    Flexibility in the receptor-binding domain of the enzymatic colicin E9 is required for toxicity against Escherichia coli cells

    The events that occur after the binding of the enzymatic E colicins to Escherichia coli BtuB receptors that lead to translocation of the cytotoxic domain into the periplasmic space and, ultimately, cell killing are poorly understood. It has been suggested that unfolding of the coiled-coil Mull receptor binding domain of the E colicins may be an essential step that leads to the loss of immunity protein from the colicin and immunity protein complex and then triggers the events of translocation. We introduced pairs of cysteine mutations into the receptor binding domain of colicin E9 (ColE9) that resulted in the formation of a disulfide bond located near the middle or the top of the R domain. After dithiothreitol reduction, the ColE9 protein with the mutations L359C and F412C (ColE9 L359C-F412C) and the ColE9 protein with the mutations Y324C and L447C (ColE9 Y324C-L447C) were slightly less active than equivalent concentrations of ColE9. On oxidation with diamide, no significant biological activity was seen with the ColE9 L359C-F412C and the ColE9 Y324C-L447C mutant proteins; however diamide had no effect on the activity of ColE9. The presence of a disulfide bond was confirmed in both of the oxidized, mutant proteins by matrix-assisted laser desorption ionization-time of flight mass spectrometry. The loss of biological activity of the disulfide-containing mutant proteins was not due to an indirect effect on the properties of the translocation or DNase domains of the mutant colicins.     Quantifying the 2.5D imaging performance of digital holographic systems

    Digital holographic systems are a class of two step, opto-numerical, three-dimensional imaging techniques. The role of the digital camera in limiting the resolution and field of view of the reconstructed image, and the interaction of these limits with a general optical system is poorly understood. The linear canonical transform describes any optical system consisting of lenses and/or free space in a unified manner. Expressions derived using it are parametrised in terms of the parameters of the optical system, as well as those of the digital camera: aperture size, pixel size and pixel pitch. We develop rules of thumb for selecting an optical system to minimise mean squared error for given input and digital camera parameters. In the limit, our results constitute a point spread function analysis. The results presented in this paper will allow digital holography practitioners to select an optical system to maximise the quality of their reconstructed image using a priori knowledge of the camera and object

    XO-7 b: A Transiting Hot Jupiter with a Massive Companion on a Wide Orbit

    Transiting planets orbiting bright stars are the most favorable targets for follow-up and characterization. We report the discovery of the transiting hot Jupiter XO-7 b and of a second, massive companion on a wide orbit around a circumpolar, bright, and metal rich G0 dwarf (V = 10.52, Teff = 6250±100 K, [Fe/H] = 0.432 ± 0.057 dex). We conducted photometric and radial velocity follow-up with a team of amateur and professional astronomers. XO-7 b has a period of 2.8641424±0.0000043 days, a mass of 0.709±0.034 MJ, a radius of 1.373±0.026 RJ, a density of 0.340±0.027 g cm-3 , and an equilibrium temperature of 1743 ± 23 K. Its large atmospheric scale height and the brightness of the host star make it well suited to atmospheric characterization. The wide orbit companion is detected as a linear trend in radial velocities with an amplitude of ~ 100 m s-1 over two years, yielding a minimum mass of 4 MJ; it could be a planet, a brown dwarf, or a low mass star. The hot Jupiter orbital parameters and the presence of the wide orbit companion point towards a high eccentricity migration for the hot Jupiter. Overall, this system will be valuable to understand the atmospheric properties and migration mechanisms of hot Jupiters and will help constrain the formation and evolution models of gas giant exoplanets.The XO project is supported by NASA grant NNX10AG30G.     Atmospheric Correction for Hyperspectral Ocean Color Retrieval with Application to the Hyperspectral Imager for the Coastal Ocean (HICO)

    The classical multi-spectral Atmospheric Correction (AC) algorithm is inadequate for the new generation of spaceborne hyperspectral sensors such as NASA's first hyperspectral Ocean Color Instrument (OCI) onboard the anticipated Plankton, Aerosol, Cloud, ocean Ecosystem (PACE) satellite mission. The AC process must estimate and remove the atmospheric path radiance contribution due to the Rayleigh scattering by air molecules and scattering by aerosols from the measured top-of-atmosphere (TOA) radiance, compensate for the absorption by atmospheric gases, and correct for reflection and refraction of the air-sea interface. In this work, we present and evaluate an improved AC for hyperspectral sensors developed within NASA's Sea-viewing Wide Field-of-view Sensor (SeaWiFS) Data Analysis System software package (SeaDAS). The improvement is based on combining the classical AC approach of multi-spectral capabilities to correct for the atmospheric path radiance, extended to hyperspectral, with a gas correction algorithm to compensate for absorbing gases in the atmosphere, including water vapor. The SeaDAS-hyperspectral version is capable of operationally processing the AC of any hyperspectral airborne or spaceborne sensor. The new algorithm development was evaluated and assessed using the Hyperspectral Imager for Coastal Ocean (HICO) scenes collected at the Marine Optical BuoY (MOBY) site, and other SeaWiFS Bio-optical Archive and Storage System (SeaBASS) and AERosol Robotic NETwork - Ocean Color (AERONET-OC) coastal sites. A hyperspectral vicarious calibration was applied to HICO, showing the validity and consistency of HICO's ocean color products.     Protection Afforded by Fluoroquinolones in Animal Models of Respiratory Infections with Bacillus anthracis, Yersinia pestis, and Francisella tularensis

    Successful treatment of inhalation anthrax, pneumonic plague and tularemia can be achieved with fluoroquinolone antibiotics, such as ciprofloxacin and levofloxacin, and initiation of treatment is most effective when administered as soon as possible following exposure. Bacillus anthracis Ames, Yersinia pestis CO92, and Francisella tularensis SCHU S4 have equivalent susceptibility in vitro to ciprofloxacin and levofloxacin (minimal inhibitory concentration is 0.03 μg/ml); however, limited information is available regarding in vivo susceptibility of these infectious agents to the fluoroquinolone antibiotics in small animal models. Mice, guinea pig, and rabbit models have been developed to evaluate the protective efficacy of antibiotic therapy against these life-threatening infections. Our results indicated that doses of ciprofloxacin and levofloxacin required to protect mice against inhalation anthrax were approximately 18-fold higher than the doses of levofloxacin required to protect against pneumonic plague and tularemia. Further, the critical period following aerosol exposure of mice to either B. anthracis spores or Y. pestis was 24 h, while mice challenged with F. tularensis could be effectively protected when treatment was delayed for as long as 72 h postchallenge. In addition, it was apparent that prolonged antibiotic treatment was important in the effective treatment of inhalation anthrax in mice, but short-term treatment of mice with pneumonic plague or tularemia infections were usually successful. These results provide effective antibiotic dosages in mice, guinea pigs, and rabbits and lay the foundation for the development and evaluation of combinational treatment modalities

    The study of vancomycin use and its adverse reactions associated to patients of a brazilian university hospital

    <p>Abstract</p> <p>Background</p> <p>Vancomycin is an antibiotic of growing importance in the treatment of hospital infections, with particular emphasis on its value in the fight against methicillin-resistant <it>Staphylococcus aureus</it>. However its usage profile must be evaluated to assure maximum benefit and minimum risk.</p> <p>Findings</p> <p>A cross-sectional retrospective study was carried out among inpatients that received vancomycin in a Brazilian quaternary hospital. The occurrence of adverse reactions reported was evaluated in medical records relating to patients taking vancomycin during a one year period. Males comprised 52% (95% CI: 41.7-60.2%) of the sample population, with a mean age of 50.6 (95% CI: 47.2-54.0) years and mean treatment period of 9.7 (95% CI: 8.0-11.5) Days. It was verified that nephrotoxicity occurred in 18.4% (95% CI: 11.3-27.5) of patients, Red man syndrome occurred in 2% (95% CI 0.2-7.2), while the occurrence of thrombocytopenia was 7.1% (95% CI: 2.9-14.2).</p> <p>Conclusions</p> <p>It may be noted that even after 50 years of use, adverse reactions associated with vancomycin continue with high frequency, presenting a public health problem, especially considering its current use in cases of multidrug resistant infections. In this context, we emphasize the importance of intensive pharmacovigilance in hospital as a surveillance tool after drug approval by the sanitary authority.</p

    Relationship of Optimism and Suicidal Ideation in Three Groups of Patients at Varying Levels of Suicide Risk

    Optimism has been associated with reduced suicidal ideation, but there have been few studies in patients at high suicide risk. We analyzed data from three study populations (total N=319) with elevated risk of suicide: (1) patients with a recent acute cardiovascular event, (2) patients hospitalized for heart disease who had depression or an anxiety disorder, and (3) patients psychiatrically hospitalized for suicidal ideation or following a suicide attempt. For each study we analyzed the association between optimism (measured by the Life-Orientation Test-Revised) and suicidal ideation, and then completed an exploratory random effects meta-analysis of the findings to synthesize this data. The meta-analysis of the three studies showed that higher levels of self-reported optimism were associated with a lower likelihood of suicidal ideation (odds ratio [OR]=.89, 95% confidence interval [CI]=.85-.95, z=3.94, pz=3.57, pz=3.61,
