    Eversão da vesícula urinária no coelho

    No setor de criação de coelhos da DTAEP- F.M. USP foi verificada em uma coelha N.Z.W. adulta a eversão completa da vesícula urinária. Ao exame macroscópico, a superfície mucosa apresentou-se de coloração avermelhada e com formações papilomatosas de 3 a 7 cm irregularmente distribuídas na porção distal do órgão. Cortes histológicos da área papilomatosa demonstraram uma erosão da mucosa recoberta por crosta de fibrina e hemáceas em alguns pontos. Preparados histológicos em outras regiões apresentaram quadro semelhante, com áreas focais de infiltrado inflamatório mais intenso. Tais resultados permitiram concluir um quadro de cistite crônica inespecífica.A rare case of complete urinary bladder inversion was verified in a female adult new zeeland while rabbit. In this region, a mucosal erosion overlaid in some places by fibrin with erythrocytes scab was observed under a microscopical aspect. The same pattern as described above was observed in other regions of the bladder, where the focal areas had a more intensive inflammatory infiltrate. These results allowed to conclude a inespecific chronic cystitis aspect

    Synaptophysin and synaptoporin expression in the developing rat olfactory system

    The expressions of two closely related synaptic vesicle antigens synaptophysin and synaptoporin were examined in the olfactory system of the adult rat and during pre- and postnatal development. In the adult, immunocytochemistry showed that the continuously regenerating olfactory receptor neurons (primary neurons) produce both synaptophysin and synaptoporin which were localized in the cell bodies of the receptor neurons in the olfactory epithelium, their dendrites, axonal processes in the olfactory nerve and their terminals in the olfactory bulb glomeruli. Furthermore, ultrastructural analysis revealed synaptophysin- and synaptoporin-immunore activities associated with synaptic vesicles in most olfactory receptor axonal terminals impinging on dendrites of the mitral and tufted neurons (secondary neurons in the olfactory bulb circuitry) in the olfactory glomeruli. In like manner, tufted neurons, granule and periglomerular neurons (interneurons in the olfactory bulb circuitry) express both synaptophysin and synaptoporin. In contrast, mitral neurons expressed only the synaptophysin antigen which was likewise associated with mitral axonal terminals in their target the olfactory cortex. The patterns of synaptophysin and synaptoporin expressions in mitral neurons (synaptophysin only) and tufted neurons (synaptophysin and synaptoporin) were similar in prenatal, postnatal and adult rats as revealed by immunocytochemistry and in situ hybridization.     A case of toxoplasmosis in an adult male emperor tamarin (Saguinus imperator) is reported. The primate was found dead and no clinical sign was noticed before death. Pathological findings included moderate to severe interstitial pneumonia, multifocal necrotizing hepatitis and multifocal to coalescing necrotizing lymphadenitis. Immunohistochemistry assays (strepto-avidin-biotin-peroxidase) performed on paraffin embedded tissues (lung, lymph nodes, liver, spleen, heart, intestine and adipose tissue) were strongly positive for Toxoplasma gondii.É relatado um caso de toxoplasmose ocorrido em sagüi-de-bigode (Saguinus imperator). O animal foi encontrado morto sem que tenham sido observados sinais clínicos. Achados de necropsia e histopatológicos incluíram pneumonia intersticial moderada, hepatite necrótica multifocal e linfadenite necrosante multifocal a coalescente. A técnica imunoistoquímica (strepto-avidina-biotina peroxidase) realizada em pulmão, fígado, linfonodos, baço, coração, intestino e tecido adiposo foi fortemente positiva para Toxoplasma gondii

    Differential expression of the neural cell adhesion molecule NCAM 140 in human pituitary tumors

    We have analyzed the expression of the intracellular marker protein neuron specific enolase (NSE), synaptophysin (SYN) and of the cell surface marker NCAM (neural cell adhesion molecule) in both normal human hypophysis and in pituitary adenomas in order to explore their potential use as diagnostic tools. All adenomas (4 prolactinomas, 3 growth hormone (GH) producing adenomas and 4 inactive adenomas) showed SYN and NSE immunoreactivity on tissue sections and this was confirmed by immunoblots. NCAM 140 (an isoform of NCAM with molecular mass 140 kDa) was detected by immunoblotting in normal human adenohypophysis, in all GH adenomas, and in three out of four inactive adenomas, but not in prolactinomas. Using highly sensitive techniques, NCAM immunoreactivity was observed by electron microscopy in all adenomas. These data indicate that NCAM 140 is a constituent of the cell surface of endocrine cells in both normal human adenohypophysis and its tumors. Since prolactinomas express very low levels of NCAM 140 compared to other hypophyseal tumors its virtual absence could be used for differential diagnosis. A combined analysis of NCAM, SYN and NSE could be useful to characterize inactive adenomas which are not immunoreactive for pituitary hormones and which may contain no or only low levels of the alpha chain of the glycoprotein hormones

    An evaluation of immunoreactivity for c-erbB-2 protein as a marker of poor short-term prognosis in breast cancer.

    Eighty-five breast carcinomas from the same number of patients have been assessed immunohistochemically using the antiserum 21N for the presence of the c-erbB-2 protein. Twenty-two of the patients had evidence of advanced disease (tumour fixation or distant metastases) at presentation. Follow-up was for a median of 24 months. c-erbB-2 protein was detected in the majority of cells in 14 (16.5%) carcinomas, and to a lesser extent in a further six (7%) tumours. There was no relationship between staining and stage, node status or size but more poorly differentiated carcinomas had evidence of staining (36%) than well (17%) or moderately (14%) differentiated carcinomas (P = 0.02). There was a significant association between staining and mortality (P = 0.009) and recurrence (P = 0.0002). The relative risk of death for staining compared to no staining (after adjusting for node status, stage and grade) was 2.97 (95% confidence interval 1.29, 6.84) and the relative risk of recurrence for staining compared to no staining after similar adjustment was 3.85 (95% confidence interval 1.86-7.97). In this particular group of patients immunoreactivity for c-erbB-2 protein is an independent indicator of poor short-term prognosis

    Immunohistochemical investigation of caspase-3 in neuronal apoptosis after experimental closed head trauma

    The aim of this study was to investigate the caspase-3 activity in neuronal apoptosis after experimental closed head trauma model in rats. Twenty adult male rats were randomly divided into two groups: control and trauma groups. In trauma group, a cranial impact was delivered to the skull from a height of 7 cm at a point just in front of the coronal suture and over the right hemisphere. Rats were sacrificed at 12 hours after the onset of injury. Brain tissues were removed for histopathological investigation. In the trauma group, the neurons became extensively dark and degenerated into picnotic nuclei. The number of apoptotic neurons in frontal cortex tissue of trauma group was significantly more than control groups. In conclusion, the caspase 3 immunopositivity was increased in degenerating neurons of the frontal cortex tissue following trauma. The present results indicate that closed head trauma caused degenerative changes and increased caspase 3 immonupositivity in neurons. © 2011 OMU All rights reserved

    Developmental expression of GLUT3 glucose transporter in the rat brain

    AbstractThe ontogeny of the GLUT3 glucose transporter gene and protein expression was studied in rat brain. Northern blot analysis using total RNA from rat brains at different developmental stages revealed that the levels of GLUT3 mRNA were very low during the embryonic stage and increased towards the postnatal stage. Immunohistochemistry using a specific antibody showed that the expression of GLUT3 protein was barely detectable in the embryonic stage, but was clearly detected on the plasma membrane of neuronal cells from 10 days after birth to the adult. Expression of GLUT3 mRNA and protein in the cerebral neuronal cell cultures was also examined during the maturation of neurons. GLUT3 glucose transporter of primary neuronal cultured cerebral cortical neurons was only detected in mature neurons after they were cultured for 14 days. These results indicate that GLUT3 plays an important role in glucose homeostasis postnatally in neurons of the rat brain

    Elevated P53 expression correlates with a history of heavy smoking in squamous cell carcinoma of the head and neck.

    Expression of the tumour suppressor gene p53 was examined in squamous cell carcinoma of the head and neck using two p53 antibodies, PAb 421 and PAb 1801. Elevated p53 expression was found in 67% of the 73 patients investigated. P53 expression was not found to correlate with whether the patient had been previously treated or not, nor any of the clinico-pathological parameters. However a correlation was found between the patients smoking history and positive p53 staining. Six out of seven non-smokers did not express p53 whereas 29 of 37 heavy smokers were found to have elevated p53 expression (P less than 0.005). Also, of a group of ten patients who had given up smoking more than 5 years ago, nine had elevated expression. Epidemiological studies have shown a correlation between heavy smoking and head and neck cancer. The present study indicate a genetic link for this correlation
