    Genetic and phylogenetic analysis of ten Gobiidae species in China based on amplified fragment length polymorphism (AFLP) analysis

    To study the genetic and phylogenetic relationship of gobioid fishes in China, the representatives of 10 gobioid fishes from 2 subfamilies in China were examined by amplified fragment length polymorphism (AFLP) analysis. We established 220 AFLP bands for 45 individuals from the 10 species, and the percentage of polymorphic bands was 100%. The percentage of polymorphic loci within species ranged from 3.61 to 58.56%. Chaeturichthys stigmatias showed the greatest percentage of polymorphic loci (58.56%), the highest Nei’s genetic diversity (0.2421 ± 0.2190) and Shannon’s information index (0.3506 ± 0.3092), while Pterogobius zacalles showed the lowest percentage polymorphic loci (3.61%), the lowest Nei’s genetic diversity (0.0150 ± 0.0778) and lowest Shannon’s information index (0.0219 ± 0.1136). The topology of UPGMA tree showed that the individuals from the same species clustered together and the 10 species formed two major clades. One clade consisted Cryptocentrus filifer, P. zacalles, Tridentiger trigonocephalus, Chaeturichthys hexanema, C. stigmatias, Acanthogobius flavimanus and Synechogobius ommaturus, and the other clade consisted Odontamblyopus rubicundus, Trypauchen vagina and Ctenotrypauchen microcephalus. The results agreed with the traditional taxonomy of the morphological characters. AFLP fingerprints were successfully used to study the phylogenetic relationship of the gobioid fishes and it identified species origins of morphologically similar taxa.Key words: Phylogenetic, amplified fragment length polymorphism (AFLP), gobiidae, Amblyopinae, gobiinae

    Cloning and characterization of a nitrite reductase gene related to somatic embryogenesis in Gossypium hirsutum

    A nitrite reductase gene related to somatic embryogenesis was first cloned from Gossypium hirsutum. The cDNA sequence of the gene, named GhNiR, is 2,257 bp in length, with 254 bp of the 5’ untranslated region and 236 bp of the 3’ untranslated region. The open reading frame is 1,767 bp in length, encoding a deduced amino acid sequence of 588 residues with a molecular weight of 65.722 kDa and an isoelectric point of 7.07. Semi-quantitative RT-PCR analysis showed that the expression level of GhNiR was higher in embryogenic calli and somatic embryoids than in nonembryogenic calli among different somatic embryogenesis stages, and that the level of GhNiR mRNA was also higher in the cultivar with higher somatic embryogenesis ability. The catalytic GhNiR was verified by transformation in E. coli BL21 (DE3) strain with the recombinant expression vector pET-28A-GhNiR.     Central inflammation and leptin resistance are attenuated by ginsenoside Rb1 treatment in obese mice fed a high-fat diet

    A low-grade pro-inflammatory state is at the pathogenic core of obesity and type 2 diabetes. We tested the hypothesis that the plant terpenoid compound ginsenoside Rb1 (Rb1), known to exert anti-inflammatory effects, would ameliorate obesity, obesity-associated inflammation and glucose intolerance in the high-fat diet-induced obese mouse model. Furthermore, we examined the effect of Rb1 treatment on central leptin sensitivity and the leptin signaling pathway in the hypothalamus. We found that intraperitoneal injections of Rb1 (14 mg/kg, daily) for 21 days significantly reduced body weight gain, fat mass accumulation, and improved glucose tolerance in obese mice on a HF diet compared to vehicle treatment. Importantly, Rb1 treatment also reduced levels of pro-inflammatory cytokines (TNF-α, IL-6 and/or IL-1β) and NF-κB pathway molecules (p-IKK and p-IκBα) in adipose tissue and liver. In the hypothalamus, Rb1 treatment decreased the expression of inflammatory markers (IL-6, IL-1β and p-IKK) and negative regulators of leptin signaling (SOCS3 and PTP1B). Furthermore, Rb1 treatment also restored the anorexic effect of leptin in high-fat fed mice as well as leptin pSTAT3 signaling in the hypothalamus. Ginsenoside Rb1 has potential for use as an anti-obesity therapeutic agent that modulates obesity-induced inflammation and improves central leptin sensitivity in HF diet-induced obesity. © 2014 Public Library of ScienceAcquired from the Public Library of Science.     Interface Coupling in Twisted Multilayer Graphene by Resonant Raman Spectroscopy of Layer Breathing Modes.

    Raman spectroscopy is the prime nondestructive characterization tool for graphene and related layered materials. The shear (C) and layer breathing modes (LBMs) are due to relative motions of the planes, either perpendicular or parallel to their normal. This allows one to directly probe the interlayer interactions in multilayer samples. Graphene and other two-dimensional (2d) crystals can be combined to form various hybrids and heterostructures, creating materials on demand with properties determined by the interlayer interaction. This is the case even for a single material, where multilayer stacks with different relative orientations have different optical and electronic properties. In twisted multilayer graphene there is a significant enhancement of the C modes due to resonance with new optically allowed electronic transitions, determined by the relative orientation of the layers. Here we show that this applies also to the LBMs, which can be now directly measured at room temperature. We find that twisting has a small effect on LBMs, quite different from the case of the C modes. This implies that the periodicity mismatch between two twisted layers mostly affects shear interactions. Our work shows that ultralow-frequency Raman spectroscopy is an ideal tool to uncover the interface coupling of 2d hybrids and heterostructures

    Learning Graph-Convolutional Representations for Point Cloud Denoising

    Point clouds are an increasingly relevant data type but they are often corrupted by noise. We propose a deep neural network based on graph-convolutional layers that can elegantly deal with the permutation-invariance problem encountered by learning-based point cloud processing methods. The network is fully-convolutional and can build complex hierarchies of features by dynamically constructing neighborhood graphs from similarity among the high-dimensional feature representations of the points. When coupled with a loss promoting proximity to the ideal surface, the proposed approach significantly outperforms state-of-the-art methods on a variety of metrics. In particular, it is able to improve in terms of Chamfer measure and of quality of the surface normals that can be estimated from the denoised data. We also show that it is especially robust both at high noise levels and in presence of structured noise such as the one encountered in real LiDAR scans.Comment: European Conference on Computer Vision (ECCV) 202

    Screening for inborn errors of metabolism in high-risk children: a 3-year pilot study in Zhejiang Province, China

    <p>Abstract</p> <p>Background</p> <p>Tandem mass spectrometry (MS/MS) has been available in China for 8 years. This technique makes it possible to screen for a wide range of previously unscreened inborn errors of metabolism (IEM) using a single test. This 3-year pilot study investigated the screening, diagnosis, treatment and outcomes of IEM in symptomatic infants and children.</p> <p>Methods</p> <p>All children encountered in the Newborn Screening Center of Zhejiang Province during a 3-year period with symptoms suspicious for IEM were screened for metabolic diseases. Dried blood spots were collected and analyzed by tandem mass spectrometry. The diagnoses were further confirmed by clinical symptoms and biochemical analysis. Neonatal intrahepatic cholestasis caused by citrin deficiency, ornithine transcarbamylase deficiency and primary carnitine deficiency were confirmed by DNA analysis.</p> <p>Results</p> <p>A total of 11,060 symptomatic patients (6,720 boys, 4,340 girls) with a median age of 28.8 months (range: 0.04-168.2 months) were screened. Among these, 62 were diagnosed with IEM, with a detection rate of 0.56%. Thirty-five were males and 27 females and the median age was 3.55 months (range 0.07-143.9 months). Of the 62 patients, 27 (43.5%) had aminoacidemias, 26 (41.9%) had organic acidemias and nine (14.5%) had fatty acid oxidation disorders.</p> <p>Conclusions</p> <p>Because most symptomatic patients are diagnosed at an older age, mental retardation and motor delay are difficult to reverse. Additionally, poor medication compliance reduces the efficacy of treatment. More extensive newborn screening is thus imperative for ensuring early diagnosis and enhancing the treatment efficacy of IEM.</p
