    5-methylcytosine DNA methylation regulates gene expression and developmental programming in a broad range of eukaryotes. However, its presence and potential roles in ciliates, complex single-celled eukaryotes with germline-somatic genome specialization via nuclear dimorphism, are largely uncharted. While canonical cytosine methyltransferases have not been discovered in published ciliate genomes, recent studies performed in the stichotrichous ciliate Oxytricha trifallax suggest de novo cytosine methylation during macronuclear development. In this study, we applied bisulfite genome sequencing, DNA mass spectrometry and antibody-based fluorescence detection to investigate the presence of DNA methylation in Paramecium tetraurelia. While the antibody-based methods suggest cytosine methylation, DNA mass spectrometry and bisulfite sequencing reveal that levels are actually below the limit of detection. Our results suggest that Paramecium does not utilize 5-methylcytosine DNA methylation as an integral part of its epigenetic arsenal

    OMI total bromine monoxide (OMBRO) data product: algorithm, retrieval and measurement comparisons

    This paper presents the retrieval algorithm for the operational Ozone Monitoring Instrument (OMI) total bromine monoxide (BrO) data product (OMBRO) developed at the Smithsonian Astrophysical Observatory (SAO) and shows comparisons with correlative measurements and retrieval results. The algorithm is based on direct nonlinear least squares fitting of radiances from the spectral range 319.0–347.5&thinsp;nm. Radiances are modeled from the solar irradiance, attenuated by contributions from BrO and interfering gases, and including rotational Raman scattering, additive and multiplicative closure polynomials, correction for Nyquist undersampling and the average fitting residual spectrum. The retrieval uses albedo- and wavelength-dependent air mass factors (AMFs), which have been pre-computed using a single mostly stratospheric BrO profile. The BrO cross sections are multiplied by the wavelength-dependent AMFs before fitting so that the vertical column densities (VCDs) are retrieved directly. The fitting uncertainties of BrO VCDs typically vary between 4 and 7×1012&thinsp;molecules cm−2 (∼10&thinsp;%–20&thinsp;% of the measured BrO VCDs). Additional fitting uncertainties can be caused by the interferences from O2-O2 and H2CO and their correlation with BrO. AMF uncertainties are estimated to be around 10&thinsp;% when the single stratospheric-only BrO profile is used. However, under conditions of high tropospheric concentrations, AMF errors due to this assumption of profile can be as high as 50&thinsp;%. The retrievals agree well with GOME-2 observations at simultaneous nadir overpasses and with ground-based zenith-sky measurements at Harestua, Norway, with mean biases less than -0.22±1.13×1013 and 0.12±0.76×1013&thinsp;molecules cm−2, respectively. Global distribution and seasonal variation of OMI BrO are generally consistent with previous satellite observations. Finally, we confirm the capacity of OMBRO retrievals to observe enhancements of BrO over the US Great Salt Lake despite the current retrieval setup considering a stratospheric profile in the AMF calculations. OMBRO retrievals also show significant BrO enhancements from the eruption of the Eyjafjallajökull volcano, although the BrO retrievals are affected under high SO2 loading conditions by the sub-optimum choice of SO2 cross sections.</p

    Six Years of Chandra Observations of Supernova Remnants

    We present a review of the first six years of Chandra X-ray Observatory observations of supernova remnants. From the official "first-light" observation of Cassiopeia A that revealed for the first time the compact remnant of the explosion, to the recent million-second spectrally-resolved observation that revealed new details of the stellar composition and dynamics of the original explosion, Chandra observations have provided new insights into the supernova phenomenon.     In the 1990s policy relating to children and young people who offend developed as a result of the interplay of political imperatives and populist demands. The ‘responsibilisation’ of young offenders and the ‘no excuses’ culture of youth justice have been ‘marketed’ through a discourse which evidences linguistic changes. This article focuses on one particular area of policy change, that relating to the prosecutorial decision, to show how particular images of children were both reflected and constructed through a changing selection of words to describe the non-adult suspect and offender. In such minutiae of discourse can be found not only the signifiers of public attitudinal and policy change but also the means by which undesirable policy developments can be challenged

    Design and Initial Performance of the Prototype for the BEACON Instrument for Detection of Ultrahigh Energy Particles

    The Beamforming Elevated Array for COsmic Neutrinos (BEACON) is a planned neutrino telescope designed to detect radio emission from upgoing air showers generated by ultrahigh energy tau neutrino interactions in the Earth. This detection mechanism provides a measurement of the tau flux of cosmic neutrinos. We have installed an 8-channel prototype instrument at high elevation at Barcroft Field Station, which has been running since 2018, and consists of 4 dual-polarized antennas sensitive between 30-80 MHz, whose signals are filtered, amplified, digitized, and saved to disk using a custom data acquisition system (DAQ). The BEACON prototype is at high elevation to maximize effective volume and uses a directional beamforming trigger to improve rejection of anthropogenic background noise at the trigger level. Here we discuss the design, construction, and calibration of the BEACON prototype instrument. We also discuss the radio frequency environment observed by the instrument, and categorize the types of events seen by the instrument, including a likely cosmic ray candidate event.Comment: 21 pages, 20 figure

    Mammographic screening for young women with a family history of breast cancer: knowledge and views of those at risk

    Although the effectiveness of mammography for women under the age of 50 years with a family history of breast cancer (FHBC) has not yet been proven, annual screening is being offered to these women to manage breast cancer risk. This study investigates women's awareness and interpretation of their familial risk and knowledge and views about mammographic screening. A total of 2231 women from 21 familial/breast/genetics centres who were assessed as moderate risk (17–30% lifetime risk) or high risk (>30% lifetime risk) completed a questionnaire before their mammographic screening appointment. Most women (70%) believed they were likely, very likely or definitely going to develop breast cancer in their lifetime. Almost all women (97%) understood that the purpose of mammographic screening was to allow the early detection of breast cancer. However, 20% believed that a normal mammogram result meant there was definitely no breast cancer present, and only 4% understood that screening has not been proven to save lives in women under the age of 50 years. Women held positive views on mammography but did not appear to be well informed about the potential disadvantages. These findings suggest that further attention should be paid to improving information provision to women with an FHBC being offered routine screening
