Elderly patients primarily present with nonclassic symptoms of PHPT that can sometimes be missed in favor of other diagnoses. Several recent studies demonstrate the safety and efficacy of outpatient, minimally invasive parathyroidectomy in elderly patients with PHPT, the majority of them reporting increased fracture-free survival and symptomatic relief postoperatively and thus experiencing improved quality of life

    Experimental measurement of stress at a four-domain junction in lead zirconate titanate

    A junction between two lamellar bands of ferroelectric domains in a lead zirconate titanate (PZT) ceramic is analysed using Kikuchi diffraction patterns in the transmission electron microscope. Indexing of the diffraction patterns allowed the determination of the 3D relative orientation of the 4 different domains at the junction and thus the characterisation of the domain boundaries. The local c/a ratio could also be determined from the misorientations at the domain boundaries. Analysis of the data showed that large stresses were concentrated at the junction, and that this is inevitable at such band junctions. Such stress concentrations could act as nuclei for cracking of the ceramic under additional loading in service, perhaps particularly as a consequence of extended electromechanical cycling. Moreover, the stresses would increase with increasing c/a making the issues all the more serious for Ti-rich compositions having larger c/a ratios

    BackgroundPatients with systemic lupus erythematosus (SLE) are at increased risk of atherosclerosis, even after accounting for traditional risk factors. High levels of leptin and low levels of adiponectin are associated with both atherosclerosis and immunomodulatory functions in the general population.ObjectiveTo examine the association between these adipokines and subclinical atherosclerosis in SLE, and also with other known inflammatory biomarkers of atherosclerosis.MethodsCarotid ultrasonography was performed in 250 women with SLE and 122 controls. Plasma leptin and adiponectin levels were measured. Lipoprotein a (Lp(a)), oxidised phospholipids on apoB100 (OxPL/apoB100), paraoxonase, apoA-1 and inflammatory high-density lipoprotein (HDL) function were also assessed.ResultsLeptin levels were significantly higher in patients with SLE than in controls (23.7±28.0 vs 13.3±12.9 ng/ml, p<0.001). Leptin was also higher in the 43 patients with SLE with plaque than without plaque (36.4±32.3 vs 20.9±26.4 ng/ml, p=0.002). After multivariate analysis, the only significant factors associated with plaque in SLE were leptin levels in the highest quartile (≥29.5 ng/ml) (OR=2.8, p=0.03), proinflammatory HDL (piHDL) (OR=12.8, p<0.001), age (OR=1.1, p<0.001), tobacco use (OR=7.7, p=0.03) and hypertension (OR=3.0, p=0.01). Adiponectin levels were not significantly associated with plaque in our cohort. A significant correlation between leptin and piHDL function (p<0.001), Lp(a) (p=0.01) and OxPL/apoB100 (p=0.02) was also present.ConclusionsHigh leptin levels greatly increase the risk of subclinical atherosclerosis in SLE, and are also associated with an increase in inflammatory biomarkers of atherosclerosis such as piHDL, Lp(a) and OxPL/apoB100. High leptin levels may help to identify patients with SLE at risk of atherosclerosis

    Deriving instrumental point spread functions from partially occulted images

    The point spread function (PSF) of an imaging system describes the response of the system to a point source. Accurately determining the PSF enables one to correct for the combined effects of focusing and scattering within the imaging system and, thereby, enhance the spatial resolution and dynamic contrast of the resulting images. We present a semi-empirical semi-blind methodology to derive a PSF from partially occulted images. We partition the two-dimensional PSF into multiple segments, set up a multilinear system of equations, and directly fit the system of equations to determine the PSF weight in each segment. The algorithm is guaranteed to converge toward the correct instrumental PSF for a large class of occultations, does not require a predefined functional form of the PSF, and can be applied to a large variety of partially occulted images, such as within laboratory settings, regular calibrations within a production line or in the field, astronomical images of distant clusters of stars, or partial solar eclipse images. We show that the central weight of the PSF, which gives the percentage of photons that are not scattered by the instrument, is accurate to better than 1.2%. The mean absolute percentage error between the reconstructed and true PSF is usually between 0.5 and 5% for the entire PSF, between 0.5 and 5% for the PSF core, and between 0.5 and 3% for the PSF tail

    Inferring Phytoplankton, Terrestrial Plant and Bacteria Bulk delta C-13 Values from Compound Specific Analyses of Lipids and Fatty Acids

    Stable isotope mixing models in aquatic ecology require delta C-13 values for food web end members such as phytoplankton and bacteria, however it is rarely possible to measure these directly. Hence there is a critical need for improved methods for estimating the delta C-13 ratios of phytoplankton, bacteria and terrestrial detritus from within mixed seston. We determined the delta C-13 values of lipids, phospholipids and biomarker fatty acids and used these to calculate isotopic differences compared to the whole-cell delta C-13 values for eight phytoplankton classes, five bacterial taxa, and three types of terrestrial organic matter (two trees and one grass). The lipid content was higher amongst the phytoplankton (9.5 +/- 4.0%) than bacteria (7.3 +/- 0.8%) or terrestrial matter (3.9 +/- 1.7%). Our measurements revealed that the delta C-13 values of lipids followed phylogenetic classification among phytoplankton (78.2% of variance was explained by class), bacteria and terrestrial matter, and there was a strong correlation between the delta C-13 values of total lipids, phospholipids and individual fatty acids. Amongst the phytoplankton, the isotopic difference between biomarker fatty acids and bulk biomass averaged -10.7 +/- 1.1 parts per thousand for Chlorophyceae and Cyanophyceae, and -6.1 +/- 1.7 parts per thousand for Cryptophyceae, Chrysophyceae and Diatomophyceae. For heterotrophic bacteria and for type I and type II methane-oxidizing bacteria our results showed a -1.3 +/- 1.3 parts per thousand, -8.0 +/- 4.4 parts per thousand, and -3.4 +/- 1.4 parts per thousand delta C-13 difference, respectively, between biomarker fatty acids and bulk biomass. For terrestrial matter the isotopic difference averaged -6.6 +/- 1.2 parts per thousand. Based on these results, the delta C-13 values of total lipids and biomarker fatty acids can be used to determine the delta C-13 values of bulk phytoplankton, bacteria or terrestrial matter with +/- 1.4 parts per thousand uncertainty (i.e., the pooled SD of the isotopic difference for all samples). We conclude that when compound-specific stable isotope analyses become more widely available, the determination of delta C-13 values for selected biomarker fatty acids coupled with established isotopic differences, offers a promising way to determine taxa-specific bulk delta C-13 values for the phytoplankton, bacteria, and terrestrial detritus embedded within mixed seston.Peer reviewe

    A cell wall reference profile for Miscanthus bioenergy crops highlights compositional and structural variations associated with development and organ origin

    Miscanthus spp. are promising lignocellulosic energy crops, but cell wall recalcitrance to deconstruction still hinders their widespread use as bioenergy and biomaterial feedstocks. Identification of cell wall characteristics desirable for biorefining applications is crucial for lignocellulosic biomass improvement. However, the task of scoring biomass quality is often complicated by the lack of a reference for a given feedstock. A multidimensional cell wall analysis was performed to generate a reference profile for leaf and stem biomass from several miscanthus genotypes harvested at three developmentally distinct time points. A comprehensive suite of 155 monoclonal antibodies was used to monitor changes in distribution, structure and extractability of noncellulosic cell wall matrix glycans. Glycan microarrays complemented with immunohistochemistry elucidated the nature of compositional variation, and in situ distribution of carbohydrate epitopes. Key observations demonstrated that there are crucial differences in miscanthus cell wall glycomes, which may impact biomass amenability to deconstruction. For the first time, variations in miscanthus cell wall glycan components were comprehensively characterized across different harvests, organs and genotypes, to generate a representative reference profile for miscanthus cell wall biomass. Ultimately, this portrait of the miscanthus cell wall will help to steer breeding and genetic engineering strategies for the development of superior energy crops
