    Light emitting diode antifuse has been integrated into a microfluidic device that is realized with extended standard CMOS technological steps. The device comprises of a microchannel sandwiched between a photodiode detector and a nanometer-scale diode antifuse light emitter. Within this contribution, the device fabrication process, working principle and properties will be discussed. Change in the interference fringe of the antifuse spectra has been measured due to the filling of the channel.     AbstractHuman β-hexosaminidase A (Hex A) (αβ) is composed of two subunits whose primary structures are ∼60% identical. Deficiency of either subunit results in severe neurological disease due to the storage of GM2 ganglioside; Tay–Sachs disease, α deficiency, and Sandhoff disease, β deficiency. Whereas both subunits contain active sites only the α-site can efficiently bind negatively charged 6-sulfated hexosamine substrates and GM2 ganglioside. We have recently identified the αArg424 as playing a critical role in the binding of 6-sulfate-containing substrates, and βAsp452 as actively inhibiting their binding. To determine if these same residues affect the binding of the sialic acid moiety of GM2 ganglioside, an αArg424Gln form of Hex A was expressed and its kinetics analyzed using the GM2 activator protein:[3H]-GM2 ganglioside complex as a substrate. The mutant showed a ∼3-fold increase in its Km for the complex. Next a form of Hex B (ββ) containing a double mutation, βAspLeu453AsnArg (duplicating the α-aligning sequences), was expressed. As compared to the wild type (WT), the mutant exhibited a >30-fold increase in its ability to hydrolyze a 6-sulfated substrate and was now able to hydrolyze GM2 ganglioside when the GM2 activator protein was replaced by sodium taurocholate. Thus, this α-site is critical for binding both types of negatively charge substrates

    Levels of chemical contaminants in nonoccupationally exposed U. S. residents

    Data are presented on the levels of all chemical contaminants resulting from environmental pollution which have been found in human tissues including blood, urine, breast milk, and tissue samples obtained at autopsy. Most data results from specific surveys to determine health hazards. The roles of trace elements and recognition of the need to determine baseline levels of chemicals introduced into the environment are factors which have motivated surveys by individual investigators. Thus, most data on chemicals in human tissues record levels of pesticides (e.g., DDT and metabolites), levels of trace metals such as lead, cadmium, and mercury, or levels of nutritionally essential elements such as zinc, copper, manganese, and fluoride. Data available on iron and calcium are not presented as their presence in the environment is generally not considered hazardous. Data on several uncommon chemicals, such as indium and ytterbium, are included basically as items of interest and to further document their presence in healthy individuals. Baseline data were presented where available to provide perspective as to chemical levels which might be expected under conditions where exposure could be considered normal or not directly related to a pollutant source. Nearly 600 cited surveys or investigations, most of which were reported within the past decade, are listed. Ninety-four different chemical contaminants, primarily trace metals and organochlorine pesticides, are reported. It is estimated that over 75% of the data published during the past 30 years on chemical contaminants derived from environmental pollution and found in human tissue in the United States are represented in this report
