Conditional gene ablation of Stat3 reveals differential signaling requirements for survival of motoneurons during development and after nerve injury in the adult
Members of the ciliary neurotrophic factor (CNTF)/leukemia inhibitory factor (LIF)/cardiotrophin gene family are potent survival factors for embryonic and lesioned motoneurons. These factors act via receptor complexes involving gp130 and LIFR-β and ligand binding leads to activation of various signaling pathways, including phosphorylation of Stat3. The role of Stat3 in neuronal survival was investigated in mice by Cre-mediated gene ablation in motoneurons. Cre is expressed under the neurofilament light chain (NF-L) promoter, starting around E12 when these neurons become dependent on neurotrophic support. Loss of motoneurons during the embryonic period of naturally occurring cell death is not enhanced in NF-L–Cre; Stat3flox/KO mice although motoneurons isolated from these mice need higher concentrations of CNTF for maximal survival in culture. In contrast, motoneuron survival is significantly reduced after facial nerve lesion in the adult. These neurons, however, can be rescued by the addition of neurotrophic factors, including CNTF. Stat3 is essential for upregulation of Reg-2 and Bcl-xl expression in lesioned motoneurons. Our data show that Stat3 activation plays an essential role for motoneuron survival after nerve lesion in postnatal life but not during embryonic development, indicating that signaling requirements for motoneuron survival change during maturation
Memory Reduction via Delayed Simulation
We address a central (and classical) issue in the theory of infinite games:
the reduction of the memory size that is needed to implement winning strategies
in regular infinite games (i.e., controllers that ensure correct behavior
against actions of the environment, when the specification is a regular
omega-language). We propose an approach which attacks this problem before the
construction of a strategy, by first reducing the game graph that is obtained
from the specification. For the cases of specifications represented by
"request-response"-requirements and general "fairness" conditions, we show that

IQB-Bildungstrend 2018
Im Skalenhandbuch werden die Erhebungsinstrumente dokumentiert, die im IQB-Bildungstrend 2018 in den Fächern Mathematik, Biologie, Chemie und Physik in der Sekundarstufe I eingesetzt wurden. Die Bildungstrend-Studien des IQB sind Teil der Gesamtstrategie der Kultusministerkonferenz zum Bildungsmonitoring. Sie werden regelmäßig durchgeführt, um zu überprüfen, inwieweit die Schülerinnen und Schüler in den einzelnen Ländern in der Bundesrepublik Deutschland die in den länderübergreifend verbindlichen Bildungsstandards formulierten Kompetenzziele erreicht haben. Mit dem IQB-Bildungstrend 2018 war es erstmalig möglich, für das Erreichen der Standards im Fach Mathematik und in den naturwissenschaftlichen Fächern in der 9. Jahrgangsstufe Trends zu berichten (Vergleich der Erhebungsjahre 2012 und 2018)
Removing Biofilms from Microstructured Titanium Ex Vivo: A Novel Approach Using Atmospheric Plasma Technology
The removal of biofilms from microstructured titanium used for dental implants is a still unresolved challenge. This experimental study investigated disinfection and removal of in situ formed biofilms from microstructured titanium using cold atmospheric plasma in combination with air/water spray. Titanium discs (roughness (Ra): 1.96 µm) were exposed to human oral cavities for 24 and 72 hours (n = 149 each) to produce biofilms. Biofilm thickness was determined using confocal laser scanning microscopy (n = 5 each). Plasma treatment of biofilms was carried out ex vivo using a microwave-driven pulsed plasma source working at temperatures from 39 to 43°C. Following plasma treatment, one group was air/water spray treated before re-treatment by second plasma pulses. Vital microorganisms on the titanium surfaces were identified by contact culture (Rodac agar plates). Biofilm presence and bacterial viability were quantified by fluorescence microscopy. Morphology of titanium surfaces and attached biofilms was visualized by scanning electron microscopy (SEM). Total protein amounts of biofilms were colorimetrically quantified. Untreated and air/water treated biofilms served as controls. Cold plasma treatment of native biofilms with a mean thickness of 19 µm (24 h) to 91 µm (72 h) covering the microstructure of the titanium surface caused inactivation of biofilm bacteria and significant reduction of protein amounts. Total removal of biofilms, however, required additional application of air/water spray, and a second series of plasma treatment. Importantly, the microstructure of the titanium discs was not altered by plasma treatment. The combination of atmospheric plasma and non-abrasive air/water spray is applicable for complete elimination of oral biofilms from microstructured titanium used for dental implants and may enable new routes for the therapy of periimplant disease
Guideline adherence and patient satisfaction in the treatment of inflammatory bowel disorders – an evaluation study
Background: Crohn's disease (CD) and ulcerative colitis (UC) are the most frequent inflammatory bowel disorders (IBD). IBD cause a significant burden to society due to extensive health care utilization from the first clinical symptoms until diagnosis and thereafter due to direct and indirect costs. Besides the socio-economic impact of CD and UC, gastrointestinal and extraintestinal symptoms affect quality of life, but there is remarkably little data about the quality of treatment as assessed by patient satisfaction, quality of life and adherence to guidelines. Thus the aim of this study was to identify variables that influence quality of treatment and quality of life as well as patient satisfaction. Methods: The Essener Zirkel Study was a cross sectional study of 86 IBD-patients with a confirmed diagnosis of CD or UC. They were recruited at primary, secondary and tertiary care settings. Quality of treatment, quality of life and patient satisfaction were evaluated. Consulting behaviour and number of examinations, duration of disease and variables regarding adherence to guidelines were evaluated, too. Results: 59 (69%) patients had CD and 27 had UC (31%). 19% spent more than four years until the suspected diagnosis of IBD was confirmed and visited more than five physicians. All patients showed a significantly reduced quality of life compared to the 1998 German normative population. In spite of being under medical treatment, nearly half of the patients suffered from strong quality of life restricting symptoms. Over all, 35% described their treatment as moderate or bad. Patients who consulted psychotherapists and non-medical practitioners suffered significantly less from depression. Conclusion: Besides structural deficiencies due to the health care policy, we revealed the adherence to guidelines to be a problem area. The impact of the COVID-19 pandemic on administrative eating disorder prevalence in the outpatient sector and on severity of anorexia nervosa.
The COVID-19 pandemic appears to have had a considerable impact on the mental health of children and adolescents, particularly regarding eating disorders. However, it remains unclear whether the pandemic affected only the frequency or also the severity of eating disorders. We examined potential pandemic-related changes in the administrative prevalence of eating disorders in the outpatient sector compared with other mental disorders using German statutory health insurance data for the age group 10 to 16 years. We also examined disorder severity of anorexia nervosa using data from the multicenter German Registry of Children and Adolescents with Anorexia Nervosa in the same age group. Our results showed a marked increase in the administrative prevalence of eating disorders (based on documented diagnoses) in the outpatient sector among girls but not among boys. A similar pattern was found for internalizing disorders, whereas the administrative prevalences of externalizing disorders decreased. Regarding the severity of anorexia nervosa among inpatients, we found no pandemic-related changes in body mass index standard deviation score at admission, body weight loss before admission, psychiatric comorbidities and psychopharmacological medication. Given the administrative prevalence increase in the outpatient sector, the lack of impact of the pandemic on the inpatient sector may also be partly due to a shift in healthcare utilization towards outpatient services during the pandemic. Thus, the higher number of children and adolescents requiring specialized and timely outpatient care may be a major concern under pandemic conditions
The New Look pMSSM with Neutralino and Gravitino LSPs
The pMSSM provides a broad perspective on SUSY phenomenology. In this paper
we generate two new, very large, sets of pMSSM models with sparticle masses
extending up to 4 TeV, where the lightest supersymmetric particle (LSP) is
either a neutralino or gravitino. The existence of a gravitino LSP necessitates
a detailed study of its cosmological effects and we find that Big Bang
Nucleosynthesis places strong constraints on this scenario. Both sets are
subjected to a global set of theoretical, observational and experimental
constraints resulting in a sample of \sim 225k viable models for each LSP type.
The characteristics of these two model sets are briefly compared. We confront
the neutralino LSP model set with searches for SUSY at the 7 TeV LHC using both
the missing (MET) and non-missing ET ATLAS analyses. In the MET case, we employ
Monte Carlo estimates of the ratios of the SM backgrounds at 7 and 8 TeV to
rescale the 7 TeV data-driven ATLAS backgrounds to 8 TeV. This allows us to
determine the pMSSM parameter space coverage for this collision energy. We find
that an integrated luminosity of \sim 5-20 fb^{-1} at 8 TeV would yield a
substantial increase in this coverage compared to that at 7 TeV and can probe
roughly half of the model set. If the pMSSM is not discovered during the 8 TeV
run, then our model set will be essentially void of gluinos and lightest first
and second generation squarks that are \lesssim 700-800 GeV, which is much less
than the analogous mSUGRA bound. Finally, we demonstrate that non-MET SUSY
searches continue to play an important role in exploring the pMSSM parameter
space. These two pMSSM model sets can be used as the basis for investigations

Turning to the negative: attention allocation to emotional faces in adolescents with dysregulation profile—an event-related potential study
Patients with irritability, temper outbursts, hyperactivity and mood swings often meet the dysregulation profile (DP) of the Child Behavior Checklist (CBCL) or the Strengths and Difficulties Questionnaire (SDQ), which have been investigated over the past few decades. While the DP has emerged as a transdiagnostic marker with a negative impact on therapeutic outcome and psychosocial functioning, little is known about its underlying mechanisms such as attention and emotion regulation processes. In this study, we tested whether adolescent psychiatric patients (n = 27) with the SDQ-DP show impaired emotional face processing for task-irrelevant stimuli compared to psychiatric patients without the SDQ-DP (n = 30) and non-clinical adolescents (n = 21). Facial processing was tested with event-related potential (ERP) measures known to be modulated by attention (i.e., P1, N1, N170, P2, and Nc) during a modified Attention Network Task, to which task-irrelevant emotional stimuli (sad, fearful, and neutral faces) were added prior to the actual trial. The results reveal group differences in the orienting and in the conflicting network. Patients with DP showed a less efficient orienting network and the clinical control group showed a less efficient conflicting network. Moreover, patients with the dysregulation profile had a shorter N1/N170 latency than did the two control groups, suggesting that dysregulation in adolescents is associated with a faster but less arousing encoding of (task-irrelevant) emotional information and less top-down control
