    Improved Training and Semen Collection Outcomes Using the Closed Box Chair for Macaques

    Collaborative semen collection in monkeys is a valuable tool in research, animal collection management, and conservation efforts. To obtain samples, monkeys are often restrained in open restraint chairs (ORC) with the “pole and collar” technique. While commonly used, this restraint is not tolerated by all individuals; some become anxious or aggressive towards the poles and people. In an effort to refine this procedure and improve welfare of the monkeys, we examined the use of a “closed box chair” (CBC), a clear, plexiglass box in which the monkey is trained to sit for sperm collection. The CBC does not require pole and collar, and although legs are secured, the arms and neck are not restrained. The use of CBCs has increased in recent years; however, there are few studies demonstrating its effects on scientific outcomes. We used positive reinforcement techniques to train 34 adult male rhesus macaques (Macaca mulatta) to provide semen samples using either the ORC or the CBC. While all CBC monkeys (n = 14) were reliably trained for this procedure, only 75% of ORC (n = 20) males completed the training (p = 0.04). It took significantly less time to train animals in the CBC than the ORC (201.0 vs. 412.4 min; p <0.001). In a controlled subset, males restrained with ORC (n = 7) produced a significantly lower ejaculatory volume than those collected by CBC (n = 10) (297.6 µL vs. 522.1 µL respectively; p = 0.04) and had a lower concentration of sperm (186.0 × 106/mL vs. 367.5 × 106/mL respectively; p = 0.017), although there were no differences with respect to sperm motility (p = 0.15). Our data suggest the closed box chair technique reduces stress on the animals while enhancing semen quality, supporting the use of the CBC as an important refinement

    The perceived effects of high stakes testing and Alabama's Plan 2020 on instructional practices and curriculum: a social studies instrumental case study

    The purpose of this study was to investigate the potential impact of Alabama's high stakes testing policy and Alabama's Plan 2020, on the instructional practices and curriculum of 12th grade secondary social studies teachers. This phenomenological qualitative instrumental case study design of four participants utilized guided interviews, document analysis, and a focus group interview. This study was grounded in the theoretical framework of constructivism, using the methodological foundations of the power relations theory of Michael Foucault. From this process, the researcher was able to understand the impact of these educational policies on instructional practices and curriculum. The results of the study indicated that the participants shifted their instruction and intensity to focus on state mandated testing due to the local school administration and internal pressures experienced. The results also displayed a lack of understanding for the new educational policy, a newly found instructional autonomy, and a fear of the return of state testing.     The impact of heat exposures on biomarkers of AKI and plasma metabolome among agricultural and non-agricultural workers

    Background: Agricultural workers are consistently exposed to elevated heat exposures and vulnerable to acute kidney injury. The underlying pathophysiology and detailed molecular mechanisms of AKI among agricultural workers, and the disproportionate burden of HRI and heat stress exposure are not well understood, especially at the level of cellular metabolism. Objective: The aim of this study was to examine the impact of heat exposures on renal biomarkers and on the human metabolome via untargeted high-resolution metabolomics among agricultural and non-agricultural workers. Methods: Blood and urine samples were collected pre- and post-work shift from 63 agricultural workers and 27 non– agricultural workers. We evaluated pre- and post-work shift renal biomarkers and completed untargeted metabolomics using high-resolution mass spectrometry with liquid chromatography. Metabolome-wide association studies (MWAS) models identified the metabolic features differentially expressed between agricultural workers and non-agricultural workers. Results: Median values of pre-shift creatinine and osteopontin (p < 0.05) were higher for agricultural workers than non-agricultural workers. Metabolic pathway enrichment analyses revealed 27 diverse pathways differed between agricultural workers and non-agricultural workers (p < 0.05) including TCA cycle and urea cycle, carbohydrate metabolism, histidine metabolism and evidence for altered microbiome shikimate pathway. Conclusion: This is the first investigation on the metabolic pathways that are affected among agricultural workers who are exposed to heat compared to non-heat exposed workers.     Help-seeking decisions among college students: the impact of mental health service affordability

    Recent statistics indicate that approximately 40% of students enrolled on U.S. college and university campuses report experiencing at least 1 mental health problem in the previous 12 months. Despite the documented benefits of counseling and mental health services on academic performance and degree attainment, only about 10% of mentally and emotionally distressed students ever seek professional help. The purpose of this study, therefore, was to gain a better understanding of why, among college students experiencing similar types of mental and emotional distress, some seek help, whereas most do not. For this study, 2 samples of students were recruited from 1 large, research university campus. The first was a clinical sample and consisted of distressed students who were attending a first screening appointment at the university's counseling center. The second was a random sample of students from the general student population who demonstrated levels of distress similar to the first sample, but who had chosen not to seek professional help. Participants in both samples completed 4 study instruments used for collecting demographic data, as well as data pertaining to help-seeking attitudes, help-seeking behaviors, treatment barriers, types of distress, and levels of distress. Data from both groups were combined to examine what variables contribute to the prediction of who, among similarly distressed college students, chooses to seek professional help and who does not. Of particular interest was the role that treatment barriers related to the affordability, availability, accessibility, and acceptability of mental health services might play in distinguishing help-seekers from non-help-seekers. A binary logistic regression model revealed that treatment-related barriers associated with cost of services, not knowing what services are available, and stigma were found to be significant predictors of help-seeking behavior. Among person-related barriers, measures of depression, generalized anxiety, eating concerns, and substance use were found to be significant predictors of help-seeking behavior. Another person-related barrier, help-seeking attitude, was found to be a significant predictor, but showed a lower rate of accuracy in predicting help-seeking behavior than the other significant predictors. A discussion of these findings is presented, along with associated implications for college campus stakeholders and directions for further research. (Published By University of Alabama Libraries

    Enhanced inflammation in New Zealand white rabbits when MERS-CoV reinfection occurs in the absence of neutralizing antibody

    <div><p>The Middle East respiratory syndrome coronavirus (MERS-CoV) is a zoonotic betacoronavirus that was first detected in humans in 2012 as a cause of severe acute respiratory disease. As of July 28, 2017, there have been 2,040 confirmed cases with 712 reported deaths. While many infections have been fatal, there have also been a large number of mild or asymptomatic cases discovered through monitoring and contact tracing. New Zealand white rabbits are a possible model for asymptomatic infection with MERS-CoV. In order to discover more about non-lethal infections and to learn whether a single infection with MERS-CoV would protect against reinfection, we inoculated rabbits with MERS-CoV and monitored the antibody and inflammatory response. Following intranasal infection, rabbits developed a transient dose-dependent pulmonary infection with moderately high levels of viral RNA, viral antigen, and perivascular inflammation in multiple lung lobes that was not associated with clinical signs. The rabbits developed antibodies against viral proteins that lacked neutralizing activity and the animals were not protected from reinfection. In fact, reinfection resulted in enhanced pulmonary inflammation, without an associated increase in viral RNA titers. Interestingly, passive transfer of serum from previously infected rabbits to naïve rabbits was associated with enhanced inflammation upon infection. We further found this inflammation was accompanied by increased recruitment of complement proteins compared to primary infection. However, reinfection elicited neutralizing antibodies that protected rabbits from subsequent viral challenge. Our data from the rabbit model suggests that people exposed to MERS-CoV who fail to develop a neutralizing antibody response, or persons whose neutralizing antibody titers have waned, may be at risk for severe lung disease on re-exposure to MERS-CoV.</p></div
