    Polyphenol extraction optimisation from Ceylon gooseberry (Dovyalis hebecarpa) pulp

    AbstractOriginally from Asia, Dovyalis hebecarpa is a dark purple/red exotic berry now also produced in Brazil. However, no reports were found in the literature about phenolic extraction or characterisation of this berry. In this study we evaluate the extraction optimisation of anthocyanins and total phenolics in D. hebecarpa berries aiming at the development of a simple and mild analytical technique. Multivariate analysis was used to optimise the extraction variables (ethanol:water:acetone solvent proportions, times, and acid concentrations) at different levels. Acetone/water (20/80 v/v) gave the highest anthocyanin extraction yield, but pure water and different proportions of acetone/water or acetone/ethanol/water (with >50% of water) were also effective. Neither acid concentration nor time had a significant effect on extraction efficiency allowing to fix the recommended parameters at the lowest values tested (0.35% formic acid v/v, and 17.6min). Under optimised conditions, extraction efficiencies were increased by 31.5% and 11% for anthocyanin and total phenolics, respectively as compared to traditional methods that use more solvent and time. Thus, the optimised methodology increased yields being less hazardous and time consuming than traditional methods. Finally, freeze-dried D. hebecarpa showed high content of target phytochemicals (319mg/100g and 1421mg/100g of total anthocyanin and total phenolic content, respectively)

    Dietary vitamin supplementation decrease stress caused by high stocking density, and boosts immunological system of farmed fish. A studied was carried out to determine vitamin A requirements of Nile tilapia (Oreochromis niloticus) in an all male group (13.8 ± 1.2 g) and a mixed sex population (9.8 ± 2.3 g). Fish stocked in 100-L plastic aquaria (26.0 ± 1.0ºC) were fed to near satiety, twice a day, seven days a week, during 75 days with vitamin A-free, semi-purified diets supplemented with 0; 600; 1,200; 1,800; 2,400; 3,000; 3,600; 4,200; 4,800 and 5,400 International Units (IU) of retinyl palmitate (30% vitamin A) per kg of diet in a completely randomized experimental design, factorial arrangement 2c10 (n = 4). Deficiency signs of vitamin A were observed in fish fed 0 to 1.200 IU vitamin A kg-1 diet; moderate signs were observed in fish fed diets with 1.800 to 3.600 IU vitamin A kg-1 diet; no interactions group*level (p 0.05). A group effect was observed regarding all performance variables (p 0,05). Foi observado efeito de grupo no desempenho dos peixes (p < 0,0001). Foi detectado o retinol hepático através de HPLC somente no grupo alimentado com 5.400 UI de retinol kg-1 de dieta, caracterizando assim que o mesmo foi utilizado e armazenado. A quantidade de 5.400 IU de retinol kg-1 de dieta é a mínima recomendada para tilápia do Nil

    The aim of the present study was determining the main organic acids in pulp and juices, as well as evaluating their stability, after opening the package, by liquid chromatography in a C18 column with isocratic elution and UV detection. In açaí pulp tartaric, malic and citric acids were found. Cashew samples presented all of the organic acids evaluated, besides high concentrations of ascorbic and malic acids. Acerola pulp had the highest ascorbic acid concentration. A small decrease in organic acid content during storage was observed. Malic and citric acids seem to be more stable than tartaric and ascorbic acids

    Broccoli is a vegetable consumed in many countries and a possible source of folates, which are water-soluble vitamins active during DNA synthesis. The folates found in the samples analyzed were 5-methyltetrahydrofolate and 5-formyltetrahydrofolate. The vitamin content varied between 413.7 and 742.2 µg/100 g for 5-methyltetrahydrofolate and from 4.8 to 12.8 µg/100 g for 5-formyltetrahydrofolate. In organic broccoli the amount of 5-methyltetrahydrofolate was significantly higher than in the same vegetable cultivated by traditional methods, for the commercial samples analyzed. The losses of these folates after cooking in water were of approximately 68%, most of it (53%) found in the cooking water

    Optimization of a colorimetric method to determine ascorbic acids in fruit jelly

    The AOAC standard methodology for determination of vitamin C in juices and preparations employs a titration with the indicator 2,6-di-chlorophenol-indophenol. This technique presents difficulties when samples with intense coloration are analyzed. Considering the need for a more adequate methodology to deal with these samples, adaptations of the traditional method were performed and their efficacy was verified. Comparative tests were performed on the standard methodology, the modified methodology and the chromatography method. Six different jelly fruits were analyzed and the proposed modifications were considered satisfactory, because these results were closest to the chromatographic results. The system proved to be linear, repeatable (standard deviation of 2.50%), with high recovery (102.10%) and an economy of 92.88% of reagents was achieved. Concentrations were between 0 to 6.08 mg of ascorbic acid per 100 g of sample.A metodologia indicada pela AOAC para a determinação de Vitamina C em sucos e preparados emprega uma titulação com o indicador 2,6-diclorofenol-indofenol. Esta técnica apresenta dificuldades quando são analisadas amostras com intensa coloração. Tendo em vista a necessidade de uma metodologia mais adequada para estas amostras, foram feitas modificações no método tradicional e sua eficácia foi comprovada. Testes comparativos foram realizados entre a metodologia original, a modificada e um método cromatográfico. Seis diferentes geleias de frutas foram analisadas e as modificações propostas foram satisfatórias, ao passo que os resultados foram mais semelhantes aos obtidos pelo método cromatográfico. O sistema apresentou-se linear, repetitivo (desvio padrão de 2,50%), com alta recuperação (102,10%), e uma economia de 92,88% de reagentes foi realizada.
