    Viral suppression of multiple escape mutants by de novo CD8+ T cell responses in a human immunodeficiency virus-1 Infected elite suppressor

    Elite suppressors or controllers (ES) are HIV-1 infected patients who maintain undetectable viral loads without treatment. While HLA-B*57-positive ES are usually infected with virus that is unmutated at CTL epitopes, a single, dominant variant containing CTL escape mutations is typically seen in plasma during chronic infection. We describe an ES who developed seven distinct and rare escape variants at an HLA-B*57-restricted Gag epitope over a five year period. Interestingly, he developed proliferative, de novo CTL responses that suppressed replication of each of these variants. These responses, in combination with low viral fitness of each variant, may contribute to sustained elite control in this ES

    Design of a Pico-Satellite for the Monitoring of the Performance of a Thin Film Solar Array

    This paper presents an overview of the design and mission of a pico-satellite designed to monitor the performance of a Thin Film Solar Array (TFSA) over a one-week period. TFSA is a solar array technology that allows solar cells to be deposited onto a thin, flexible substrate. This substrate can be easily folded, which allows large solar arrays to collapse into a small space. The Aerospace Corporation has developed a small deployable solar array based on this technology. During this mission, the pico-satellite will deploy the TFSA. Once deployed, the voltage and current generated by the array will be monitored. The majority of the pico-satellite’s subsystems are constructed from off-the-shelf components that have been modified for space flight. These are a Kenwood amateur radio communications system, a Basic-X micro-controller based computer system, and a power system. These components, along with the ability to be launched as a secondary payload help reduce costs. This mission is ideally suited for a pico-size satellite due to its short duration, low power requirements, and lack of pointing requirements. It is hoped that information gathered on this mission will allow larger TFSA’s to be built in the future, enabling them to act as primary power sources for future satellites

    Genetic-geographic correlation revealed across a broad European ecotypic sample of perennial ryegrass (Lolium perenne) using array-based SNP genotyping

Applied to 90 European ecotypes composed of 716 individuals identifies a significant genetic–geographic correlation. ABSTRACT: Grassland ecosystems are ubiquitous across temperate and tropical regions, totalling 37 % of the terrestrial land cover of the planet, and thus represent a global resource for understanding local adaptations to environment. However, genomic resources for grass species (outside cereals) are relatively poor. The advent of next-generation DNA sequencing and high-density SNP genotyping platforms enables the development of dense marker assays for population genetics analyses and genome-wide association studies. A high-density SNP marker resource (Illumina Infinium assay) for perennial ryegrass (Lolium perenne) was created and validated in a broad ecotype collection of 716 individuals sampled from 90 sites across Europe. Genetic diversity within and between populations was assessed. A strong correlation of geographic origin to genetic structure was found using principal component analysis, with significant correlation to longitude and latitude (P < 0.001). The potential of this array as a resource for studies of germplasm diversity and identifying traits underpinning adaptive variation is highlighted.     Duplication of the mitochondrial control region is associated with increased longevity in birds

    Despite a number of biochemical and lifestyle differences which should increase risk of oxidative damage to their mitochondrial DNA (mtDNA) and thus reduce expected lifespan, avian species often display longer lifespans than mammals of similar body mass. Recent work in mammalian ageing has demonstrated that functional mitochondrial copy number declines with age. We noted that several bird species display duplication of the control region (CR) of the mtDNA to form a pseudo-control region (YCR), apparently an avian-specific phenomenon. To investigate whether the presence of this duplication may play a similar role in longevity to mitochondrial copy number in mammals, we correlated body mass and longevity in 92 avian families and demonstrate a significant association. Furthermore, outlier analysis demonstrated a significant (p=0.01) difference associated with presence of the YCR duplication in longer-lived avian species. Further research is required to determine if the YCR does indeed alter mitochondrial function or resilience to oxidative damage, but these findings provide an intriguing hint of how mitochondrial sequences may be related to an extended lifespan

    Germplasm dynamics:The role of ecotypic diversity in shaping the patterns of genetic variation in Lolium perenne

    Perennial ryegrass (Lolium perenne) is the most widely grown temperate grass species globally. Intensive plant breeding in ryegrass compared to many other crops species is a relatively recent exercise (last 100 years) and provides an interesting experimental system to trace the extent, impact and trajectory of undomesticated ecotypic variation represented in modern ryegrass cultivars. To explore germplasm dynamics in Lolium perenne, 2199 SNPs were genotyped in 716 ecotypes sampled from 90 European locations together with 249 cultivars representing 33 forage/amenity accessions. In addition three pseudo-cross mapping populations (450 individual recombinants) were genotyped to create a consensus genetic linkage map. Multivariate analyses revealed strong differentiation between cultivars with a small proportion of the ecotypic variation captured in improved cultivars. Ryegrass cultivars generated as part of a recurrent selection programme (RSP) are strongly associated with a small number of geographically localised Italian ecotypes which were among the founders of the RSP. Changes in haplotype frequency revealed signatures of selection in genes putatively involved in water-soluble carbohydrate (WSC) accumulation (a trait selected in the RSP). Retrospective analysis of germplasm in breeding programmes (germplasm dynamics) provides an experimental framework for the identification of candidate genes for novel traits such as WSC accumulation in ryegrass
