    The effect of type II toxin-antitoxin systems on methicillinresistant Staphylococcus aureus persister cell formation and antibiotic tolerance

    Persister cells are defi ned as a subpopulation of bacteria in a dormant state with the ability to reduce bacterial metabolism and they are involved in antibiotic tolerance. Toxin-antitoxin (TA) systems have been previously suggested as important players in persistence. Therefore, this study aimed to study the involvement of TA systems in persister cell formation in methicillin-resistant Staphylococcus aureus following antibiotic exposure. Using TADB and RASTA database, two type II TA systems including MazF/MazE and RelE/RelB were predicted in S. aureus. The presence of these TA genes was determined in 5 methicillin-resistant S. aureus isolates and the standard strain S. aureus subsp. aureus N315 using PCR method. To induce persistence, isolates were exposed to lethal doses of ciprofl oxacin and the expression of the studied TA system genes was measured after 5 h using Real-Time PCR. According to our results, all the studied isolates harbored the TA system genes. S. aureus was highly capable of persister cell formation following exposure to sub-MIC of ciprofl oxacin and RT-qPCR showed a signifi cant increase in the expression of the MazEF and RelBE loci, indicating their potential role in antibiotic tolerance. Considering the importance of antibiotic tolerance, further studies on persister cell formation and TA systems involved in this phenomenon are required to effi ciently target these systems

    A comparative study of the effect of massage with sunflower oil or sesame oil on infants' crying and sleep times: A randomized clinical trial

    Background & Aim: Infants are very sensitive and special attention is needed to take care of their physical and mental health. Some studies showed that massage by parents can improve infants' sleep quality. The aim of this study was to compare the effect of massage with sunflower oil or sesame oil on infants' crying and sleep times. Methods & Materials: This triple-blinded clinical trial was conducted on 120 infants who were 10-15 days old, full-term, single, exclusively breastfed and with no history of hospitalization. The infants were assigned randomly into three groups of sunflower oil massage, sesame oil massage and massage with no oils. Mothers administered 15 minutes of massage to their infants twice per day (morning and afternoon) for 28 days. Times of crying and sleep were measured by a parents' information form at baseline, and at the end of the first, second, third, and fourth weeks of the study. Data were analyzed using the repeated measures ANOVA. Result: Within four weeks of following, decreasing pattern of infants crying time was not significant (P=0.18); however, the mean of crying time between the three groups was significant (P=0.007). The interaction effect of crying time and groups was not significant (P=0.18). Increasing pattern of infant sleep time was statistically significant in the fourth weeks of the following (P<0.001); however, the mean of sleep time between three groups was not significant (P<0.56). The interaction effect of sleeping time and groups was not significant (P<0.10). Conclusion: Since there were not statistical significant differences between the infants' crying and sleep times between the three groups, and despite the fact that increasing pattern of infants sleep time is a normal pattern, it is not possible to have a proper conclusion

Specialists in various sciences conduct various researches in order to eliminate or reduce the problems caused by this disease. Nutrition is one of the sciences that plays a very important supportive role in this regard. It is important for patients to pay attention to the potential of different diets in preventing or accelerating the healing process. The relationship between nutrition and microbiome regulation or the occurrence of food microbial poisoning is one of the factors that can directly or indirectly play a key role in the body's resilience to COVID-19. In this article, we introduce a link between nutrition, the microbiome, and the incidence of food microbial poisoning that may have great potential in preventing, treating COVID-19, or preventing deterioration in patients. In linking the components of this network, artificial intelligence (AI), machine learning (ML) and data mining (DM) can be important strategies and lead to the creation of a conceptual model called "Balance square", which we will introduce

    Large oncosomes (LO) are atypically large (1-10 mu m diameter) cancer-derived extracellular vesicles (EVs), originating from the shedding of membrane blebs and associated with advanced disease. We report that 25% of the proteins, identified by a quantitative proteomics analysis, are differentially represented in large and nano-sized EVs from prostate cancer cells. Proteins enriched in large EVs included enzymes involved in glucose, glutamine and amino acid metabolism, all metabolic processes relevant to cancer. Glutamine metabolism was altered in cancer cells exposed to large EVs, an effect that was not observed upon treatment with exosomes. Large EVs exhibited discrete buoyant densities in iodixanol (OptiPrep (TM)) gradients. Fluorescent microscopy of large EVs revealed an appearance consistent with LO morphology, indicating that these structures can be categorized as LO. Among the proteins enriched in LO, cytokeratin 18 (CK18) was one of the most abundant (within the top 5th percentile) and was used to develop an assay to detect LO in the circulation and tissues of mice and patients with prostate cancer. These observations indicate that LO represent a discrete EV type that may play a distinct role in tumor progression and that may be a source of cancer-specific markers.1182Ysciescopu
