Antioxidantien als Strategie zur Optimierung der Kryokonservierung von Stammzellen für das Tissue Engineering
Kontrolliertes Einfrieren in den Temperaturbereich zwischen – 80 °C und – 196 °C erlaubt es Zellen, bei gleichzeitigem Erhalt ihrer Vitalität nach dem Auftauprozess, langfristig zu lagern. Durch die Verwendung von Antioxidantien sollte in der vorliegenden Arbeit eine Steigerung der Rekultivierbarkeit von mesenchymalen Stammzellen nach der Kryokonservierung erzielt werden. Der Zusatz von 100 μM α–Tocopherol steigerte die Rekultivierungseffzienz der Stammzellen um 117,5 % gegenüber der Kontrolle ohne Antioxidans. Darüber hinaus konnte gezeigt werden, dass eine Reduzierung der DMSO-Konzentration möglich ist, da Versuche mit 200 μM α–Tocopherol und 2,5 % bzw. 5 % DMSO vergleichbare Resultate von ca. 164 % bzw. 170 % Rekultivierbarkeit erzielten.EXC/62/
Encapsulating Non-Human Primate Multipotent Stromal Cells in Alginate via High Voltage for Cell-Based Therapies and Cryopreservation
Alginate cell-based therapy requires further development focused on clinical application. To assess engraftment, risk of mutations and therapeutic benefit studies should be performed in an appropriate non-human primate model, such as the common marmoset (Callithrix jacchus). In this work we encapsulated amnion derived multipotent stromal cells (MSCs) from Callithrix jacchus in defined size alginate beads using a high voltage technique. Our results indicate that i) alginate-cell mixing procedure and cell concentration do not affect the diameter of alginate beads, ii) encapsulation of high cell numbers (up to 10×106 cells/ml) can be performed in alginate beads utilizing high voltage and iii) high voltage (15–30 kV) does not alter the viability, proliferation and differentiation capacity of MSCs post-encapsulation compared with alginate encapsulated cells produced by the traditional air-flow method. The consistent results were obtained over the period of 7 days of encapsulated MSCs culture and after cryopreservation utilizing a slow cooling procedure (1 K/min). The results of this work show that high voltage encapsulation can further be maximized to develop cell-based therapies with alginate beads in a non-human primate model towards human application.DFG/EXC/62/

M+D: conceptual guidelines for compiling a materials library
This article proposes to present a study conducted by the Raw Materials research group, the results of which comprise the conceptual guidelines for compiling an M+D material library. The study includes the topic, materials and design taking the impact of the changes that came into being in the post industrial era on project methodologies and the search for information regarding materials.
Taking into account the importance and complexity that these relationships have taken on currently, we have studied the issue of materials based on Manzini (1983) and Ashby and Johnson (2002). Afterward different databases and materials libraries located in the Brazil, the United States, France and Italy geared toward design professionals and students were analyzed to understand what information and means of access to them were available. The project methodologies were approached based on Löbach (1991), Bürdeck (1994), Schulmann (1994), Baxter (1998), Dantas (1998 and 2005) and Papanek (1995 and 2000). This study sought to identify the key elements of the role of materials in the project process today, to serve as a parameter for the analysis of the models studied.
A comparative analysis of the models investigated enabled identification of positive and negative aspects to adapt to the needs previously mentioned and identify conceptual guidelines for compiling a collection of materials for use in design projects.

Impaired patient satisfaction and maladaption in ICD patients with and without shocks
In einer retrospektiven Querschnittstudie wurde bei 138 ICD-Patienten mittels standardisierter psychometrischer Testverfahren untersucht, welche Risikofaktoren zu verminderter Therapieakzeptanz und erhöhter psychischer Komorbidität führen. Ferner wurde überprüft, ob das Konzept der Posttraumatischen Belastungsstörung als Erklärungsmodell für Maladaptation geeignet ist. Trotz insgesamt hoher Therapiezufriedenheit und guter Adaptation an Krankheit und Therapie bei den meisten der untersuchten Patienten, wurde eine Untergruppe mit hoher Therapieablehnung und behandlungsbedürftiger affektiver Komorbidität identifiziert. Maladaptation ging insbesondere einher mit gehäuften Schockentladungen, aber auch mit dysfunktionalen Copingstrategien, wie Alexithymie u.a.In a retrospective cross-sectional study 138 ICD patients have been surveyed with standardised psychometric tests regarding risk factors for impaired patient satisfaction and increased psychiatric comorbidity. Furthermore, it has been examined whether or not the posttraumatic stress disorder paradigm can explain maladaption. In general the patients showed high satisfaction and good adaptation to disease and treatment. Nevertheless a subgroup of patients has been identified which had a highly negative attitude toward the device, as well as significant affective comorbidity. In particular, maladaption was associated not only with frequent shocks but also with dysfunctional coping strategies, such as alexithymia and others
An Evaluation of a Suicide Prevention E-Learning Program for Police Officers (COPS): Improvement in Knowledge and Competence
Background: Police officers are at high risk for mental and physical health problems and
suicidal ideation. The specific risk factors are numerous and concern stressful missions
and administrative aspects of the police profession. So far, the police get only little training
on specific missions as well as on coping with stress and suicidal ideation in the police
profession. In this study we test the efficacy of the online training COPS (Coping with
Suicide) for police officers.
Methods: A total of 142 police officers fromGermany and Switzerland participated in the
study; complete data (baseline and post) are available from 102 participants. The training
consisted of three modules covering the topics of delivering death notifications, dealing
with individuals with suicidal ideation and dealing with one’s own distress and suicidal
ideation in the police profession. The primary outcomes are perceived knowledge and
self-rated competence regarding the contents of the program, actual knowledge as well
as symptoms of depression and anxiety (PHQ-9), and attitudes toward suicide (ATTS).
The data are collected at baseline and after completing the training.
Results: We found a significant increase in knowledge as well as in perceived
competence after completing the training. Mental health and attitudes toward suicide did
not change significantly. Years on the job had no moderating effect on the effectiveness
of the training.
Discussion: The results suggest that a short e-learning program significantly improves
knowledge and self-rated competence in delivering death notifications, in suicide
prevention and stress management. It can be easily integrated into the daily routine
of police-officers, and participants could participate at their own pace and from any
location. One limitation of this study is the lack of a control-group. Further advantages
and limitations of this study are discussed
Cryopreservation of cells using defined serum-free cryoprotective agents
For regenerative purposes, there is a high demand for viable and active cells. A big issue is to have enough viable cells available at any given time. One solution is cryopreservation. In this context, DMSO is used as cryoprotective agent (CPA) along with fetal bovine serum for nutrient supply and stress shielding effects. To use these cells for human clinical studies, it is important to eliminate the serum to prevent foreign immune reactions and virus transmittance and DMSO for its toxic effect. In this study a serum free cryopreservation solution and protocol has been established. The combination of methylcellulose and poloxamer 188 provide the basis for the new CPA. Other additves are -tocopherol, ectoine, prolin and ascorbic acid. The CPAs were examined with 3T3-cells and multipotent stromal cells from the common marmoset monkey (Callithrix jacchus). The cells were preserved with various CPA concentrations, incubation times and different cooling rates. To enable a higher throughput of encouraging conditions a fluorescence microscopy analysis was used. The use of methylcellulose, poloxamer 188 and -tocopherol enables the reduction of DMSO [up to 2.5% (v/v)] and the elimination of serum without viability losses compared to control
