Indigo plant improves serum lipid profiles
We investigated the effects of Polygonum tinctorium Lour (PTL), a plant commonly known as indigo, on biological parameters in an animal model of high-fat diet-induced obesity. Wistar rats fed a high-fat diet and treated with PTL showed lower serum levels of triglycerides and total cholesterol levels and a higher serum levels of HDL cholesterol than those in Wistar rats fed a high-fat diet without PTL treatment. The weight of mesenteric fat in PTL-treated rats was decreased compared to that in control rats not treated with PTL. In addition, energy metabolic rate in the dark period, but not in the light period, in PTL-treated rats was higher than that in control rats. Although a significant difference was not observed, body weight in PTL-treated rats tended to be decreased compared to that in control rats. Breast Cancer with Cartilaginous and/or Osseous Metaplasia Diagnosed by Lymph Nodal Metastasis:A Case Report
Breast cancer with cartilaginous and/or osseous metaplasia is a type of metaplastic carcinomas and is a rare disease. We report the case of a 49 year-old female who underwent right mastectomy for a large breast tumor. Histological examinations revealed a mixed tumor with both stromal and epithelial elements;the stroma showed poor differentiated spindle-shape and multiform cells with a massive osseous matrix, and atypical epithelial cells, which mainly existed on the surface of the cysts, showed nucleic atypia. The tumor was diagnosed as a malignant phyllodes tumor with osteosarcomatous differentiation;it was not identified as a metaplastic carcinoma because of the lack of proof of a cancerous component. Two years after a mastectomy, swelling of the axillary lymph nodes was found and a biopsy was performed. Histological findings for the lymph node indicated a metastasis of the invasive ductal carcinoma. The primary tumor was re-examined and was considered to be the origin of the lymph nodal metastasis. Lymph nodal metastasis of cancer proved that the primary tumor had cancerous potential, and the pathological diagnosis was altered to a breast cancer with cartilaginous and/or osseous metaplasia.</p
Analysis of Communication between Inpatient Children and Their Mothers - I -
We observed the communication between inpatient children and their mothers mainly at the scenes of "play", "drug administration", and "meal" and classified their communication state into 6 types to evaluate the mother-child relationship. The 6 types were such as, good response type (Type I), repair type (Type II) , good response discontinuation type (Type III), response setback type (Type IV), rejective ending type (Type V), rejectin type (Type VI). 1. Sixty-two communication scenes were evaluated in 10 mother-child pairs. 2. Concerning child\u27s words and behavior, attachment for their mothers was observed at 82.5% of the "play" scenes. However, both attachment and rejection were observed at similar percentages of "drug administration" or "meal" scenes . 3. Concerning mother\u27s responses, good responses were most frequently observed at play scenes (88.0%). At "meal" scenes, good responses, good response discontinuation, and rejection were observed. In "drug administration" scenes, no rejection was observed. 4. The mother-child communication was classified into 6 types according to mother\u27s responses. 5. Type I and II indicating good responses were most frequently observed (77.4% of the scenes) . Type V and VI indicating rejective responses were observed in 9.7% of the scenes. 6. In "play" and "drug administration" scenes, good com- munication types were frequently observed. In "meal" scenes, various types were present. 7. The frequency of the communication types indicating rejection was significantly lower in relatively aged mothers group than in young mothers. It is clinically important to clarify the association between mother-child communication types and the situation in which the mother and child are placed based on the results of this study. The participation-observation method used in this study is an effective method of evaluating the relationship between inpatient children and their mothers for appro- priate nursing support
Split luciferase complementation assay to detect regulated protein-protein interactions in rice protoplasts in a large-scale format

Background: The rice interactome, in which a network of protein-protein interactions has been elucidated in rice, is a useful resource to identify functional modules of rice signal transduction pathways. Protein-protein interactions occur in cells in two ways, constitutive and regulative. While a yeast-based high-throughput method has been widely used to identify the constitutive interactions, a method to detect the regulated interactions is rarely developed for a large-scale analysis. Results: A split luciferase complementation assay was applied to detect the regulated interactions in rice. A transformation method of rice protoplasts in a 96-well plate was first established for a large-scale analysis. In addition, an antibody that specifically recognizes a carboxyl-terminal fragment of Renilla luciferase was newly developed. A pair of antibodies that recognize amino- and carboxyl- terminal fragments of Renilla luciferase, respectively, was then used to monitor quality and quantity of interacting recombinant-proteins accumulated in the cells. For a proof-of-concept, the method was applied to detect the gibberellin-dependent interaction between GIBBERELLIN INSENSITIVE DWARF1 and SLENDER RICE 1. Abdominal wall and thigh abscess resulting from the penetration of ascending colon cancer.
An unusual case is described in which an abdominal wall and thigh abscess was an initial symptom of ascending colon cancer. A 76-year-old woman was referred to our hospital for investigation of fever and abdominal and thigh swelling. Computed tomography revealed a right abdominal wall, retroperitoneal, psoas and thigh abscess formation suspected to be caused by colon perforation. Due to the patient's poor general condition, local drainage of the abscess was performed on the following day of hospitalization. Histological examination of necrotic tissues removed form the retroperitoneal cavity demonstrated adenocarcinoma of the colon. The patient subsequently underwent right hemicolectomy with lymph nodal dissection after 19 days of the drainage procedure and was transferred to another hospital on the 49th day following the second surgery.</p
Design and synthesis of caged ceramide : UV-responsive ceramide releasing system based on UV-induced amide bond cleavage followed by O–N acyl transfer
Sphingolipids, recognized as membrane constructs and as key signaling molecules, have been studied to examine intracellular function. Some caged sphingolipids that release parent sphingolipids after exposure to UV-irradiation have been previously developed, but caged ceramide has yet to be reported. In this study, we report the design and synthesis of a caged ceramide. Photo-irradiation experiment clarified that the caged ceramide can be successfully converted to the parent ceramide by UV-irradiation. Introduction of an alkyne-handle moiety for further modification of the caged ceramide is also reported
