    Interferons Regulate the Phenotype of  Wild-type and Mutant Herpes Simplex Viruses In Vivo

    Mechanisms responsible for neuroattenuation of herpes simplex virus (HSV) have been defined previously by studies of mutant viruses in cultured cells. The hypothesis that null mutations in host genes can override the attenuated phenotype of null mutations in certain viral genes was tested. Mutants such as those in infected cell protein (ICP) 0, thymidine kinase, ribonucleotide reductase, virion host shutoff, and ICP34.5 are reduced in their capacity to replicate in nondividing cells in culture and in vivo. The replication of these viruses was examined in eyes and trigeminal ganglia for 1–7 d after corneal inoculation in mice with null mutations (−/−) in interferon receptors (IFNR) for type I IFNs (IFN-α/βR), type II IFN (IFN-γR), and both type I and type II IFNs (IFN-α/β/γR). Viral titers in eyes and ganglia of IFN-γR−/− mice were not significantly different from congenic controls. However, in IFN-α/βR−/− or IFN-α/β/γR−/− mice, growth of all mutants, including those with significantly impaired growth in cell culture, was enhanced by up to 1,000-fold in eyes and trigeminal ganglia. Blepharitis and clinical signs of infection were evident in IFN-α/βR−/− and IFN-α/β/γR−/− but not control mice for all viruses. Also, IFNs were shown to significantly reduce productive infection of, and spread from intact, but not scarified, corneas. Particularly striking was restoration of near-normal trigeminal ganglion replication and neurovirulence of an ICP34.5 mutant in IFN-α/βR−/− mice. These data show that IFNs play a major role in limiting mutant and wild-type HSV replication in the cornea and in the nervous system. In addition, the in vivo target of ICP34.5 may be host IFN responses. These experiments demonstrate an unsuspected role for host factors in defining the phenotypes of some HSV mutants in vivo. The phenotypes of mutant viruses therefore cannot be interpreted based solely upon studies in cell culture but must be considered carefully in the context of host factors that may define the in vivo phenotype

    Fronto-striatal organization: Defining functional and microstructural substrates of behavioural flexibility.

    Discrete yet overlapping frontal-striatal circuits mediate broadly dissociable cognitive and behavioural processes. Using a recently developed multi-echo resting-state functional MRI (magnetic resonance imaging) sequence with greatly enhanced signal compared to noise ratios, we map frontal cortical functional projections to the striatum and striatal projections through the direct and indirect basal ganglia circuit. We demonstrate distinct limbic (ventromedial prefrontal regions, ventral striatum - VS, ventral tegmental area - VTA), motor (supplementary motor areas - SMAs, putamen, substantia nigra) and cognitive (lateral prefrontal and caudate) functional connectivity. We confirm the functional nature of the cortico-striatal connections, demonstrating correlates of well-established goal-directed behaviour (involving medial orbitofrontal cortex - mOFC and VS), probabilistic reversal learning (lateral orbitofrontal cortex - lOFC and VS) and attentional shifting (dorsolateral prefrontal cortex - dlPFC and VS) while assessing habitual model-free (SMA and putamen) behaviours on an exploratory basis. We further use neurite orientation dispersion and density imaging (NODDI) to show that more goal-directed model-based learning (MBc) is also associated with higher mOFC neurite density and habitual model-free learning (MFc) implicates neurite complexity in the putamen. This data highlights similarities between a computational account of MFc and conventional measures of habit learning.     Review of Composite Marine Risers for Deep-Water Applications: Design, Development and Mechanics

    In recent times, the utilisation of marine composites in tubular structures has grown in popularity. These applications include composite risers and related SURF (subsea umbilicals, risers and flowlines) units. The composite industry has evolved in the development of advanced composites, such as thermoplastic composite pipes (TCP) and hybrid composite structures. However, there are gaps in the understanding of its performance in composite risers, hence the need for this review on the design, hydrodynamics and mechanics of composite risers. The review covers both the structure of the composite production riser (CPR) and its end-fittings for offshore marine applications. It also reviews the mechanical behaviour of composite risers, their microstructure and strength/stress profiles. In principle, designers now have a greater grasp of composite materials. It was concluded that composites differ from standard materials such as steel. Basically, composites have weight savings and a comparative stiffness-to-strength ratio, which are advantageous in marine composites. Also, the offshore sector has grown in response to newer innovations in composite structures such as composite risers, thereby providing new cost-effective techniques. This comprehensive review shows the necessity of optimising existing designs of composite risers. Conclusions drawn portray issues facing composite riser research. Recommendations were made to encourage composite riser developments, including elaboration of necessary standards and specifications

    The Automated Palomar 60-Inch Telescope

    We have converted the Palomar 60-inch telescope (P60) from a classical night assistant-operated telescope to a fully robotic facility. The automated system, which has been operational since September 2004, is designed for moderately fast (t <~ 3 minutes) and sustained (R <~ 23 mag) observations of gamma-ray burst afterglows and other transient events. Routine queue-scheduled observations can be interrupted in response to electronic notification of transient events. An automated pipeline reduces data in real-time, which is then stored on a searchable web-based archive for ease of distribution. We describe here the design requirements, hardware and software upgrades, and lessons learned from roboticization.     Childhood vision impairment and blindness in West Africa: public health measures and implications for systemic health

    Childhood blindness is an issue of global health impact, affecting approximately 2 million children worldwide. Vision 2020 and the United Nations Sustainable Development Goals previously identified childhood blindness as a key issue in the twentieth century, and while public health measures are underway, the precise etiologies and management require ongoing investigation and care, particularly within resource-limited settings such as sub-Saharan Africa. We systematically reviewed the literature on childhood blindness in West Africa to identify the anatomic classification and etiologies, particularly those causes of childhood blindness with systemic health implications. Treatable causes included cataract, refractive error, and corneal disease. Systemic etiologies identified included measles, rubella, vitamin A deficiency, and Ebola virus disease. While prior public health measures including vitamin A supplementation and vaccination programs have been deployed in most countries with reported data, multiple studies reported preventable or reversible etiologies of blindness and vision impairment. Ongoing research is necessary to standardize reporting for anatomies and/or etiologies of childhood blindness to determine the necessity of further development and implementation of public health measures that would ameliorate childhood blindness and vision impairment
