    A call for high quality early childhood education in the early years of school in Western Australia

    This paper has been written in response to the growing concerns from educators of children in the early years of school (kindergarten to year 2) and university teacher educators who are experiencing first-hand a rapid increase in the formalisation of the early years of schooling in many classrooms in Western Australia (The West Australian, 2013). Concerns for the general and long term health and wellbeing of young children have surfaced in the early childhood profession as we witness increased pressure placed on improving school achievement in national literacy and numeracy tests (King & Janson, 2009; Wescombe-Down, 2013)

    A call for a WA Play Strategy in Early Childhood Education and Care

    In 2013, ECA WA members were invited to contribute to the development of a strategic plan that would set priorities, focus energy and resources, and work toward the core function of Early Childhood Australia: to advocate for the rights and well-being of children in early childhood education and care (ECEC). Members identified the diminishing opportunities for children (aged birth to eight years) to access quality play in these environments as a major concern necessitating it as a key priority in future planning. The vision that child-initiated and self-directed play be reinstated as a fundamental right in early education and care settings, including the early years of schooling, was similarly shared by members of other leading early childhood advocacy organisations in WA (OMEP, ECTA, EYES and the SSTUWA). Moreover, the demise of play in ECEC was considered by key stakeholder representatives (of Playgroup WA, CareWest, Family Day Care WA, Meerilinga, and Childcare Association of WA) at the ECA WA AGM (2014) during a Panel Discussion on Play. Shortly thereafter, ECA WA pledged its commitment to developing a play strategy initiative

    The Ubiquitin Ligase Adaptor NDFIP1 Selectively Enforces a CD8<sup>+</sup> T Cell Tolerance Checkpoint to High-Dose Antigen

    Escape from peripheral tolerance checkpoints that control cytotoxic CD8+ T cells is important for cancer immunotherapy and autoimmunity, but pathways enforcing these checkpoints are mostly uncharted. We reveal that the HECT-type ubiquitin ligase activator, NDFIP1, enforces a cell-intrinsic CD8+ T cell checkpoint that desensitizes TCR signaling during in vivo exposure to high antigen levels. Ndfip1-deficient OT-I CD8+ T cells responding to high exogenous tolerogenic antigen doses that normally induce anergy aberrantly expanded and differentiated into effector cells that could precipitate autoimmune diabetes in RIP-OVAhi mice. In contrast, NDFIP1 was dispensable for peripheral deletion to low-dose exogenous or pancreatic islet-derived antigen and had little impact upon effector responses to Listeria or acute LCMV infection.     The role of the exit in the initial screening of investment opportunities: The case of business angel syndicate gatekeepers

    The exit process has been largely ignored in business angel research.. The practitioner community identifies the difficulty in achieving exits as the most pressing problem for investors. This has been attributed to the failure of investors to adopt an exit-centric approach to investing. The validity of this claim is examined via a study of the investment approach of 21 ‘gatekeepers’ (managers) of angel groups in Scotland and Northern Ireland. Most gatekeepers say that they do consider the exit when they invest. However, this is contradicted by a verbal protocol analysis which indicates that the exit is not a significant consideration in their initial screening process. The small number of exits achieved by the groups is consistent with the general lack of an exit-centric approach to investing. Only three groups exhibit evidence of a strong exit-centric approach to investing. The lack of exits may have a negative impact on the level of future angel investment activity

    Paedophiles in the community: inter-agency conflict, news leaks and the local press

    This article explores the leaking of confidential information about secret Home Office plans to house convicted paedophiles within a local community (albeit inside a prison). It argues that a politics of paedophilia has emerged in which inter-agency consensus on the issue of ‘what to do’ with high-profile sex offenders has broken down. Accordingly, the article situates newspaper ‘outing’ of paedophiles in the community in relation to vigilante journalism and leaked information from official agencies. The article then presents research findings from a case study of news events set in train following a whistle-blowing reaction by Prison Officers’ Association officials to Home Office plans. Drawing from a corpus of 10 interviews with journalists and key protagonists in the story, the article discusses both the dynamics of whistle blowing about paedophiles and also what happens after the whistle has blown

    Intensification of hydrological drought in California by human water management

    We analyze the contribution of human water management to the intensification and mitigation of hydrological drought over California using the PCR-GLOBWB hydrological model for the period 1979-2014. We demonstrate that considering water management results in more accurate discharge representation. During the severe 2014 drought, water management alleviated the drought deficit by ∌50% in Southern California through reservoir operation during low flow periods. However, human water consumption (mostly irrigation) in the Central Valley increased drought duration and deficit by 50% and 50-100%, respectively. Return level analysis indicates that there is more than 50% chance that the probability of occurrence of an extreme 2014-magnitude drought event was at least doubled under the influence of human activities compared to natural variability. This impact is most significant over the San Joaquin Drainage basin with a 50% and 75% likelihood that the return period is more than 3.5 and 1.5 times larger, respectively, because of human activities

    A manifesto for researching entrepreneurial ecosystems

    Entrepreneurial ecosystems are the focus of government economic policies around the world for their potential to generate entrepreneur-led economic development. The paper identifies key research questions and challenges to building effective public policy: (i) the limitations of existing data sources, (ii) the need to balance findings from quantitative and qualitative studies, (iii) the danger that entrepreneurial ecosystems will be just a policy fad, (iv) the narrow focus of policy and research on high tech firms and scale-ups, and (v) the need to balance research approaches between simplified models and a complex systems approach. There is a need for a better understanding of the diversity of policy contexts (level of government, country context) and model of ecosystem governance. A more granulated understanding of ecosystem thinking is required, with greater consideration of the diversity of actors and the institutional context, with more attention given to the heterogeneous nature of places and complex interactions between actors and networks. Looking to the future, the potential of new data sources and methodologies is identified. Future research should give greater consideration to the institutional context to understand how policy can better support entrepreneurial activity and the extent to which specific policies can be replicated elsewhere

    Sodium and potassium excretion in an adult Caribbean population of African descent with a high burden of cardiovascular disease

    Abstract Background High sodium diets with inadequate potassium and high sodium-to-potassium ratios are a known determinant of hypertension and cardiovascular disease (CVD). The Caribbean island of Barbados has a high prevalence of hypertension and mortality from CVD. Our objectives were to estimate sodium and potassium excretion, to compare estimated levels with recommended intakes and to identify the main food sources of sodium in Barbadian adults. Methods A sub-sample (n = 364; 25–64 years) was randomly selected from the representative population-based Health of the Nation cross-sectional study (n = 1234), in 2012–13. A single 24-h urine sample was collected from each participant, following a strictly applied protocol designed to reject incomplete samples, for the measurement of sodium and potassium excretion (in mg), which were used as proxy estimates of dietary intake. In addition, sensitivity analyses based on estimated completeness of urine collection from urine creatinine values were undertaken. Multiple linear regression was used to examine differences in sodium and potassium excretion, and the sodium-to-potassium ratio, by age, sex and educational level. Two 24-h recalls were used to identify the main dietary sources of sodium. All analyses were weighted for the survey design. Results Mean sodium excretion was 2656 (2488–2824) mg/day, with 67% (62–73%) exceeding the World Health Organization (WHO) recommended limit of 2000 mg/d. Mean potassium excretion was 1469 (1395–1542) mg/d; < 0.5% met recommended minimum intake levels. Mean sodium-to-potassium ratio was 2.0 (1.9–2.1); not one participant had a ratio that met WHO recommendations. Higher potassium intake and lower sodium-to-potassium ratio were independently associated with age and tertiary education. Sensitivity analyses based on urine creatinine values did not notably alter these findings. Conclusions In this first nationally representative study with objective assessment of sodium and potassium excretion in a Caribbean population in over 20 years, levels of sodium intake were high, and potassium intake was low. Younger age and lower educational level were associated with the highest sodium-to-potassium ratios. These findings provide baseline values for planning future policy interventions for non-communicable disease prevention

    Rapid isolation of mycoviral double-stranded RNA from Botrytis cinerea and Saccharomyces cerevisiae

    <p>Abstract</p> <p>Background</p> <p>In most of the infected fungi, the mycoviruses are latent or cryptic, the infected fungus does not show disease symptoms, and it is phenotypically identical to a non-infected strain of the same species. Because of these properties, the initial stage in the search for fungi infected with mycoviruses is the detection of their viral genome, which in most of the described cases corresponds to double-stranded RNA (dsRNA). So to analyze a large number of fungal isolates it is necessary to have a simple and rapid method to detect dsRNA.</p> <p>Results</p> <p>A rapid method to isolate dsRNA from a virus-infected filamentous fungus, <it>Botrytis cinerea</it>, and from a killer strain of <it>Saccharomyces cerevisiae </it>using commercial minicolumns packed with CF11 cellulose was developed. In addition to being a rapid method, it allows to use small quantities of yeasts or mycelium as starting material, being obtained sufficient dsRNA quantity that can later be analyzed by agarose gel electrophoresis, treated with enzymes for its partial characterization, amplified by RT-PCR and cloned in appropriate vectors for further sequencing.</p> <p>Conclusions</p> <p>The method yields high quality dsRNA, free from DNA and ssRNA. The use of nucleases to degrade the DNA or the ssRNA is not required, and it can be used to isolate dsRNA from any type of fungi or any biological sample that contains dsRNA.</p
