    The causal influence of visual salience on gaze guidance during scene search and memorization

    Visual salience at fixation is often reported as significantly higher than chance. However, it is unclear if this gaze behaviour is caused by visual salience as comparisons have always been between objects or scenes. In the present study the salience of a target object was manipulated within scenes. Removing visual salience had a minimal effect on gaze behaviour but increasing saliency significantly increased the probability of early fixation during both search and memorisation. These results suggest that visual salience may play a causal role in fixation probability but its contribution independent from cognitive relevance is currently unknown

    Looking back at Waldo: Oculomotor inhibition of return does not prevent return fixations

    Inhibition of Return (IOR) is a difficulty in processing stimuli presented at recently attended locations. IOR is widely believed to facilitate foraging of a visual scene by decreasing the probability that gaze will return to previously fixated locations. However, there is a lack of clear evidence in support of the foraging facilitator hypothesis during scene search. The original R. M. Klein and W. J. MacInnes' (1999) Where's Waldo study reported a forward bias in the distribution of fixations that was taken as evidence for the foraging facilitator hypothesis. The present study was designed to replicate R. M. Klein and W. J. MacInnes' (1999) but include detailed analysis of fixation distributions in order to test the precise predictions of the foraging facilitator hypothesis. The results indicate that latencies of saccades returning to 1-back (and possibly 2-back) locations during visual search are elevated. However, there is no evidence that the probability of returning to these locations is significantly less than control locations. The nursing shortage diminishes quality of care and increases the costs of providing services. Resolution of the long-term care nursing workforce challenge will require improvement of the recruitment and retention of nurses. The leadership for the efforts to improve the long-term care workforce must come from the community itself. New and sustained leadership for action by long-term care leaders is the critical factor necessary to the development of the recommended initiatives and the achievement of the goals of the National Commission on Nursing Workforce for Long-Term Care

    The influence of clutter on real-world scene search: Evidence from search efficiency and eye movements

    We investigated the relationship between visual clutter and visual search in real-world scenes. Specifically, we investigated whether visual clutter, indexed by feature congestion, sub-band entropy, and edge density, correlates with search performance as assessed both by traditional behavioral measures (response time and error rate) and by eye movements. Our results demonstrate that clutter is related to search performance. These results hold for both traditional search measures and for eye movements. The results suggest that clutter may serve as an image-based proxy for search set size in real-world scenes

    Dissociating temporal inhibition of return and saccadic momentum across multiple eye-movement tasks

    Saccade latencies are longer prior to an eye movement to a recently fixated location than to control locations, a phenomenon known as oculomotor inhibition of return (O-IOR). There are theoretical reasons to expect that O-IOR would vary in magnitude across different eye movement tasks, but previous studies have produced contradictory evidence. However, this may have been because previous studies have not dissociated O-IOR and a related phenomenon, saccadic momentum, which is a bias to repeat saccade programs that also influences saccade latencies. The present study dissociated the influence of O-IOR and saccadic momentum across three complex visual tasks: scene search, scene memorization, and scene aesthetic preference. O-IOR was of similar magnitude across all three tasks, while saccadic momentum was weaker in scene search

    North to south: ecosystem features determine seagrass community response to sea otter foraging

    We compared sea otter recovery in California (CA) and British Columbia (BC) to determine how key ecosystem properties shape top-down effects in seagrass communities. Potential ecosystem drivers of sea otter foraging in CA and BC seagrass beds that we examined include the role of coastline complexity and environmental stress on sea otter effects. In BC, we found greater species richness across seagrass trophic assemblages. Furthermore, Cancer spp. crabs, an important link in the seagrass trophic cascade observed in CA, was less common. Additionally, the more recent reintroduction of sea otters, more complex coastline, and reduced environmental stress in BC seagrass habitats supported the hypothesis that sea otter foraging pressure is currently reduced in more northern latitudes. In order to manage the ecosystem features that lead to regional differences in top predator effects in seagrass communities, we review our findings, their spatial and temporal constraints, and present a social-ecological framework for future re- search

    Do the eyes really have it? Dynamic allocation of attention when viewing moving faces

    What controls gaze allocation during dynamic face perception? We monitored participants' eye movements while they watched videos featuring close-ups of pedestrians engaged in interviews. Contrary to previous findings using static displays, we observed no general preference to fixate eyes. Instead, gaze was dynamically directed to the eyes, nose, or mouth in response to the currently depicted event. Fixations to the eyes increased when a depicted face made eye contact with the camera, while fixations to the mouth increased when the face was speaking. When a face moved quickly, fixations concentrated on the nose, suggesting that it served as a spatial anchor. To better understand the influence of auditory speech during dynamic face perception, we presented participants with a second version of the same video, in which the audio speech track had been removed, leaving just the background music. Removing the speech signal modulated gaze allocation by decreasing fixations to faces generally and the mouth specifically. Since the task was to simply rate the likeability of the videos, the decrease of attention allocation to the mouth region implies a reduction of the functional benefits of mouth fixations given that speech comprehension was not required. Together, these results argue against a general prioritization of the eyes and support a more functional, information-seeking use of gaze allocation during dynamic face viewing

    L-5-methyltetrahydrofolate supplementation increases blood folate concentrations to a greater extent than folic acid supplementation in Malaysian women

    Background: Folic acid fortification of grains is mandated in many countries to prevent neural tube defects. Concerns regarding excessive intakes of folic acid have been raised. A synthetic analog of the circulating form of folate, l-5-methyltetrahydrofolate (l-5-MTHF), may be a potential alternative. Objective: The objective of this study was to determine the effects of folic acid or l-5-MTHF supplementation on blood folate concentrations, methyl nutrient metabolites, and DNA methylation in women living in Malaysia, where there is no mandatory fortification policy. Methods: In a 12-wk, randomized, placebo-controlled intervention trial, healthy Malaysian women (n = 142, aged 20–45 y) were randomly assigned to receive 1 of the following supplements daily: 1 mg (2.27 μmol) folic acid, 1.13 mg (2.27 μmol) l-5-MTHF, or a placebo. The primary outcomes were plasma and RBC folate and vitamin B-12 concentrations. Secondary outcomes included plasma total homocysteine, total cysteine, methionine, betaine, and choline concentrations and monocyte long interspersed nuclear element-1 (LINE-1) methylation. Results: The folic acid and l-5-MTHF groups had higher (P < 0.001) RBC folate (mean ± SD: 1498 ± 580 and 1951 ± 496 nmol/L, respectively) and plasma folate [median (25th, 75th percentiles): 40.1 nmol/L (24.9, 52.7 nmol/L) and 52.0 nmol/L (42.7, 73.1 nmol/L), respectively] concentrations compared with RBC folate (958 ± 345 nmol/L) and plasma folate [12.6 nmol/L (8.80, 17.0 nmol/L)] concentrations in the placebo group at 12 wk. The l-5-MTHF group had higher RBC folate (1951 ± 496 nmol/L; P = 0.003) and plasma folate [52.0 nmol/L (42.7, 73.1 nmol/L); P = 0.023] at 12 wk than did the folic acid group [RBC folate, 1498 ± 580 nmol/L; plasma folate, 40.1 nmol/L (24.9, 52.7 nmol/L)]. The folic acid and l-5-MTHF groups had 17% and 15%, respectively, lower (P < 0.001) plasma total homocysteine concentrations than did the placebo group at 12 wk; there were no differences between the folic acid and l-5-MTHF groups. No differences in plasma vitamin B-12, total cysteine, methionine, betaine, and choline and monocyte LINE-1 methylation were observed. Conclusion: These findings suggest differential effects of l-5-MTHF compared with folic acid supplementation on blood folate concentrations but no differences on plasma total homocysteine lowering in Malaysian women.     Smoking-Relevant Nicotine Concentration Attenuates the Unfolded Protein Response in Dopaminergic Neurons

    Retrospective epidemiological studies show an inverse correlation between susceptibility to Parkinson's disease and a person's history of tobacco use. Animal model studies suggest nicotine as a neuroprotective agent and nicotinic acetylcholine (ACh) receptors (nAChRs) as targets for neuroprotection, but the underlying neuroprotective mechanism(s) are unknown. We cultured mouse ventral midbrain neurons for 3 weeks. Ten to 20% of neurons were dopaminergic (DA), revealed by tyrosine hydroxylase (TH) immunoreactivity. We evoked mild endoplasmic reticulum (ER) stress with tunicamycin (Tu), producing modest increases in the level of nuclear ATF6, phosphorylated eukaryotic initiation factor 2α, nuclear XBP1, and the downstream proapoptotic effector nuclear C/EBP homologous protein. We incubated cultures for 2 weeks with 200 nm nicotine, the approximate steady-state concentration between cigarette smoking or vaping, or during nicotine patch use. Nicotine incubation suppressed Tu-induced ER stress and the unfolded protein response (UPR). Study of mice with fluorescent nAChR subunits showed that the cultured TH+ neurons displayed α4, α6, and β3 nAChR subunit expression and ACh-evoked currents. Gene expression profile in cultures from TH-eGFP mice showed that the TH+ neurons also express several other genes associated with DA release. Nicotine also upregulated ACh-induced currents in DA neurons by ∼2.5-fold. Thus, nicotine, at a concentration too low to activate an appreciable fraction of plasma membrane nAChRs, induces two sequelae of pharmacological chaperoning in the ER: UPR suppression and nAChR upregulation. Therefore, one mechanism of neuroprotection by nicotine is pharmacological chaperoning, leading to UPR suppression. Measuring this pathway may help in assessing neuroprotection

    Rapid, ultra low coverage copy number profiling of cell-free DNA as a precision oncology screening strategy.

    Current cell-free DNA (cfDNA) next generation sequencing (NGS) precision oncology workflows are typically limited to targeted and/or disease-specific applications. In advanced cancer, disease burden and cfDNA tumor content are often elevated, yielding unique precision oncology opportunities. We sought to demonstrate the utility of a pan-cancer, rapid, inexpensive, whole genome NGS of cfDNA approach (PRINCe) as a precision oncology screening strategy via ultra-low coverage (~0.01x) tumor content determination through genome-wide copy number alteration (CNA) profiling. We applied PRINCe to a retrospective cohort of 124 cfDNA samples from 100 patients with advanced cancers, including 76 men with metastatic castration-resistant prostate cancer (mCRPC), enabling cfDNA tumor content approximation and actionable focal CNA detection, while facilitating concordance analyses between cfDNA and tissue-based NGS profiles and assessment of cfDNA alteration associations with mCRPC treatment outcomes. Therapeutically relevant focal CNAs were present in 42 (34%) cfDNA samples, including 36 of 93 (39%) mCRPC patient samples harboring AR amplification. PRINCe identified pre-treatment cfDNA CNA profiles facilitating disease monitoring. Combining PRINCe with routine targeted NGS of cfDNA enabled mutation and CNA assessment with coverages tuned to cfDNA tumor content. In mCRPC, genome-wide PRINCe cfDNA and matched tissue CNA profiles showed high concordance (median Pearson correlation = 0.87), and PRINCe detectable AR amplifications predicted reduced time on therapy, independent of therapy type (Kaplan-Meier log-rank test, chi-square = 24.9, p &lt; 0.0001). Our screening approach enables robust, broadly applicable cfDNA-based precision oncology for patients with advanced cancer through scalable identification of therapeutically relevant CNAs and pre-/post-treatment genomic profiles, enabling cfDNA- or tissue-based precision oncology workflow optimization
