    The cell cycle program of polypeptide labeling in syndhronous cultures of wild-type Chlamydomonas reinhardtii was analyzed by pulse-labeling cells with 35SO4 = or [3H]arginine at different cell cycle stages. Nearly 100 labeled membrane and soluble polypeptides were resolved and studied using one-dimensional sodium dodecyl sulfate (SDS)- polyacrylamide gel electrophoresis. The labeling experiments produced the following results. (a) Total 35SO4 = and [3H]arginine incorporation rates varied independently throughout the cell cycle. 35SO4 = incorporation was highest in the mid-light phase, while [3H]arginine incorporation peaked in the dark phase just before cell division. (b) The relative labeling rate for 20 of 100 polypeptides showed significant fluctuations (3-12 fold) during the cell cycle. The remaining polypeptides were labeled at a rate commensurate with total 35SO4 = or [3H]arginine incorporation. The polypeptides that showed significant fluctuations in relative labeling rates served as markers to identify cell cycle stages. (c) The effects of illumination conditions on the apparent cell cycle stage-specific labeling of polypeptides were tested. Shifting light-grown asynchronous cells to the dark had an immediate and pronounced effect on the pattern of polypeptide labeling, but shifting dark-phase syndhronous cells to the light had little effect. The apparent cell cycle variations in the labeling of ribulose 1,5-biphosphate (RUBP)-carboxylase were strongly influenced by illumination effects. (d) Pulse-chase experiments with light-grown asynchronous cells revealed little turnover or inter- conversion of labeled polypeptides within one cell generation, meaning that major polypeptides, whether labeled in a stage-specific manner or not, do not appear transiently in the cell cycle of actively dividing, light-grown cells. The cell cycle program of labeling was used to analyze effects of a temperature-sensitive cycle blocked (cb) mutant. A synchronous culture of ts10001 was shifted to restrictive temperature before its block point to prevent it from dividing. The mutant continued its cell cycle program of polypeptide labeling for over a cell generation, despite its inability to divide

    This paper describes a simple mathematical model for one-dimensional elastoplastic wave propagation in a metal in the regime where the applied stress greatly exceeds the yield stress. Attention is focussed on the increasing ductility that occurs in the over-driven limit when the plastic wave speed approaches the elastic wave speed. Our model predicts that a plastic compression wave is unable to travel faster than the elastic wave speed, and instead splits into a compressive elastoplastic shock followed by a plastic expansion wave

    A theoretical model for single molecule incoherent scanning tunneling spectroscopy

    Single molecule scanning tunneling spectroscopy (STS), with dephasing due to elastic and inelastic scattering, is of some current interest. Motivated by this, we report an extended Huckel theory (EHT) based mean-field Non-equilibrium Green's function (NEGF) transport model with electron-phonon scattering treated within the self-consistent Born approximation (SCBA). Furthermore, a procedure based on EHT basis set modification is described. We use this model to study the effect of the temperature dependent dephasing, due to low lying modes in far-infrared range for which hw<<kT, on the resonant conduction through highest occupied molecular orbital (HOMO) level of a phenyl dithiol molecule sandwiched between two fcc-Au(111) contacts. Furthermore, we propose to include dephasing in room temperature molecular resonant conduction calculations.Comment: 12 pages, 5 figure

    Space shuttle external tank performance improvements: The challenge

    The external tank (ET) has been actively involved in performance improvements since the inception of the space shuttle program, primarily by weight savings. Weight savings were realized on the first block of flight articles (standard weight tank). With a need for further performance improvements, the ET Program Office was requested to develop a program to reduce tank weight an additional 6000 lb and schedule delivery of the first lightweight ET (LWT) for June 1982. The weight savings program was accomplished by: (1) a unique approach to use of factors of safety; (2) design optimization; and (3) redesign of structures with large margins of safety which resulted in an actual weight savings of 7294 lb. Additional studies have identified further weight savings which are to be implemented at appropriate times in production flow. Examples are an improved thermal protection system for the LH2 tank aft dome and reduction of slosh baffles in the LO2 tank based on flight data. All performance improvements were compared and selected based on non-recurring and recurring cost and technical risk

    Shuttle active thermal control system development testing. Volume 2: Modular radiator system tests

    Tests were designed to investigate the validity of the "modular" approach to space radiator system design for space shuttle and future applications by gathering performance data on various systems comprised of different numbers of identical panels, subject to nominal and extreme heat loads and environments. Both one-sided and two-sided radiation was tested, and engineering data was gathered on simulated low a/e coatings and system response to changes in outlet temperature control point. The results of the testing showed system stability throughout nominal orbital transients, unrealistically skewed environments, freeze-thaw transients, and rapid changes in outlet temperature control point. Various alternative panel plumbing arrangements were tested with no significant changes in performance being observed. With the MRS panels arranged to represent the shuttle baseline system, a maximum heat rejection of 76,600 Btu/hr was obtained in segmented tests under the expected worst case design environments. Testing of an alternate smaller two-sided radiation configuration yielded a maximum heat rejection of 52,931 Btu/hr under the maximum design environments

    Thanks, but no thanks: women's avoidance of help-seeking in the context of a dependency-related stereotype

    The stereotype that women are dependent on men is a commonly verbalized, potentially damaging aspect of benevolent sexism. We investigated how women may use behavioral disconfirmation of the personal applicability of the stereotype to negotiate such sexism. In an experiment (N = 86), we manipulated female college students’ awareness that women may be stereotyped by men as dependent. We then placed participants in a situation where they needed help. Women made aware of the dependency stereotype (compared to controls who were not) were less willing to seek help. They also displayed a stronger negative correlation between help-seeking and post help-seeking affect - such that the more help they sought, the worse they felt. We discuss the relevance of these findings for research concerning women’s help-seeking and their management of sexist stereotyping in everyday interaction. We also consider the implications of our results for those working in domains such as healthcare, teaching and counseling, where interaction with individuals in need and requiring help is common

    A summary is given of a series of thermal vacuum tests designed to support the development of the orbiter active thermal control system (ATCS) and included testing of a wide heat load range modular radiator system (MRS) configured to the March 1973 orbiter baseline system, a candidate weight reducing radiator/water cooling system, and a smaller radiator system with a high performance radiator coating. The tests verified the performance of the baseline system and obtained detailed design information for application of a wide heat load range modular radiator system to the orbiter. The two candidate ATCS weight reducing designs have undergone extensive concept verification testing and their system operating characteristics have been determined in sufficient detail for application to the orbiter. Design information has been obtained for an integrated radiator/water cooling system that provides for vehicle heat rejection as well as water management of the excess fuel cell water. Processing techniques have been developed and verified for the application of a high performance thermal control coating to large radiator areas subjected to a temperature range of -280 F to +160 F

    Orbiter radiator panel solar focusing test

    A test was conducted to determine the solar reflections from the Orbiter radiator panels. A one-tenth scale model of the forward and mid-forward radiator panels in the deployed position was utilized in the test. Test data was obtained to define the reflected one-sun envelope for the embossed silver/Teflon radiator coating. The effects of the double contour on the forward radiator panels were included in the test. Solar concentrations of 2 suns were measured and the one-sun envelope was found to extend approximately 86 inches above the radiator panel. A limited amount of test data was also obtained for the radiator panels with the smooth silver/Teflon coating to support the planned EVA on the Orbiter STS-5 flight. Reflected solar flux concentrations as high as 8 suns were observed with the smooth coating and the one-sun envelope was determined to extend 195 inches above the panel. It is recommended that additional testing be conducted to define the reflected solar environment beyond the one-sun boundary

    Development of a contact heat exchanger for a constructable radiator system

    A development program for a contact heat exchanger to be used to transfer heat from a spacecraft coolant loop to a heat pipe radiator is described. The contact heat exchanger provides for a connectable/disconnectable joint which allows for on-orbit assembly of the radiator system and replacement or exchange of radiator panels for repair and maintenance. The contact heat exchanger does not require the transfer of fluid across the joint; the spacecraft coolant loop remains contained in an all welded system with no static or dynamic fluid seals. The contact interface is also "dry' with no conductive grease or interstitial material required
