    Corollary discharge inhibition of ascending auditory neurons in the stridulating cricket

    Acoustically communicating animals are able to process external acoustic stimuli despite generating intense sounds during vocalization. We have examined how the crickets' ascending auditory pathway copes with self-generated, intense auditory signals (chirps) during singing (stridulation). We made intracellular recordings from two identified ascending auditory interneurons, ascending neuron 1 (AN1) and ascending neuron 2 (AN2), during pharmacologically elicited sonorous (two-winged), silent (one-winged), and fictive (isolated CNS) stridulation. During sonorous chirps, AN1 responded with bursts of spikes, whereas AN2 was inhibited and rarely spiked. Low-amplitude hyperpolarizing potentials were recorded in AN1 and AN2 during silent chirps. The potentials were also present during fictive chirps. Therefore, they were the result of a centrally generated corollary discharge from the stridulatory motor network. The spiking response of AN1 and AN2 to acoustic stimuli was inhibited during silent and fictive chirps. The maximum period of inhibition occurred in phase with the maximum spiking response to self-generated sound in a sonorously stridulating cricket. In some experiments (30%) depolarizing potentials were recorded during silent chirps. Reafferent feedback elicited by wing movement was probably responsible for the depolarizing potentials. In addition, two other sources of inhibition were present in AN1: (1) IPSPs were elicited by stimulation with 12.5 kHz stimuli and (2) a long-lasting hyperpolarization followed spiking responses to 4.5 kHz stimuli. The hyperpolarization desensitized the response of AN1 to subsequent quieter stimuli. Therefore, the corollary discharge will reduce desensitization by suppressing the response of AN1 to self-generated sounds

    Motor-Incompatibility of Facial Reactions : The influence of valence and stimulus content on voluntary facial reactions

    Emotional cues facilitate motor responses that are associated with approach or avoidance. Previous research has shown that evaluative processing of positive and negative facial expression stimuli is also linked to motor schemata of facial muscles. To further investigate the influence of different types of emotional stimuli on facial reactions, we conducted a study with pictures of emotional facial expressions (KDEF) and scenes (IAPS). Healthy participants were asked to respond to the positive or negative facial expressions (KDEF) and scenes (IAPS) with specific facial muscles in a valence-congruent (stimulus valence matches muscle related valence) or a valence-incongruent condition (stimulus valence is contrary to muscle related valence). Additionally, they were asked to rate pictures in terms of valence and arousal. Muscular response latencies were recorded by an electromyogram. Overall, response latencies were shorter in response to facial expressions than to complex pictures of scenes. For both stimulus categories, response latencies with valence-compatible muscles were shorter compared to reactions with incompatible muscles. Moreover, correlations between picture ratings and facial muscle reactions for happy facial expressions as well as positive scenes reflect a direct relationship between perceived intensity of the subjective emotional experience and physiological responding. Results replicate and extend previous research, indicating that incompatibility effects are reliable across different stimulus types and are not limited to facial mimicry

    Electrophoresis of polar fluorescent tracers through the nerve sheath labels neuronal populations for anatomical and functional imaging

    The delivery of tracers into populations of neurons is essential to visualize their anatomy and analyze their function. In some model systems genetically-targeted expression of fluorescent proteins is the method of choice; however, these genetic tools are not available for most organisms and alternative labeling methods are very limited. Here we describe a new method for neuronal labelling by electrophoretic dye delivery from a suction electrode directly through the neuronal sheath of nerves and ganglia in insects. Polar tracer molecules were delivered into the locust auditory nerve without destroying its function, simultaneously staining peripheral sensory structures and central axonal projections. Local neuron populations could be labelled directly through the surface of the brain, and in-vivo optical imaging of sound-evoked activity was achieved through the electrophoretic delivery of calcium indicators. The method provides a new tool for studying how stimuli are processed in peripheral and central sensory pathways and is a significant advance for the study of nervous systems in non-model organisms.MDI was funded by Howard Hughes Medical Institute, the equipment used in this study was funded by the BBSRC (BB/G018723/1)

    Substrate texture affects female cricket walking response to male calling song.

    Field crickets are extensively used as a model organism to study female phonotactic walking behaviour, i.e. their attraction to the male calling song. Laboratory-based phonotaxis experiments generally rely on arena or trackball-based settings; however, no attention has been paid to the effect of substrate texture on the response. Here, we tested phonotaxis in female Gryllus bimaculatus, walking on trackballs machined from methyl-methacrylate foam with different cell sizes. Surface height variations of the trackballs, due to the cellular composition of the material, were measured with profilometry and characterized as smooth, medium or rough, with roughness amplitudes of 7.3, 16 and 180 µm. Female phonotaxis was best on a rough and medium trackball surface, a smooth surface resulted in a significant lower phonotactic response. Claws of the cricket foot were crucial for effective walking. Females insert their claws into the surface pores to allow mechanical interlocking with the substrate texture and a high degree of attachment, which cannot be established on smooth surfaces. These findings provide insight to the biomechanical basis of insect walking and may inform behavioural studies that the surface texture on which walking insects are tested is crucial for the resulting behavioural response.This study is supported by a Mexican CONACyT Cambridge Trust Scholarship to E.J.S.P., Newnham College, the Royal Entomological Society, the Philosophical Society and the Department of Zoology.     The binaural masking level difference: cortical correlates persist despite severe brain stem atrophy in progressive supranuclear palsy.

    Under binaural listening conditions, the detection of target signals within background masking noise is substantially improved when the interaural phase of the target differs from that of the masker. Neural correlates of this binaural masking level difference (BMLD) have been observed in the inferior colliculus and temporal cortex, but it is not known whether degeneration of the inferior colliculus would result in a reduction of the BMLD in humans. We used magnetoencephalography to examine the BMLD in 13 healthy adults and 13 patients with progressive supranuclear palsy (PSP). PSP is associated with severe atrophy of the upper brain stem, including the inferior colliculus, confirmed by voxel-based morphometry of structural MRI. Stimuli comprised in-phase sinusoidal tones presented to both ears at three levels (high, medium, and low) masked by in-phase noise, which rendered the low-level tone inaudible. Critically, the BMLD was measured using a low-level tone presented in opposite phase across ears, making it audible against the noise. The cortical waveforms from bilateral auditory sources revealed significantly larger N1m peaks for the out-of-phase low-level tone compared with the in-phase low-level tone, for both groups, indicating preservation of early cortical correlates of the BMLD in PSP. In PSP a significant delay was observed in the onset of the N1m deflection and the amplitude of the P2m was reduced, but these differences were not restricted to the BMLD condition. The results demonstrate that although PSP causes subtle auditory deficits, binaural processing can survive the presence of significant damage to the upper brain stem.This work has been supported by the Wellcome Trust (Grants 088324 and 088263); Medical Research Council (G0700503 to B.     The general theme of the 2020 Community Service Program is “Community Empowerment”. Meanwhile, the special theme is "Entrepreneurship". The implementation time starts from August 3 to September 3, 2020, in Onekore Village, Ende Tengah District, Ende Regency. On the program. With this special theme, the main activity carried out is supporting entrepreneurship in Onekore Village. The purpose of implementing this activity is that students are expected to gain learning experience through direct involvement in the community in finding, formulating, solving, and overcoming problems in the field. In this KKN activity, the problems found were the lack of utilization of existing garden products, namely bananas. So far, it is consumed by boiling or eating it without further processing. Olso besides, we also found woven products from weaving that were difficult to market. Students socialize and practice entrepreneurship as an effort to solve problems faced by weaving craftsmen. Efforts made to help the household economy include making flower pots and making wallets made from woven fabrics. Supporting activities during the Community Service Program are assisting with the administration of the Village, community service, building a gate for the Indonesian Independence Day, making sample gardens, socializing COVID-19 and, health service for the elderly
