    Tribomaterial factors in space mechanism brake performance

    The asbestos/phenolic pads of Shuttle Remote Manipulator System (SRMS) brakes are unsuitable for use in long life space mechanisms because their friction decreases on extended sliding in high vacuum. Dehydration of the material and accumulation of wear debris in the conforming interface of this tribosystem induces the permanent friction changes. Other polymer and some ceramic based materials exhibit similar frictional torque behavior due to the development of minimal contact patches by the interfacial debris. In contrast, high friction occurs when other ceramics form many small contacts throughout fine debris beds. Generating this latter interfacial structure during run-in ensures that the in-vacuo friction remains stable thereafter. Such materials with low wear rates are potential candidates for friction elements in SSRMS and similar mechanisms

    Effectiveness of Helminth Therapy in the Prevention of Allograft Rejection: A Systematic Review of Allogeneic Transplantation.

    Background: The unique immunomodulatory capacity of helminth parasites has been investigated as a novel strategy in the prevention of allograft rejection after transplantation. This review was conducted to fully evaluate the specific effects of helminth therapy on allograft survival reported in published studies of animal models of allogeneic transplantation. Method: Following PRISMA protocol guidelines, a literature search was conducted using PubMed, MEDLINE via OvidSP, along with additional manual searches of selected reference lists. Publications describing helminth intervention within allograft transplantation models were screened for relevance to eligibility criteria. Primary and secondary outcomes were extracted using standardized data collection tables. The SYRCLE risk of bias assessment tool was used for quality assessment. Due to heterogeneity of study designs, meta-analysis could not be performed; rather outcomes are presented as a narrative synthesis with concept mapping. This review was registered in PROSPERO with ID: CRD42018097175. Results: The literature search generated 1,443 publications, which after screening for relevance to the eligibility criteria yielded 15 publications for qualitative analysis. All 15 publications reported improvement to allograft survival as a result of helminth therapy. This prolonged allograft survival was not significantly different when helminth-derived products were used compared to live infection. However, the extent of positive impact on allograft survival was noted to be dependent on study design factors, such as the chronicity of the live helminth infection, allograft type and the species/genus of helminth selected. Conclusion: Both live and product-based helminth therapy have potential applications as novel immune regulators or adjuncts for the prevention of allograft rejection. However, there were differences in efficacy between different worms and preparations of worm-derived products. Therefore, further studies are required to determine the most appropriate worm for a specific allograft, to elucidate the optimal dose and route of administration, and to better understand the modulation of immune responses that can mediate tolerance

    Molecular determinants of ginkgolide binding in the glycine receptor pore

    Ginkgolides are potent blockers of the glycine receptor Cl- channel (GlyR) pore. We sought to identify their binding sites by comparing the effects of ginkgolides A, B and C and bilobalide on alpha1, alpha2, alpha1beta and alpha2beta GlyRs. Bilobalide sensitivity was drastically reduced by incorporation of the beta subunit. In contrast, the sensitivities to ginkgolides B and C were enhanced by beta subunit expression. However, ginkgolide A sensitivity was increased in the alpha2beta GlyR relative to the alpha2 GlyR but not in the alpha1beta GlyR relative to the alpha1 GlyR. We hypothesised that the subunit-specific differences were mediated by residue differences at the second transmembrane domain 2' and 6' pore-lining positions. The increased ginkgolide A sensitivity of the alpha2beta GlyR was transferred to the alpha1beta GlyR by the G2'A (alpha1 to alpha2 subunit) substitution. In addition, the alpha1 subunit T6'F mutation abolished inhibition by all ginkgolides. As the ginkgolides share closely related structures, their molecular interactions with pore-lining residues were amenable to mutant cycle analysis. This identified an interaction between the variable R2 position of the ginkgolides and the 2' residues of both alpha1 and beta subunits. These findings provide strong evidence for ginkgolides binding at the 2' pore-lining position

    Relationship of glucose to prevalence of ECG abnormalities at baseline and to 6-yr mortality in Scottish males aged 45-64 yr

    A community study of more than 3000 persons screened in the Burgh of Renfrew, Scotland, included 1134 men age 45-64 whose baseline casual glucose level was measured in 1972 and who were followed for 6 yr to determine mortality rate. A positive association was found, in univariate analysis, between baseline glucose level and prevalence of ECG abnormalities as defined both by the London Whitehall Study and the U.S. Pooling Project Study. In multivariate analyses of these cross-sectional data, an association was seen for Whitehall but not Pooling Project abnormalities. No association was found between asymptomatic hyperglycemia and coronary mortality at 6 yr, either in univariate, bivariate or multivariate analyses. The clinically-observed excess in coronary mortality among those with symptomatic diabetes mellitus was not in evidence among those with asymptomatic hyperglycemia in this group of middle-aged men.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/23750/1/0000723.pd

    Resolved Spectroscopy of the Narrow-Line Region in NGC 1068. II. Physical Conditions Near the NGC 1068 ``Hot-Spot''

    The physical conditions near the optical continuum peak (``hot spot'') in the inner narrow line region (NLR) of the Seyfert 2 galaxy, NGC 1068. Spectra were taken with HST/STIS through the 0.1X52 arcsec slit, covering the full STIS 1200 to 10000 Angstrom waveband, and are from a region that includes the hot spot, extending 0.2, or ~ 14 pc (for H= 75 km/sec/Mpc). Perhaps the most striking feature of these spectra is the presence of strong coronal emission lines, including [S XII] 7611 which has hitherto only been identified in spectra of the solar corona. There is an apparent correlation between ionization energy and velocity of the emission lines with respect to the systemic velocity of the host galaxy, with the coronal lines blueshifted, most other high excitation lines near systemic, and some of the low ionization lines redshifted. From the results of our modeling, we find that the emission-line gas consists of three principal components: 1) one in which most of the strong emission-lines, such as [O III] 5007, [Ne V] 3426, C IV 1550, arise, 2) a more tenuous, highly ionized component, which is the source of the coronal-line emission, and 3) a component, which is not co-planar with the other two, in which the low ionization and neutral lines, such as [N II] 6548 and [O I] 6300, are formed. The first two components are directly ionized by the EUV-Xray continuum emitted by the central source, while the low ionization gas is ionized by a combination of highly absorbed continuum radiation and a small fraction of unabsorbed continuum scattered by free electrons associated with the hot spot. The combination of covering factor and Thomson optical depth of the high ionization components is insufficient to scatter the observed fraction of continuum radiation into our line-of-sight.Comment: 42 pages, Latex, includes 5 figures (postscript), to appear in the Astrophysical Journa

    Effect of poly(ethylene glycol) on insulin stability and cutaneous cell proliferation in vitro following cytoplasmic delivery of insulin-loaded nanoparticulate carriers – A potential topical wound management approach

    We describe the development of a nanoparticulate system, with variation of poly(ethylene glycol) (PEG) content, capable of releasing therapeutic levels of bioactive insulin for extended periods of time. Recombinant human insulin was encapsulated in poly(d,l-lactide-co-glycolide) nanoparticles, manufactured with variation in poly(ethylene glycol) content, and shown to be stable for 6days using SDS-PAGE, western blot and MALDI MS. To determine if insulin released from this sustained release matrix could stimulate migration of cell types normally active in dermal repair, a model wound was simulated by scratching confluent cultures of human keratinocytes (HaCaT) and fibroblasts (Hs27). Although free insulin was shown to have proliferative effect, closure of in vitro scratch fissures was significantly faster following administration of nano-encapsulated insulin. This effect was more pronounced in HaCaT cells when compared to Hs27 cells. Variation in PEG content had the greatest effect on NP size, with a lesser influence on scratch closure times. Our work supports a particulate uptake mechanism that provides for intracellular insulin delivery, leading to enhanced cell proliferation. When placed into an appropriate topical delivery vehicle, such as a hydrogel, the extended and sustained topical administration of active insulin delivered from a nanoparticulate vehicle shows promise in promoting tissue healing

    Tuberculosis incidence correlates with sunshine : an ecological 28-year time series study

    Birmingham is the largest UK city after London, and central Birmingham has an annual tuberculosis incidence of 80 per 100,000. We examined seasonality and sunlight as drivers of tuberculosis incidence. Hours of sunshine are seasonal, sunshine exposure is necessary for the production of vitamin D by the body and vitamin D plays a role in the host response to tuberculosis. Methods: We performed an ecological study that examined tuberculosis incidence in Birmingham from Dec 1981 to Nov 2009, using publicly-available data from statutory tuberculosis notifications, and related this to the seasons and hours of sunshine (UK Meteorological Office data) using unmeasured component models. Results: There were 9,739 tuberculosis cases over the study period. There was strong evidence for seasonality, with notifications being 24.1% higher in summer than winter (p<0.001). Winter dips in sunshine correlated with peaks in tuberculosis incidence six months later (4.7% increase in incidence for each 100 hours decrease in sunshine, p<0.001). Discussion and Conclusion: A potential mechanism for these associations includes decreased vitamin D levels with consequent impaired host defence arising from reduced sunshine exposure in winter. This is the longest time series of any published study and our use of statutory notifications means this data is essentially complete. We cannot, however, exclude the possibility that another factor closely correlated with the seasons, other than sunshine, is responsible. Furthermore, exposure to sunlight depends not only on total hours of sunshine but also on multiple individual factors. Our results should therefore be considered hypothesis-generating. Confirmation of a potential causal relationship between winter vitamin D deficiency and summer peaks in tuberculosis incidence would require a randomized-controlled trial of the effect of vitamin D supplementation on future tuberculosis incidence

    Applying Machine Learning to Neutron-Gamma Ray Discrimination from Scintillator Readout Using Wavelength Shifting Fibers

    Advances in machine learning have found wide applications including radiation detection. In this work, machine learning is applied to neutron-gamma ray discrimination of an organic liquid scintillator (OLS) readout using wavelength shifting (WLS) fibers. The objective of using WLS fiber is to enable the transfer of the light signal from the scintillation medium, with almost any active volume geometry, to a low-profile photomultiplier. This is a common practice in high-energy physics research and has proven to be very effective for such applications. The drawback of this approach is the light pulses carried to the photomultiplier through the WLS fibers do not perfectly replicate the original OLS light pulses’ intensities or timing. This drawback causes traditional pulse shape discrimination algorithms applied to the degraded light pulses to fail to discriminate between neutron and gamma ray events. However, differences in the degraded light pulses for neutrons and gamma rays still exist and various machine learning algorithms can be applied to identify these differences. An experimental system was constructed to simultaneously capture part of the scintillation medium signal and the corresponding signal through the WLS fibers. Using the known neutron-gamma ray discrimination characteristics directly measured in the scintillation medium to provide the ground truth, supervised machine learning algorithms were applied to the corresponding light pulses carried to the photomultiplier through the WLS fibers. The results indicate that this approach will enable enhanced recovery of neutron-gamma ray discrimination information. This research effort will focus on two aspects of the OLS-WLS system: 1) developing an experimental system to create machine learning training data and 2) applying and evaluating various machine learning algorithms

    The open future, bivalence and assertion

    It is highly intuitive that the future is open and the past is closed—whereas it is unsettled whether there will be a fourth world war, it is settled that there was a first. Recently, it has become increasingly popular to claim that the intuitive openness of the future implies that contingent statements about the future, such as ‘there will be a sea battle tomorrow,’ are non-bivalent (neither true nor false). In this paper, we argue that the non-bivalence of future contingents is at odds with our pre-theoretic intuitions about the openness of the future. These are revealed by our pragmatic judgments concerning the correctness and incorrectness of assertions of future contingents. We argue that the pragmatic data together with a plausible account of assertion shows that in many cases we take future contingents to be true (or to be false), though we take the future to be open in relevant respects. It follows that appeals to intuition to support the non-bivalence of future contingents is untenable. Intuition favours bivalence

    Health-related Quality of Life among hospitalized older people awaiting residential aged care

    BACKGROUND: Health related quality of life (HRQoL) in very late life is not well understood. The aim of the present study was to assess HRQoL and health outcomes at four months follow-up in a group of older people awaiting transfer to residential aged care. METHODS: Secondary analysis of data from a randomized controlled trial conducted in three public hospitals in Adelaide. A total of 320 patients in hospital beds awaiting a residential aged care bed participated. Outcome measurements included HRQoL (Assessment of Quality of Life; AQoL), functional level (Modified Barthel Index), hospital readmission rates, survival, and place of residence at four months follow-up. RESULTS: In this frail group the median AQoL was poor at baseline (median 0.02; 95%CI -0.01 - 0.04) and at follow-up (0.05; 95%CI 0.03 - 0.06). On leaving hospital, more than one third of participants who were moving for the first time into nursing home care rated themselves in a state worse than death (AQoL < or = 0.0). Poor HRQoL at discharge from hospital (AQoL < or = 0.0) was a significant predictor of mortality (HR 1.7; 95%CI 1.2 - 2.7), but not hospital readmission nor place of residence at four months follow-up. Improved function was a predictor of improved HRQoL among the surviving cohort. CONCLUSION: People making the transition to residential aged care from hospital have very poor HRQoL, but small gains in function seem to be related to improvement. While functional gains are unlikely to change discharge destination in this frail group, they can contribute to improvements in HRQoL.
