    Application of Microsatellite Markers for Genetic Diversity Analysis of Indonesian Local Cattle

    Animal genetic resources (AnGR), including cattle, have been valuable national assets that need to be preserved and developed. There are at least 16 recognized breeds of cattle that have been registered as local and new breeds by the Ministry of Agriculture of the Republic of Indonesia. Conservation and development programs of these local cattle breeds require basic information regarding their genetic diversity, relationships, and structures. There are several types of DNA markers that can be used for genetic diversity analysis, such as microsatellite markers. Microsatellites or short tandem repeats (STRs) are a group of DNA sequences consisting of tandemly repeated units (1–6 bp), which are abundant throughout the genome and can be found in both coding and non-coding regions. The primary advantages of microsatellites are that they are inherited in a Mendelian pattern (codominant markers), high polymorphism rates, and high abundances throughout the genome. The aim of this review is to discuss the application of microsatellite markers for genetic diversity analysis in Indonesian local cattle based on 3 indices: number alleles per locus, expected heterozygosity (He), and polymorphisms information content (PIC). There are at least 28 microsatellite markers that have been studied in Indonesian local cattle, with the number of alleles per locus ranging from 2 to 32, He values ranging from 0.100 to 0.985, and PIC values from 0.095 to 0.935. Based on the PIC values, several microsatellites are classified as highly informative, e.g. BM1824, ILST6, TGLA126, TGLA53, TGLA227, TGLA122, ETH225, INRA23, SPS113, SPS115, BM1818, CSSM66, ETH10, INRA005, INRA037, ETH185, HEL017, and ILSTS029. Therefore, these microsatellite markers can be potentially used for future genetic diversity analysis of other breeds of cattle

    Respon Tiga Varietas Kacang Hijau (Vigna radiata L.) terhadap Kapasitas Lapang

    Kacang hijau merupakan komoditas penting secara agronomi maupun pangan fungsional. Namun produksi kacang hijau di Indonesia masih tergolong rendah. Solusi dari permasalahan tersebut ialah peningkatan produksi kacang hijau dengan memanfaatkan lahan kering sebagai lahan budidaya. Pada lahan kering sering mencul permasalahan kurangnya ketersediaan air bagi tanaman. Oleh karena itu diperlukan penelitian ketersediaan air pada lahan kering dan pengujian beberapa varietas tahan kekeringan. Penelitian ini bertujuan untuk mengetahui varietas yang sesuai dibudidayakan di lahan kering. Penelitian dilakukan pada bulan Februari hingga April 2019 di lahan percobaan Joyo Agung, Lowokwaru, Malang. Penelitian ini menggunakan rancangan petak terbagi dengan menginteraksikan dua faktor yaitu varietas sebagai main treatment dan persentase kapasitas lapang sebagai sub treatment. Perlakuan varietas terdiri dari 3 taraf perlakuan yaitu : V1 : Varietas Vima I, V2 : Varietas Vima II dan V3 : Varietas Kutilang. Kemudian perlakuan persentase kapasitas lapang terdiri atas : K1 : 100% kapasitas lapang, K2 : 70% dari kapasitas lapang dan K3 : 40% dari kapasitas lapang. Pengambilan sempel menggunakan metode non-destruktif. Data yang diperoleh dianalisis menggunakan analisis ragam dengan taraf 5%. Hasil penelitian menunjukkan bahwa terjadi interaksi antar perlakuan terhadap bobot polong per tanaman. Perlakuan varietas berpengaruh terhadap umur berbunga dan bobot 100 biji dan perlakuan persentase kapasitas lapang berpengaruh terhadap tinggi tanaman, bobot polong, jumlah biji per polong dan bobot biji

    Tanaman Buncis (Phaseolus vulgaris L.) merupakan tanaman kacang-kacangan yang biasa dikonsumsi sebagai sayuran karena merupakan salah satu sumber protein nabati dan kaya vitamin. Salah satu  faktor yang menyebabkan produksi buncis yang semakin menurun adalahkualitas tanah yang menurun. Alternatif untuk meningkatkan kesuburan tanah dapat menggunakan pupuk organik yang dibantu dengan Effective Microorganisms 4 (EM4) yang merupakan kultur campuran mikroorganisme yang dapat mempercepat dekomposisi bahan organik sehingga dapat diserap langsung oleh perakaran tanaman. Tujuan dari penelitian ini adalah mempelajari pengaruh interaksi pemberian pupuk kandang sapi dan EM4 terhadap pertumbuhan dan hasil tanaman buncis.Penelitian dilaksanakan  di lahan pertanian desa Wringinsongo, kecamatan Tumpang, kabupaten Malang pada bulan Mei 2017 sampai Juli 2017. Penelitian ini dilakasanakan menggunakan Rancangan Acak Kelompok Faktorial dengan 2 faktor dan 3 ulangan.Faktor pertama adalah dosis pupuk kandang sapi yang terdiri dari 3 taraf yaitu dosis pupuk kandang sapi 15, 25, dan 35 ton ha-1. Faktor kedua adalah konsentrasi EM4 yang terdiri dari 3 taraf yaitu 5, 7,5, dan 10 ml L-1. Hasil penelitian menunjukkan tidak terdapat interaksi antara perlakuan dosis pupuk kandang sapi dengan konsentrasi EM4 pada semua parameter pertumbuhan dan hasil tanaman buncis. Pemberian pupuk kandang sapi 35 ton ha -1 berpengaruh nyata pada 24, 34, dan 44 hst pada parameter jumlah daun. Pemberian EM4 dengan Konsentrasi 10 ml L-1 berpengaruh nyata pada parameter jumlah daun 34 hst

In Indonesia, the maximum length of work time is 8 hours and the rest is for the rest of life in the family and community. Extending more work time than that will only reduce work efficiency, increase work fatigue, accidents and illness due to work. Fatigue is a complex physiological and psychological phenomenon which is characterized by symptoms of fatigue and physiological changes in the body. Fatigue will result in decreased workability and endurance.     Abstract This research was aimed at describing and analyzing the efforts devoted to escalating family prosperity through empowerment on Sido Makmur of woman community in Pasrujambe village. Research question under investigation was germane to how the efforts devoted to improving family prosperity on Sido Makmur woman community were carried out to improve the prosperity of families in Pasrujambe village. The presence of empowerment carried out in Sido Makmur community was one a kind of farming-based woman community which was intended to rally the appreciations of farming women. The research method operative in the study was descriptive, which was coupled with qualitative approach. The research site was Pasrujambe village, Pasrujambe sub-district of Lumajang district. Research informants were determined by purposive sampling, leading to including primary and secondary informants. To gain research data, observation, interview, and documentation were operationalized. Upon testing data validity, the researcher applied source trianglation. Research findings revealed that the empowerment to household women was done by enacting Sido Makmur woman community, through which various soft skill and  hard skills trainings were given. As a corollary, such undertakings exerted benefits to family prosperity in the form of economy improvement, knowledge improvement, and improvement.&nbsp

    Pengaruh Pemberian Dosis Pupuk Urea dan Variasi Jarak Tanam Terhadap Produksi Tanaman Ciplukan (Physalis angulata L.)

    This research was aimed to determine the cultivation techniques of ciplukan (Physalis angulata L.) by administering variations in the dose of urea fertilizer and spacing on the production of ciplukan plants cultivated in the field. This research was conducted from March 2020 until completion at the Jubung Agroteknopark, Jember city. This study used a factorial randomized block design with two treatment factors and three replications. The first treatment was by giving a dose of urea fertilizer, consisting of three levels N1: 1.5 g/plant, N2: 2.5 g/plant and N3: 3.5 g/plant. The second factor is setting spacing (J1) 20 x 40, (J2) 30 x40, (J3) 40 x 40, and (J4) 50x40. Each treatment was repeated 3 times. The data were analyzed using the ANOVA test and if there were differences in data then it was continued with the Duncan Multiple Range Test (DMRT) at a 95% confidence level. The results showed that the interaction was highly significant in the treatment of urea fertilizer 3.5 grams per plant with a spacing of 50 cm x 40 cm (N3J4) in the observation variable number of branches and number of harvestable fruit, and significantly different in the variable number of total fruit, number of fruit not harvestable, total fruit weight and average fruit weight. A single treatment of 3.5 gram urea per plant (N3) showed significant differences in the treatment of plant height, plant weight, and number of flowers. A single treatment with a spacing of 50 cm x 40 cm (J4) showed significantly different results on the variable number of leaves and very significantly different on the variable plant weight and number of flowers
