Investigation of dendritic cells as a vector for vaccine delivery
Dendritic cells (DC) are an attractive target for both vaccine targeting and use as a delivery vehicle due to their ability to direct innate and adaptive immune responses. In this study a procedure was developed where DC were employed as a vehicle to induce cell-mediated immune responses to the Gram negative bacillus Burkholderia pseudomallei. This bacterium is the causative agent of melioidosis, a disease endemicto areas of south-east Asia and northern Australia, to which presently no licensed vaccine exists.Purified DC were pulsed with heat-killed (HK) whole cell B. pseudomallei and used to immunise syngeneic mice. The use of this immunisation method elicited strong cellular immune responses against B. pseudomallei, which have historically been difficult to generate. Antibody responses following immunisation with DC were low. Booster immunisations of either a second dose of HK B. pseudomallei pulsed DC or HK B.pseudomallei in MPL + TDM adjuvant were administered to increase the immuneresponse. Immunised animals were challenged with fully virulent B. pseudomallei and protection was demonstrated in animals immunised with a combination of HK B. pseudomallei pulsed DC and HK B. pseudomallei delivered in MPL + TDM adjuvant. The protective immune response demonstrated was then further elucidated to reveal arole for both cell-mediated and humoral immunity in protection against B. pseudomallei infection. The data also demonstrate the potential for DC as vaccine delivery vectors and as a screening mechanism for the evaluation of potential vaccine candidates
Technology for an intelligent, free-flying robot for crew and equipment retrieval in space
Crew rescue and equipment retrieval is a Space Station Freedom requirement. During Freedom's lifetime, there is a high probability that a number of objects will accidently become separated. Members of the crew, replacement units, and key tools are examples. Retrieval of these objects within a short time is essential. Systems engineering studies were conducted to identify system requirements and candidate approaches. One such approach, based on a voice-supervised, intelligent, free-flying robot was selected for further analysis. A ground-based technology demonstration, now in its second phase, was designed to provide an integrated robotic hardware and software testbed supporting design of a space-borne system. The ground system, known as the EVA Retriever, is examining the problem of autonomously planning and executing a target rendezvous, grapple, and return to base while avoiding stationary and moving obstacles. The current prototype is an anthropomorphic manipulator unit with dexterous arms and hands attached to a robot body and latched in a manned maneuvering unit. A precision air-bearing floor is used to simulate space. Sensor data include two vision systems and force/proximity/tactile sensors on the hands and arms. Planning for a shuttle file experiment is underway. A set of scenarios and strawman requirements were defined to support conceptual development. Initial design activities are expected to begin in late 1989 with the flight occurring in 1994. The flight hardware and software will be based on lessons learned from both the ground prototype and computer simulations
A Statistical Description of AGN Jet Evolution from the VLBA Imaging and Polarimetry Survey (VIPS)
A detailed analysis of the evolution of the properties of core-jet systems
within the VLBA Imaging and Polarimetry Survey (VIPS) is presented. We find a
power-law relationship between jet intensity and width that suggests for the
typical jet, little if any energy is lost as it moves away from its core. Using
VLA images at 1.5 GHz, we have found evidence that parsec-scale jets tend to be
aligned with the the direction of emission on kiloparsec scales. We also found
that this alignment improves as the jets move farther from their cores on
projected scales as small as ~50-100 pc. This suggests that realignment of jets
on these projected scales is relatively common. We typically find a modest
amount of bending (a change in jet position angle of ~5 deg.) on these scales,
suggesting that this realignment may typically occur relatively gradually.Comment: Accepted to ApJ, 20 pages, 8 figure
24R,25-Dihydroxyvitamin D3 Protects against Articular Cartilage Damage following Anterior Cruciate Ligament Transection in Male Rats
Osteoarthritis (OA) in humans is associated with low circulating 25-hydroxyvitamin D3 [25 (OH)D3]. In vitamin D replete rats, radiolabeled 24R,25-dihydroxyvitamin D3 [24R,25 (OH)2D3] accumulates in articular cartilage following injection of [3 H]-25(OH)D3. Previously, we showed that 24R,25(OH)2D3 blocks chondrocyte apoptosis via phospholipase D and p53, suggesting a role for 24R,25(OH)2D3 in maintaining cartilage health. We examined the ability of 24R,25(OH)2D3 to prevent degenerative changes in articular cartilage in an OAlike environment and the potential mechanisms involved. In vitro, rat articular chondrocytes were treated with IL-1β with and without 24R,25(OH)2D3 or 1α,25(OH)2D3. 24R,25(OH)2D3 but not 1α,25(OH)2D3 blocked the effects of IL-1β in a dose-dependent manner, and its effect was partially mediated through the TGF-β1 signaling pathway. In vivo, unilateral anterior cruciate ligament transections were performed in immunocompetent rats followed by intra-articular injections of 24R,25(OH)2D3 or vehicle (t = 0, 7, 14, 21 days). Tissues were harvested on day 28. Joints treated with vehicle had changes typical of OA whereas joints treated with 24R,25(OH)2D3 had less articular cartilage damage and levels of inflammatory mediators. These results indicate that 24R,25(OH)2D3 protects against OA, and suggest that it may be a therapeutic approach for preventing trauma-induced osteoarthritis

Prerequisites for Affective Signal Processing (ASP) - Part V: A response to comments and suggestions
In four papers, a set of eleven prerequisites for affective signal processing (ASP) were identified (van den Broek et al., 2010): validation, triangulation, a physiology-driven approach, contributions of the signal processing community, identification of users, theoretical specification, integration of biosignals, physical characteristics, historical perspective, temporal construction, and real-world baselines. Additionally, a review (in two parts) of affective computing was provided. Initiated by the reactions on these four papers, we now present: i) an extension of the review, ii) a post-hoc analysis based on the eleven prerequisites of Picard et al.(2001), and iii) a more detailed discussion and illustrations of temporal aspects with ASP
Gendered experiences of academic staff in relation to research activity and the REF2014
This report is based on research commissioned by the institutional Research and Knowledge Transfer Office between June 2015 and June 2016. This research has focused on generating qualitative and quantitative data as to the potential reasons why there appears to be a gender disparity in research productivity within the commissioning institution. In particular, the number of women self-selecting for representation in the REF2014 was comparatively low. This research was led by Dr Chantal Davies (as part of her broader remit in relation to the Forum for Research into Equality and Diversity) with Dr Ruth Healey as co-researcher and Anthony Cliffe as research assistant. A Steering Group made up of representatives from across the institution oversaw the process
