    Fusion of Bacterial Flagellin to a Dendritic Cell-Targeting αCD40 Antibody Construct Coupled With Viral or Leukemia-Specific Antigens Enhances Dendritic Cell Maturation and Activates Peptide-Responsive T Cells

    Conventional dendritic cell (DC) vaccine strategies, in which DCs are loaded with antigens ex vivo, suffer biological issues such as impaired DC migration capacity and laborious GMP production procedures. In a promising alternative, antigens are targeted to DC-associated endocytic receptors in vivo with antibody–antigen conjugates co-administered with toll-like receptor (TLR) agonists as adjuvants. To combine the potential advantages of in vivo targeting of DCs with those of conjugated TLR agonists, we generated a multifunctional antibody construct integrating the DC-specific delivery of viral- or tumor-associated antigens and DC activation by TLR ligation in one molecule. We validated its functionality in vitro and determined if TLR ligation might improve the efficacy of such a molecule. In proof-of-principle studies, an αCD40 antibody containing a CMV pp65-derived peptide as an antigen domain (αCD40CMV) was genetically fused to the TLR5-binding D0/D1 domain of bacterial flagellin (αCD40.FlgCMV). The analysis of surface maturation markers on immature DCs revealed that fusion of flagellin to αCD40CMV highly increased DC maturation (3.4-fold elevation of CD80 expression compared to αCD40CMV alone) by specifically interacting with TLR5. Immature DCs loaded with αCD40.FlgCMV induced significantly higher CMVNLV-specific T cell activation and proliferation compared to αCD40CMV in co-culture experiments with allogeneic and autologous T cells (1.8-fold increase in % IFN-Îł/TNF-α+ CD8+ T cells and 3.9-fold increase in % CMVNLV-specific dextramer+ CD8+ T cells). More importantly, we confirmed the beneficial effects of flagellin-dependent DC stimulation using a tumor-specific neoantigen as the antigen domain. Specifically, the acute myeloid leukemia (AML)-specific mutated NPM1 (mNPM1)-derived neoantigen CLAVEEVSL was delivered to DCs in the form of αCD40mNPM1 and αCD40.FlgmNPM1 antibody constructs, making this study the first to investigate mNPM1 in a DC vaccination context. Again, αCD40.FlgmNPM1-loaded DCs more potently activated allogeneic mNPM1CLA-specific T cells compared to αCD40mNPM1. These in vitro results confirmed the functionality of our multifunctional antibody construct and demonstrated that TLR5 ligation improved the efficacy of the molecule. Future mouse studies are required to examine the T cell-activating potential of αCD40.FlgmNPM1 after targeting of dendritic cells in vivo using AML xenograft models

    Die molekularen Parameter PSMA-TV und TL-PSMA im 68Ga-PSMA PET/CT leiten sich ab von MTV und TLG im FDG PET/CT. Mit der vorliegenden Arbeit wurden die Grenzen neuer Autosegmentierungsprogramme durch eine maximale Belastung mit großen Tumorvolumina von Patienten unter taxanbasierter Chemotherapie ausgelotet. Die Programme Syngo.via und FIJI kamen zu vergleichbaren Ergebnissen. Patienten mit einem Gleason Score von 8-10 zeigten unter Therapie eine signifikante Zunahme des PSMA-TV und TL-PSMA im Gegensatz zu Patienten mit Gleason Score 6-7b. Ein hoher PSA-Wert korrelierte zu allen Zeitpunkten signifikant mit einem hohen PSMA-TV und TL-PSMA, ebenso korrelierte ein steigender PSA-Wert signifikant mit steigenden Werten in PSMA-TV und TL-PSMA. Patienten mit einem biochemischen Progress und einem Progress nach modifiziertem PERCIST zeigten vor Therapie ein signifikant höheres PSMA-TV und TL-PSMA als Patienten ohne Progress und unter Therapie eine signifikante Zunahme des PSMA-TV und TL-PSMA im Vergleich zu Patienten ohne Progress. Eine Einteilung des Therapieansprechens aller Patienten in CR, PR, SD und PD nach PSMA-TV, TL-PSMA, PSA-Wert und modifiziertem PERCIST stimmte nicht in allen Patienten ĂŒberein. Ein signifikant kĂŒrzeres GesamtĂŒberleben zeigten lediglich Patienten mit einem nach dem PSA-Wert definiertem Progress. Im praktischen Vergleich der beiden Programme benötigte Syngo.via fĂŒr eine komplette Segmentierung signifikant mehr Zeit als FIJI, vor allem da der Wechsel von VOI zu VOI signifikant lĂ€nger dauerte. UnabhĂ€ngig vom Autosegmentierungsprogramm dauerte eine komplette Segmentierung lĂ€nger, je grĂ¶ĂŸer das PSMA-TV und das TL-PSMA war, je mehr VOIs das Programm automatisch setzte und je mehr VOIs manuell gelöscht und neu gesetzt wurden. In der Gesamtschau bieten PSMA-TV und TL-PSMA in Kombination mit den sich schnell weiterentwickelnden Autosegmentierungs-Programmen die Möglichkeit, auch sehr hohe Tumorlasten des PCas objektiv und vergleichbar zu beschreiben.The molecular parameters PSMA-TV and TL-PSMA in 68Ga-PSMA PET/CT are derived from MTV and TLG in FDG PET/CT. The present work explored the limits of new autosegmentation programs with large tumor volumes from patients receiving taxane-based chemotherapy. The programs Syngo.via and FIJI achieved comparable results. Patients with a Gleason score of 8-10 showed a significant increase in PSMA-TV and TL-PSMA during therapy in contrast to patients with Gleason scores 6-7b. A high PSA value was significantly correlated with a high PSMA-TV and TL-PSMA at all time points, and an increasing PSA value was also significantly correlated with increasing values in PSMA-TV and TL-PSMA. Patients with biochemical progression and progression according to modified PERCIST showed a significantly higher PSMA-TV and TL-PSMA before therapy than patients without progression and a significant increase in PSMA-TV and TL-PSMA during therapy compared to patients without progression. A classification of therapy response in all patients into CR, PR, SD and PD based on PSMA-TV, TL-PSMA, PSA value and modified PERCIST was not consistent in all patients. Only patients with progression defined by the PSA value showed a significantly shorter overall survival. In a practical comparison of the two programs, Syngo.via required significantly more time for complete segmentation than FIJI, especially since switching from VOI to VOI took significantly longer. Regardless of the auto-segmentation program, complete segmentation took longer as the PSMA-TV and TL-PSMA increased, and as the program automatically set more VOIs and more VOIs were manually deleted and reset. Overall, PSMA-TV and TL-PSMA in combination with the rapidly developing auto-segmentation programs offer the possibility of objectively and comparably describing even very high PCa tumor burdens

    Temporally resolved intra-annual wood density variations in European beech (Fagus sylvatica L.) as affected by climate and aspect

    We investigated the temporal variability of intra-annual wood density variations in European beech (Fagus sylvatica L.) in a valley in southwestern Germany. Samples were collected from 11 beech trees growing at north-west (NW) and south-west (SW) exposed slopes. High-frequency densitometry was used to obtain wood density profiles. We converted radial positions within these profiles to a seasonal time scale over automatic point dendrometer data for the period 2001-2006. Temporally resolved wood density data was analyzed both visually and statistically, using correlation analysis and multiple linear regressions. Water availability was found to be of major importance for wood formation. Further, our results suggest that climatic forcing of wood density is not necessarily restricted to the late growing season only, but that strong associations may exist during a major part of the growing season. Combining wood property data with point dendrometer measurements was demonstrated to be valuable for increasing the understanding on the effects of changing environmental conditions on wood formation.</span

    Metabolic tumour volume from PSMA PET/CT scans of prostate cancer patients during chemotherapy - do different software solutions deliver comparable results?

    Background: Prostate-specific membrane antigen (PSMA)-derived tumour volume (PSMA-TV) and total lesion PSMA (TL-PSMA) from PSMA PET/CT scans are promising biomarkers for assessing treatment response in prostate cancer (PCa). Currently, it is unclear whether different software tools for assessing PSMA-TV and TL-PSMA produce comparable results. (2) Methods: 68Ga-PSMA PET/CT scans from n = 21 patients with castration-resistant PCa (CRPC) receiving chemotherapy were identified from our single-centre database. PSMA-TV and TL-PSMA were calculated with Syngo.via (Siemens) as well as the freely available Beth Israel plugin for FIJI (Fiji Is Just ImageJ) before and after chemotherapy. While statistical comparability was illustrated and quantified via Bland-Altman diagrams, the clinical agreement was estimated by matching PSMA-TV, TL-PSMA and relative changes of both variables during chemotherapy with changes in serum PSA (ΔPSA) and PERCIST (Positron Emission Response Criteria in Solid Tumors). (3) Results: Comparing absolute PSMA-TV and TL-PSMA as well as Bland–Altman plotting revealed a good statistical comparability of both software algorithms. For clinical agreement, classifying therapy response did not differ between PSMA-TV and TL-PSMA for both software solutions and showed highly positive correlations with BR. (4) Conclusions: due to the high levels of statistical and clinical agreement in our CRPC patient cohort undergoing taxane chemotherapy, comparing PSMA-TV and TL-PSMA determined by Syngo.via and FIJI appears feasible
