Pathologically Activated Neuroprotection via Uncompetitive Blockade of \u3cem\u3eN\u3c/em\u3e-Methyl-d-aspartate Receptors with Fast Off-rate by Novel Multifunctional Dimer Bis(propyl)-cognitin
Uncompetitive N-methyl-d-aspartate (NMDA) receptor antagonists with fast off-rate (UFO) may represent promising drug candidates for various neurodegenerative disorders. In this study, we report that bis(propyl)-cognitin, a novel dimeric acetylcholinesterase inhibitor and γ-aminobutyric acid subtype A receptor antagonist, is such an antagonist of NMDA receptors. In cultured rat hippocampal neurons, we demonstrated that bis(propyl)-cognitin voltage-dependently, selectively, and moderately inhibited NMDA-activated currents. The inhibitory effects of bis(propyl)-cognitin increased with the rise in NMDA and glycine concentrations. Kinetics analysis showed that the inhibition was of fast onset and offset with an off-rate time constant of 1.9 s. Molecular docking simulations showed moderate hydrophobic interaction between bis(propyl)-cognitin and the MK-801 binding region in the ion channel pore of the NMDA receptor. Bis(propyl)-cognitin was further found to compete with [3H]MK-801 with a Ki value of 0.27 μm, and the mutation of NR1(N616R) significantly reduced its inhibitory potency. Under glutamate-mediated pathological conditions, bis(propyl)-cognitin, in contrast to bis(heptyl)-cognitin, prevented excitotoxicity with increasing effectiveness against escalating levels of glutamate and much more effectively protected against middle cerebral artery occlusion-induced brain damage than did memantine. More interestingly, under NMDA receptor-mediated physiological conditions, bis(propyl)-cognitin enhanced long-term potentiation in hippocampal slices, whereas MK-801 reduced and memantine did not alter this process. These results suggest that bis(propyl)-cognitin is a UFO antagonist of NMDA receptors with moderate affinity, which may provide a pathologically activated therapy for various neurodegenerative disorders associated with NMDA receptor dysregulation

Chip Implementation with a Combined Wireless Temperature Sensor and Reference Devices Based on the DZTC Principle
This paper presents a novel CMOS wireless temperature sensor design in order to improve the sensitivity and linearity of our previous work on such devices. Based on the principle of CMOS double zero temperature coefficient (DZTC) points, a combined device is first created at the chip level with two voltage references, one current reference, and one temperature sensor. It was successfully fabricated using the 0.35 μm CMOS process. According to the chip results in a wide temperature range from −20 °C to 120 °C, two voltage references can provide temperature-stable outputs of 823 mV and 1,265 mV with maximum deviations of 0.2 mV and 8.9 mV, respectively. The result for the current reference gives a measurement of 23.5 μA, with a maximum deviation of 1.2 μA. The measurements also show that the wireless temperature sensor has good sensitivity of 9.55 mV/°C and high linearity of 97%. The proposed temperature sensor has 4.15-times better sensitivity than the previous design. Moreover, to facilitate temperature data collection, standard wireless data transmission is chosen; therefore, an 8-bit successive-approximation-register (SAR) analog-to-digital converter (ADC) and a 433 MHz wireless transmitter are also integrated in this chip. Synthesis and dissociation of soliton molecules in parallel optical-soliton reactors
Mode-locked lasers have been widely used to explore interactions between optical solitons, including bound-soliton states that may be regarded as “photonic molecules”. Conventional mode-locked lasers normally, however, host at most only a few solitons, which means that stochastic behaviours involving large numbers of solitons cannot easily be studied under controlled experimental conditions. Here we report the use of an optoacoustically mode-locked fibre laser to create hundreds of temporal traps or “reactors” in parallel, within each of which multiple solitons can be isolated and controlled both globally and individually using all-optical methods. We achieve on-demand synthesis and dissociation of soliton molecules within these reactors, in this way unfolding a novel panorama of diverse dynamics in which the statistics of multi-soliton interactions can be studied. The results are of crucial importance in understanding dynamical soliton interactions and may motivate potential applications for all-optical control of ultrafast light fields in optical resonators
Development of a P300 Brain–Machine Interface and Design of an Elastic Mechanism for a Rehabilitation Robot
This paper focuses on the development of a P300 speller and the design of a rehabilitation robot using a brain-machine interface. The combined feature set provides a norm that can be used to assess trends of the user’s increased or decreased independence. The combined feature set is found to maintain a 90% sorting rate; it can also reduce the relationship of individual independence for each subject. Among the results, the highest P300 classification accuracy can be increased by 36.04%. A novel adaptive coupled elastic actuator (ACEA) is proposed that uses adjustable characteristics to adapt to the applied output and input forces, thus ensuring safe human-machine interaction without the use of complex control strategies. The proposed robotic system uses variable impedance to achieve adaptability and safety in dynamic unstructured environments.
