    Nové Město pod Smrkem in 1945-1960

    katedra: KHI; rozsah: 120 s. (205 086 znaků)This work discusses the development of Nové Město pod Smrkem in 1945-1960. It starts at the end of WWII, then follows the beginning and the consolidation of Communist power and terminates with 1960, when Nové Město became a part of district of Liberec. The submitted text aims to a postwar trasfer of German population mainly, also to an administration and a development of the city, working opporutnities, education and culture. An increased attention belongs to local manufactory of Textilana. This company was founded in the first half of the 19th Century and started off the biggest prosperity of Nové Město. The heyday lasted from the 2nd half of the 19th Century till the end of the 20th Century. The mining city turned to the textil one and this period of time is the main point of my work. The fate of the New-Town Hall is also analyzed in this work. It used to stand in the square until 1948. It burned out and totally came to an end. Other changes of the whole square were determined after it. The old pictures shows us the life in the town and also its status. There are samples of archival materials and interviews with contemporaries previous time.Tato práce pojednává o vývoji Nového Města pod Smrkem v letech 1945-1960. Zahrnuje tedy období od konce druhé světové války, pokračuje nástupem a upevněním komunistické moci a končí rokem 1960, kdy se Nové Město stalo součástí libereckého okresu. Předkládaný text se zaměřuje zejména na poválečný odsun německého obyvatelstva, správu a rozvoj města, pracovní příležitosti, vzdělání a kulturu. Zvýšená pozornost náleží místnímu závodu Textilana. Tento podnik vznikl přibližně v polovině 19. století a právě on odstartoval největší rozkvět Nového Města. Ten trval od druhé poloviny 19. století a téměř po celé století 20. Kdysi hornické město se tak stalo městem textilním a právě do téhle etapy spadá text této práce. Podrobněji rozebírán je taktéž osud původní novoměstské radnice, která stála do roku 1948 na náměstí. V tomto roce shořela a zcela zanikla. To rozhodlo o dalších proměnách celého náměstí. Dobové fotografie pak dokumentují stav a život v obci. Práci doplňují ukázky z archivních materiálů a rozhovory s pamětníky

    The use of phytohormones in production of fruit tree rootstocks in nursery without irrigation

    The most commonly used phytohormone to reduce the impact of drought is 24-epibrassinolide (EBR). In this study, EBR was applied over two years on young rootstocks of peach (B-VA-1) and myrobalan (Vanovice myrobalan) seedlings. Young rootstocks were treated by EBR at different concentrations, IAA and a mixture of EBR with IAA. The best results in seedling height of Vanovice myrobalan in both years had the treatment of 0.05% IAA, 549.69 mm (2016) and 553.77 mm (2017). On the other hand, on the crown root thickness was affected the most in 2016 by treatment of EBR 0.06 ppm (5.5 mm) and in 2017 by EBR 0.01 ppm (7.5mm). At B-VA-1 rootstock, the highest seedlings in 2016 were measured after EBR + IAA treatment (1573.18 mm) and in 2017 after IAA treatment (682.75 mm). The highest thickness of the crown root was recorded in 2016 after EBR and IAA (17.76 mm) and in 2017 after EBR 0.1 ppm (8.08 mm) treatments. At more than half of the treatments the control variant was evaluated as a variant with the least satisfying results. According to the results EBR and IAA are possibly increasing the quality of the rootstocks in conditions of a nursery without an irrigation.O

    The effect of freezing temperature on physiological traits in sunflower

    This study was conducted to identify the physiological mechanisms associated with the resistance and tolerance of young sunflower plants to freezing temperatures. The effect of overnight temperature –3°C on the maximal quantum efficiency of PSII (Fv/Fm), the relative electrolyte leakage (REL) and the osmotic potential (Ψπ) was determined in five genotypes of sunflower: C33, C98, C124 and C148 were chosen from the population of recombinant inbred lines (RILs) based on contrasted responses to low temperature, and a wild genotype 2603 that was chosen for its ability to maintain activities in cold conditions. The night temperature –3°C over the course of 10 h caused an immediate significant decrease of Fv/Fm in C33, C98, C124 and C148. In the case of genotype C98, the effect of this freezing temperature was manifested by a significant increase of REL. Significant changes of Ψπ, as a reaction to the effect of freezing temperatures, were not found in any of the monitored genotypes. The measurements of the physiological traits after 5 days of regeneration indicated the renewal of integrity of cellular structures and an increase of PSII reaction centre efficiency in all monitored genotypes. From the point of view of tolerance or sensitivity, the wild genotype 2603 showed itself as tolerant towards the tested freezing temperature, displaying insignificant differences with control plants in all monitored traits. Genotype C98 appears to be the most sensitive from the monitored set, with evident changes in two traits signalling frost damage

    After the change of political system in Czechoslovakia (1989) came also a lot of social, economical and cultural changes. Today, all the Czech cities stay in front of the biggest change of city planning philosophy in last two decades. Prague, the capital city of Czech Republic, decided for a big institutional transition in 2012. The municipality, in cooperation with Faculty of Architecture CTU in Prague, is preparing completely pioneering methodology for quality commissioning of land use plans and, in cooperation with the new Institute of Planning and Development, is preparing innovative system of city planning. There are new ordinances, laws, regulations, tourist trade strategies and many other documents. Prague, as one of the strongest regions in East-Central Europe, can be seen like a laboratory of current development of post-socialist city. The new methodology of Metropolitan Plan could be a key to success

    The Physiology and Proteomics of Drought Tolerance in Maize: Early Stomatal Closure as a Cause of Lower Tolerance to Short-Term Dehydration?

    Understanding the response of a crop to drought is the first step in the breeding of tolerant genotypes. In our study, two maize (Zea mays L.) genotypes with contrasting sensitivity to dehydration were subjected to moderate drought conditions. The subsequent analysis of their physiological parameters revealed a decreased stomatal conductance accompanied by a slighter decrease in the relative water content in the sensitive genotype. In contrast, the tolerant genotype maintained open stomata and active photosynthesis, even under dehydration conditions. Drought-induced changes in the leaf proteome were analyzed by two independent approaches, 2D gel electrophoresis and iTRAQ analysis, which provided compatible but only partially overlapping results. Drought caused the up-regulation of protective and stress-related proteins (mainly chaperones and dehydrins) in both genotypes. The differences in the levels of various detoxification proteins corresponded well with the observed changes in the activities of antioxidant enzymes. The number and levels of up-regulated protective proteins were generally lower in the sensitive genotype, implying a reduced level of proteosynthesis, which was also indicated by specific changes in the components of the translation machinery. Based on these results, we propose that the hypersensitive early stomatal closure in the sensitive genotype leads to the inhibition of photosynthesis and, subsequently, to a less efficient synthesis of the protective/detoxification proteins that are associated with drought tolerance

The work is closely focused on methods and methodology of marketing research ands justifies chosen research plan and methods determined to effective achievement of set goals. These methods are consequently used to analyse contemporary situation of enterprise and actual state of company customer satisfaction. The outcome of the work is above of others exposure of links among adjustment of marketing mix, actual customer satisfaction and company ability to be one step ahead of competitors. The goal of the work is to propose such measurements, which are going to lead to increase in customer satisfaction and improvements in company market position
