    A Novel Approach to Sequencing West Nile Virus Genome using IDT xGen and Illumina MiniSeq

    The West Nile virus (WNV) has been identified as a cause of mosquito-borne illness in the continental United States (CDC, 2023). It is a member of the Japanese encephalitis antigenic complex of the Flaviviridae family. WNV is predominantly maintained in an enzootic transmission cycle between Culex species mosquitoes and birds as vertebrate hosts. Humans and horses are considered WNV incidental hosts (a.k.a dead-end hosts) and cannot be transferred to another host. Within the last five years (2018-2022), 8,386 WNV human infection cases have been recorded. In addition, 90 WNV human infection cases have been reported in 17 different states this year. Nebraska has recorded the 4th highest total number of cases since WNV was introduced into the US. Monitoring the genetic variability of WNV will allow researchers to elucidate transmission patterns and ultimately incorporate WNV genomics into estimates of human risk. Understanding virus evolution through time requires an in-depth understanding of genomics. This research project aims to develop a more efficient and effective method for sequencing WNV genomes to better understand evolution. In addition, a novel approach was adopted to sequence WNV from mosquito pools collected in Nebraska.     Homelessness and Incarceration: An Examination of Barriers to Misdemeanor Resolution for the Homeless

    Abstract: The relationship between homelessness and crime has been investigated in few studies despite the implications of prior offenses and incarceration on job seekers. This study examined the relationship among homelessness, misdemeanors, incarceration, and misdemeanor resolution. Factors hindering misdemeanor resolution were explored by surveying participants at two Veterans Administration (VA) Stand Downs (n=186). The sample includes the primary service population of homeless veterans and other homeless persons. Respondents reported on factors perceived as limiting them from resolving misdemeanors, along with socio-demographic, homeless status, misdemeanor, and incarceration data. Factor Analysis yielded three factors as barriers that have implications for social work practice

    Determining Schistosoma haematobium Population Structures in Ethiopia

    Schistosomiasis, also known as bilharzia, is a Neglected Tropical Disease caused by parasitic helminths that affects over 240 million people around the world. Praziquantel has been used to treat individuals infected with schistosomiasis through Mass Drug Administration (MDA) programs but a recent reduction in its efficacy has been observed, creating concern that the parasite will evolve to become resistant to the chemotherapy drug. Monitoring changes in the population structure of Schistosoma haematobium using microsatellite markers can be a useful metric to determine praziquantel efficacy since variations in the repeat sequences of microsatellites indicate genetic diversity. Since little is known about the population structure of S. haematobium–despite urogenital schistosomiasis being a pressing issue in Ethiopia–18 microsatellite multiplex assays developed in a prior study were tested on stock DNA to validate them for use to study the effects of praziquantel on parasitic S. haematobium in Ethiopia. Through a combination of bioinformatic analysis, PCR, and Next Generation Sequencing on the MinION, 13 of the 18 microsatellite markers were validated, highlighting the importance of developing microsatellite multiplex assays not only based on length distribution, but also based on Next Generation Sequencing data.https://digitalcommons.unmc.edu/surp2023/1015/thumbnail.jp

    Workplace Traumatic Stress and Mental Health Sequelae among Public Safety Telecommunications Officers in Florida

    Background: Public safety telecommunications officers (PSTCOs), aka emergency “dispatchers,” are exposed to workplace traumatic stress and can experience situations characterized by uncertainty, communication difficulties, and a lack of resources. Traumatic stress experienced by emergency dispatchers has led to mental health symptoms. Purpose: This paper aims to describe the results of a study examining the patterns of workplace traumatic stress and the relationship between workplace traumatic stress and mental health concerns among a sample of PSTCOs. Methods: PSTCOs (n=54) participated in a cross-sectional, anonymous survey including screeners for depression, anxiety, post-traumatic stress disorder (PTSD), suicidal thoughts, and harmful alcohol use. Respondents also completed the Life Events Checklist (LEC) to screen for exposure to emergency calls identified as having the potential to induce traumatic stress. Results: 18.4% of respondents reported experiencing moderate to severe levels of depression, and 12% reported moderate to severe anxiety. Of the sample, 14% met the criteria for a provisional diagnosis of PTSD, 40% reported hazardous drinking levels, and 10% met the criteria of being at risk for suicide. Overall, 72.2% of PSTCOs experienced at least one form of job-related trauma and varied significantly by marital status. Poisson regression revealed statistically significant relationships between exposure to job-related traumatic events and screener scale scores. For example, those exposed to assault with a weapon had depression scores 2.29 points higher compared to those who were not exposed (p≤0.05). Discussion: Employing organizations of PSTCOs should strive to incorporate comprehensive wellness programs emphasizing education, prevention, early intervention, and recognition of traumatic stress among dispatchers. These programs should emphasize peer support and non-punitive policies to encourage help-seeking. In addition, given that findings in this study indicate exposure to traumatic emergency calls predicts PTSD symptomology, legislation could consider including PSTCOs in Florida Statute 118.1215

    Towards strange metallic holography

    We initiate a holographic model building approach to `strange metallic' phenomenology. Our model couples a neutral Lifshitz-invariant quantum critical theory, dual to a bulk gravitational background, to a finite density of gapped probe charge carriers, dually described by D-branes. In the physical regime of temperature much lower than the charge density and gap, we exhibit anomalous scalings of the temperature and frequency dependent conductivity. Choosing the dynamical critical exponent zz appropriately we can match the non-Fermi liquid scalings, such as linear resistivity, observed in strange metal regimes. As part of our investigation we outline three distinct string theory realizations of Lifshitz geometries: from F theory, from polarised branes, and from a gravitating charged Fermi gas. We also identify general features of renormalisation group flow in Lifshitz theories, such as the appearance of relevant charge-charge interactions when z≥2z \geq 2. We outline a program to extend this model building approach to other anomalous observables of interest such as the Hall conductivity.Comment: 71 pages, 8 figure

    Semi-Holographic Fermi Liquids

    We show that the universal physics of recent holographic non-Fermi liquid models is captured by a semi-holographic description, in which a dynamical boundary field is coupled to a strongly coupled conformal sector having a gravity dual. This allows various generalizations, such as a dynamical exponent and lattice and impurity effects. We examine possible relevant deformations, including multi-trace terms and spin-orbit effects.     An ethical framework for global vaccine allocation

    In this article, we propose the Fair Priority Model for COVID-19 vaccine distribution, and emphasize three fundamental values we believe should be considered when distributing a COVID-19 vaccine among countries: Benefiting people and limiting harm, prioritizing the disadvantaged, and equal moral concern for all individuals. The Priority Model addresses these values by focusing on mitigating three types of harms caused by COVID-19: death and permanent organ damage, indirect health consequences, such as health care system strain and stress, as well as economic destruction. It proposes proceeding in three phases: the first addresses premature death, the second long-term health issues and economic harms, and the third aims to contain viral transmission fully and restore pre-pandemic activity. To those who may deem an ethical framework irrelevant because of the belief that many countries will pursue "vaccine nationalism," we argue such a framework still has broad relevance. Reasonable national partiality would permit countries to focus on vaccine distribution within their borders up until the rate of transmission is below 1, at which point there would not be sufficient vaccine-preventable harm to justify retaining a vaccine. When a government reaches the limit of national partiality, it should release vaccines for other countries. We also argue against two other recent proposals. Distributing a vaccine proportional to a country's population mistakenly assumes that equality requires treating differently situated countries identically. Prioritizing countries according to the number of front-line health care workers, the proportion of the population over 65, and the number of people with comorbidities within each country may exacerbate disadvantage and end up giving the vaccine in large part to wealthy nations

    Circadian clock proteins regulate neuronal redox homeostasis and neurodegeneration

    Brain aging is associated with diminished circadian clock output and decreased expression of the core clock proteins, which regulate many aspects of cellular biochemistry and metabolism. The genes encoding clock proteins are expressed throughout the brain, though it is unknown whether these proteins modulate brain homeostasis. We observed that deletion of circadian clock transcriptional activators aryl hydrocarbon receptor nuclear translocator–like (Bmal1) alone, or circadian locomotor output cycles kaput (Clock) in combination with neuronal PAS domain protein 2 (Npas2), induced severe age-dependent astrogliosis in the cortex and hippocampus. Mice lacking the clock gene repressors period circadian clock 1 (Per1) and period circadian clock 2 (Per2) had no observed astrogliosis. Bmal1 deletion caused the degeneration of synaptic terminals and impaired cortical functional connectivity, as well as neuronal oxidative damage and impaired expression of several redox defense genes. Targeted deletion of Bmal1 in neurons and glia caused similar neuropathology, despite the retention of intact circadian behavioral and sleep-wake rhythms. Reduction of Bmal1 expression promoted neuronal death in primary cultures and in mice treated with a chemical inducer of oxidative injury and striatal neurodegeneration. Our findings indicate that BMAL1 in a complex with CLOCK or NPAS2 regulates cerebral redox homeostasis and connects impaired clock gene function to neurodegeneration
