 pc) it can not irradiate strong enough gravitational wave for LISA. More double degenerate systems are expected to be discovered and parameterized as the LAMOST survey goes on.Comment: 12 pages, 11 figure

    New tranylcypromine derivatives containing sulfonamide motif as potent LSD1 inhibitors to target acute myeloid leukemia: design, synthesis and biological evaluation

    Lysine-specific demethylase 1 (LSD1) is frequently elevated in acute myeloid leukemia (AML) and often leads to tumorigenesis. In recent years, numerous LSD1 inhibitors based on tranylcypromine (TCP) scaffolding have reached clinical trials. Most TCP derivatives were modified at the amino site of cyclopropane motif. Herein, we for the first time introduced a sulfonamide group in TCP benzene ring of series a compounds and performed a systematical study on structure and activity relationships by varying sulfonamide groups. The introduction of sulfonamide significantly increased the targeting capacity of TCP against LSD1. Moreover, we discovered that the Boc attached LSD1 inhibitors (labelled as series b compounds) substantially improved their anti-proliferation capacity towards AML cells. The intracellular thermal shift and LC-MS/MS results implied that Boc enhanced the drug lipophilicity and might be removed under the cancerous acidic environment to release the real pharmacophore, evidenced by the fact that a structurally similar but acidic inert pivaloyl to replace Boc dramatically dropped the cellular anti-proliferation effect. Finally, a benzyl group installed at the amino site to appropriately increase lipophilicity led to trans-4-(2-(benzylamino)-cyclopropyl)-N,N-diethylbenzenesulfonamide a10 that showed better anti-proliferation activity in AML cells and enzymatic inhibition against LSD1. Taken together, our work offers a novel TCP-based structure and provides a prodrug strategy for the discovery of potent LSD1 inhibitors by having appropriate lipophilicity

    SERPINB3 (SCCA1) inhibits cathepsin L and lysoptosis, protecting cervical cancer cells from chemoradiation

    The endogenous lysosomal cysteine protease inhibitor SERPINB3 (squamous cell carcinoma antigen 1, SCCA1) is elevated in patients with cervical cancer and other malignancies. High serum SERPINB3 is prognostic for recurrence and death following chemoradiation therapy. Cervical cancer cells genetically lacking SERPINB3 are more sensitive to ionizing radiation (IR), suggesting this protease inhibitor plays a role in therapeutic response. Here we demonstrate that SERPINB3-deficient cells have enhanced sensitivity to IR-induced cell death. Knock out of SERPINB3 sensitizes cells to a greater extent than cisplatin, the current standard of care. IR in SERPINB3 deficient cervical carcinoma cells induces predominantly necrotic cell death, with biochemical and cellular features of lysoptosis. Rescue with wild-type SERPINB3 or a reactive site loop mutant indicates that protease inhibitory activity is required to protect cervical tumor cells from radiation-induced death. Transcriptomics analysis of primary cervix tumor samples and genetic knock out demonstrates a role for the lysosomal protease cathepsin L in radiation-induced cell death in SERPINB3 knock-out cells. These data support targeting of SERPINB3 and lysoptosis to treat radioresistant cervical cancers
