Charakterisierung der Legionella pneumophila GDSL Hydrolasen PlaA und PlaD in Bezug auf EnzymaktivitÀt und Modulierung von Signalkaskaden des Wirts
The facultative intracellular bacterium Legionella pneumophila is the causative agent for Legionnairesâ disease, a potentially fatal pneumonia. The bacterium is ubiquitous in aqueous habitats and replicates inside protozoa which are its natural host. Infections of humans occur after inhalation of contaminated aerosols. Then, L. pneumophila replicates in a specialized phagosome termed Legionella containing vacuole inside alveolar macrophages. During infection, L. pneumophila secretes proteins, among others phospholipases, via its type II and type IVB secretion systems (T4BSS). L. pneumophila possesses 15 phospholipases A, 3 phospholipases C and 1 phospholipase D. The phospholipases A are further divided into the patatin-like proteins, the PlaB-like proteins and the GDSL hydrolases. The aim of this thesis was the characterization of the L. pneumophila GDSL hydrolases PlaA and PlaD with regard to enzymatic activity and the analysis of the interactions of PlaD with the host cell. Thus, L. pneumophila plaA- and plaD- mutants were analyzed in infection assays and purified recombinant proteins were used for investigation of lipolytic enzyme activity and determination of protein structure. Moreover, translocation and interaction assays were performed for the characterization of PlaD. PlaA exhibits strong lysophospholipase A activity and is processed by the L. pneumophila zinc metalloproteinase ProA within a disulfide loop in the C-terminal half of the protein increasing the lysophospholipase A activity of PlaA. Additionally, unprocessed PlaA shows glycerophospholipid:cholesterol acyltransferase activity which is diminished upon processing by ProA. Only minor activity was detected for PlaD. Instead, it was shown that PlaD is Dot/Icm-dependently injected into the host cell cytoplasm where it interacts with regulatory 14-3-3 proteins. Thus, PlaD was described as a novel T4BSS dependent effector protein of L. pneumophila. Moreover, it was found that PlaD is involved in the inhibition of host cell apoptosis upon L. pneumophila infection. The generated data lead to the hypothesis that the lysophospholipase A activity of PlaA might be directed towards the membrane of the Legionella containing vacuole and might thus be important for exit from the host cell. It is suspected that PlaD has to be activated by a so far unidentified activator to develop its full activity. Additionally, it is assumed that PlaD functions to modulate the host immune response during infection.Legionella pneumophila ist ein fakultativ intrazellulĂ€res Bakterium und Ursache der LegionĂ€rskrankheit, einer potentiell tödlichen Pneumonie. Menschen können durch das Inhalieren kontaminierter Aerosole infiziert werden. In dem Fall repliziert L. pneumophila in einem spezialisierten Phagosom namens âLegionella containing vacuoleâ in Alveolarmakrophagen. WĂ€hrend der Infektion sekretiert L. pneumophila Proteine, u.a. Phospholipasen, ĂŒber Typ II und Typ IVB Sekretionssystem (T4BSS). L. pneumophila besitzt 15 Phospholipasen A, 3 Phospholipasen C und 1 Phospholipase D. Die Phospholipasen A werden zusĂ€tzlich in die âPatatin-like proteinsâ, âdie PlaB-like proteinsâ und die âGDSL Hydrolasenâ eingeteilt. Ziel dieser Arbeit war die Charakterisierung der GDSL Hydrolasen PlaA und PlaD hinsichtlich enzymatischer AktivitĂ€t sowie die Untersuchung von PlaD hinsichtlich Interaktionen mit dem Wirt. Hierzu wurden Infektionsassays mit L. pneumophila plaA- und plaD- Mutanten durchgefĂŒhrt und aufgereinigtes rekombinantes Protein auf lipolytische AktivitĂ€t analysiert sowie zur Bestimmung der Proteinstruktur verwendet. AuĂerdem wurden fĂŒr PlaD Translokations- und Interaktionsstudien durchgefĂŒhrt. PlaA zeigt starke Lysophospholipase A (LPLA) AktivitĂ€t und wird innerhalb eines Disulfidloops in der C-terminalen HĂ€lfte des Proteins durch die L. pneumophila Zink Metalloproteinase ProA prozessiert, was die LPLA AktivitĂ€t steigert. Unprozessiertes PlaA zeigt zudem Glycerophospholipid:Cholesterol Acyltransferase AktivitĂ€t, welche durch die Prozessierung verschwindet. Im Gegensatz zeigt PlaD nur sehr geringe AktivitĂ€t detektiert. Stattdessen wurde gezeigt, dass PlaD Dot/Icm-abhĂ€ngig in die Wirtszelle injiziert wird und mit regulatorischen 14-3-3 Proteinen interagiert. PlaD konnte somit als neuer T4BSS abhĂ€ngiger Effektor von L. pneumophila beschrieben werden. DarĂŒber hinaus wurde gezeigt, dass PlaD die Apoptose der Wirtszelle inhibiert. Die Daten haben zu der Hypothese gefĂŒhrt, dass die Lysophospholipase A AktivitĂ€t von PlaA gegen die Membran der âLegionella containing vacuoleâ gerichtet und wichtig fĂŒr das Verlassen der Wirtszelle sein könnte. Es wird vermutet, dass ein bisher nicht identifizierter Aktivator notwendig ist, um die volle AktivitĂ€t von PlaD zu entwickeln. AuĂerdem wird angenommen, dass die Funktion von PlaD in der Modulation der Immunantwort des Wirts liegt
Zusammenhang zwischen Helicobacter pylori und angiographisch nachgewiesener koronarer Herzkrankheit : Erregernachweis mittels verschiedener Testmethoden
Trotz des Nachweises einer inflammatorischen Komponente bei der Entstehung der Atherosklerose herrscht nach wie vor Uneinigkeit ĂŒber die Erreger und Mechanismen welche diese Prozesse hervorrufen. Verschiedene Studien welche sich mit einer möglichen Beteiligung von H. pylori an diesen Prozessen kamen zu uneinheitlichen Ergebnissen. Ziel dieser Studie war es diesen Zusammenhang fĂŒr den GroĂraum Kiel zu untersuchen. Es wurden 186 Patienten (159 MĂ€nner, 27 Frauen) mit angiographisch nachgewiesener koronarer Herzkrankheit und 179 gesunden Kontrollpersonen (120 MĂ€nner, 59 Frauen) im Bezug auf signifikante Unterschiede in der SeroprĂ€valenz fĂŒr H. pylori-Antikörper untersucht. Gesondert untersucht wurde eine Gruppe von 66 Patienten, welche zu einer Kontrolluntersuchung nach vorangegangener Dilatations- oder Stentbehandlung gekommen waren, im Bezug auf signifikante Unterschiede in der SeroprĂ€valenz von H. pylori bei Patienten mit und ohne Restenose. Hierzu wurden ein Enzyme linked immuno sorbent assay (ELISA) sowie ein Immunoblot, jeweils zum Nachweis von IgG- und IgA-Antikörpern, verwendet. Die gesonderte Gruppe wurde auĂerdem mit zwei verschiedenen kommerziellen Immunoblot-Testen untersucht. Die statistische Auswertung der Ergebnisse erfolgte sowohl im Vergleich der gesamten Gruppen als auch in AbhĂ€ngigkeit des Alters (Personen †60 Jahre vs. Personen > 60 Jahre) und des Geschlechtes. Des Weiteren sollte die Nachweisbarkeit von H. pylori im Oropharynx, sowohl direkt als auch indirekt, an 137 Gurgelwasserproben aus der Patientengruppe untersucht werden. Der Direktnachweis erfolgte mittels Enzymimmoassay zum Nachweis von H. pylori-Antigenen, Polymerase-Chain-Reaction (PCR) und kulturellem Nachweis, der indirekte Nachweis mittels ELISA, Immunoblot und Ureasetest. Die verschiedenen Testmethoden wurden hinsichtlich der Ăbereinstimmung miteinander verglichen.
Im ELISA zeigten sich keine signifikanten Unterschiede in der SeroprĂ€valenz zwischen Patienten- und Kontrollgruppe. Im Immunoblot zeigte sich eine signifikant erhöhte IgG-SeroprĂ€valenz, welche ausschlieĂlich zwischen den mĂ€nnlichen Personen und denen †60 Jahren bestand. Diese Beobachtung deckt sich mit den Ergebnissen vorangegangener Studien, unterstĂŒtzt aber nicht die Theorie einer Beteilugung von H. pylori an atherosklerotischen Prozessen. Im Test der Patienten mit und ohne rezivierende Koronarstenosen zeigte sich im IgA-ELISA eine signifikant erhöhte SeroprĂ€valenz, welche sich allerdings im als BestĂ€tigungstest durchgefĂŒhrten Immunoblot nicht bestĂ€tigen lieĂ. Bei der Untersuchung des Gurgelwassers gelang der H. pylori-Nachweis mittels ELISA und Enzymimmunoassay. Der Nachweis mittels Enzymimmunoassay ist nach weiterer Evaluation grundsĂ€tzlich zur Diagnostik geeignet, der ELISA hingegen nur bedingt, da hier vor allem der IgA-Nachweis gelang. Kein Nachweis gelang mittels PCR, kulturellem Nachweis und Immunoblot, welche hiermit zum Nachweis auf Gurgelwasser weniger geeignet erscheinen. Im Vergleich der verschiedenen Testmethoden ergab sich zwischen ELISA und Immunoblot eine deutlich prozentuale Ăbereinstimmung, welches wahrscheinlich auf die höhere SpezifitĂ€t des Immunoblots zurĂŒckzufĂŒhren ist. Eine BestĂ€tigung eines positiven Ergebnisses im ELISA mittels Immunoblot erscheint zur Reduktion falsch positiver Ergebnisse sinnvoll. Im Vergleich von Serologie und den Ergebnissen der Untersuchungen des Gurgelwassers zeigten sich schwĂ€chere Ăbereinstimmungen, vor allem bezĂŒglich des IgG-Nachweises. Dies lĂ€sst auf eine prinzipielle Eignung zum Nachweis aus Gurgelwasser jedoch auf eine bedingte Eignung zur routinemĂ€Ăigen Testung, bei mĂ€Ăiger Nachweisbarkeit von mehr als einer Antikörperklasse, schlieĂen. Der Vergleich der verschiedenen Immunoblots zeigte insgesamt eine gute Ăbereinstimmung, jedoch mit deutlichen Unterschieden vor allem im Nachweis von VacA, was wahrscheinlich auf die Verwendung unterschiedlicher Antigene zurĂŒckzufĂŒhren ist
Fast Mapping of Terahertz Bursting Thresholds and Characteristics at Synchrotron Light Sources
Dedicated optics with extremely short electron bunches enable synchrotron
light sources to generate intense coherent THz radiation. The high degree of
spatial compression in this so-called low-alpha optics entails a complex
longitudinal dynamics of the electron bunches, which can be probed studying the
fluctuations in the emitted terahertz radiation caused by the micro-bunching
instability ("bursting"). This article presents a "quasi-instantaneous" method
for measuring the bursting characteristics by simultaneously collecting and
evaluating the information from all bunches in a multi-bunch fill, reducing the
measurement time from hours to seconds. This speed-up allows systematic studies
of the bursting characteristics for various accelerator settings within a
single fill of the machine, enabling a comprehensive comparison of the measured
bursting thresholds with theoretical predictions by the bunched-beam theory.

Simultaneous Detection of Longitudinal and Transverse Bunch Signals at a Storage Ring
To understand and control the dynamics in the longitudinal phase space,
time-resolved measurements of different bunch parameters are required. For a
reconstruction of this phase space, the detector systems have to be
synchronized. This reconstruction can be used e.g. for studies of the
micro-bunching instability. It occurs if the interaction of the bunch with its
own radiation leads to the formation of sub-structures on the longitudinal
bunch profile. These sub-structures can grow rapidly -- leading to a
sawtooth-like behaviour of the bunch. At KARA, we use a fast-gated intensified
camera for energy spread studies, Schottky diodes for coherent synchrotron
radiation studies as well as electro-optical spectral decoding for longitudinal
bunch profile measurements. For a synchronization, a hardware synchronization
scheme is used which compensates for eventual hardware delays. In this paper,
the different experimental setups and their synchronization are discussed and
first results of synchronous measurements are presented

A network analysis to identify mediators of germline-driven differences in breast cancer prognosis.
Identifying the underlying genetic drivers of the heritability of breast cancer prognosis remains elusive. We adapt a network-based approach to handle underpowered complex datasets to provide new insights into the potential function of germline variants in breast cancer prognosis. This network-based analysis studies ~7.3 million variants in 84,457 breast cancer patients in relation to breast cancer survival and confirms the results on 12,381 independent patients. Aggregating the prognostic effects of genetic variants across multiple genes, we identify four gene modules associated with survival in estrogen receptor (ER)-negative and one in ER-positive disease. The modules show biological enrichment for cancer-related processes such as G-alpha signaling, circadian clock, angiogenesis, and Rho-GTPases in apoptosis
From self-organization in relativistic electron bunches to coherent synchrotron light: observation using a photonic time-stretch digitizer
In recent and future synchrotron radiation facilities, relativistic electron
bunches with increasingly high charge density are needed for producing
brilliant light at various wavelengths, from X-rays to terahertz. In such
conditions, interaction of electrons bunches with their own emitted
electromagnetic fields leads to instabilities and spontaneous formation of
complex spatial structures. Understanding these instabilities is therefore key
in most electron accelerators. However, investigations suffer from the lack of
non-destructive recording tools for electron bunch shapes. In storage rings,
most studies thus focus on the resulting emitted radiation. Here, we present
measurements of the electric field in the immediate vicinity of the electron
bunch in a storage ring, over many turns. For recording the ultrafast electric
field, we designed a photonic time-stretch analog-to-digital converter with
terasamples/second acquisition rate. We could thus observe the predicted link
between spontaneous pattern formation and giant bursts of coherent synchrotron
radiation in a storage ring.Comment: 9 pages, 5 figure
