Regulating the Motor for GLUT4 Vesicle Traffic

    Get PDF
Insulin-triggered trafficking of GLUT4 glucose transporter-loaded vesicles and their fusion with the plasma membrane are mechanical processes involving multiprotein complexes that coordinate and facilitate vesicle movement. Now, Yip et al. (2008) link myosin-1c to insulin signaling by demonstrating direct CaMKII-driven phosphorylation of this critical motor protein

CORRELATION OF FINE STRUCTURE AND PHYSIOLOGY OF THE INNERVATION OF SMOOTH MUSCLE IN THE GUINEA PIG VAS DEFERENS

An electron microscope study of the innervation of smooth muscle of the guinea pig vas deferens was undertaken in order to find a structural basis for recent electrophysiological observations. The external longitudinal muscle coat was examined in transverse section. Large areas of the surfaces of adjacent muscle cells were 500 to 800 A apart. Closer contacts were rare. A special type of close contact suggested cytoplasmic transfer between neighbouring cells. Groups of non-myelinated axons from ganglia at the distal end of the hypogastric nerve ramified throughout the muscle. Some small axon bundles and single axons lay in narrow fissures within closely packed muscle masses. Many axons contained "synaptic vesicles." About 25 per cent of the muscle fibres in the plane of section were within 0.25 µ of a partly naked axon; of these 15 per cent were within 500 A of the axon, and about 1 per cent made close contact (200 A) with a naked axon. It is unlikely that every muscle fibre is in close contact with an axon, and it is not possible for every fibre to have many such contacts. Muscle fibres are probably activated by both diffusion of transmitter from naked portions of axons a fraction of a micron distant, and electrotonic spread of activity from neighbouring cells

HAT-P-7: A Retrograde or Polar Orbit, and a Third Body

We show that the exoplanet HAT-P-7b has an extremely tilted orbit, with a true angle of at least 86 degrees with respect to its parent star's equatorial plane, and a strong possibility of retrograde motion. We also report evidence for an additional planet or companion star. The evidence for the unparalleled orbit and the third body is based on precise observations of the star's apparent radial velocity. The anomalous radial velocity due to rotation (the Rossiter-McLaughlin effect) was found to be a blueshift during the first half of the transit and a redshift during the second half, an inversion of the usual pattern, implying that the angle between the sky-projected orbital and stellar angular momentum vectors is 182.5 +/- 9.4 degreees. The third body is implicated by excess radial-velocity variation of the host star over 2 yr. Some possible explanations for the tilted orbit are a close encounter with another planet, the Kozai effect, and resonant capture by an inward-migrating outer planet.Comment: ApJ Letters, in press [7 pages

Insulin signaling meets vesicle traffic of GLUT4 at a plasma-membrane-activated fusion step

    Get PDF
SummaryA hypothesis that accounts for most of the available literature on insulin-stimulated GLUT4 translocation is that insulin action controls the access of GLUT4 vesicles to a constitutively active plasma-membrane fusion process. However, using an in vitro fusion assay, we show here that fusion is not constitutively active. Instead, the rate of fusion activity is stimulated 8-fold by insulin. Both the magnitude and time course of stimulated in vitro fusion recapitulate the cellular insulin response. Fusion is cell cytoplasm and SNARE dependent but does not require cell cytoskeleton. Furthermore, insulin activation of the plasma-membrane fraction of the fusion reaction is the essential step in regulation. Akt from the cytoplasm fraction is required for fusion. However, the participation of Akt in the stimulation of in vitro fusion is dependent on its in vitro recruitment onto the insulin-activated plasma membrane
