    Comparative morphometric analysis of bloodstream and lymph forms of Trypanosoma (T.) brucei brucei grown in vitro and in vivo

    The fine structure of bloodstream forms of Trypanosoma brucei cultivated in vitro, and of trypanosomes from lymph and blood of mammalian hosts, was compared morphometrically. The cell volume, quantitative parameters of the mitochondrion and of glycosomes were mainly investigated. A Coulter Channelyzer was used for the first time to measure the mean cell volume of living parasites. In vitro, the monomorphic trypanosomes between the feeder layer cells showed lower values for mitochondrial parameters than the slightly pleomorphic forms from the supernatant medium. Trypanosomes in culture were very similar morphologically to forms from lymph nodes of rats. Despite some morphometric differences between cultivated blood stream forms and those grown in vivo, the similarity of both populations was clear. Both populations, however, differed significantly from stages found in the vector or from procyclic culture form

    Background: Idiopathic pulmonary fibrosis (IPF) is a fatal interstitial lung disease characterised by accumulation of activated (myo)fibroblasts and excessive extracellular matrix deposition. The enhanced accumulation of (myo)fibroblasts may be attributed, in part, to the process of transforming growth factor \textgreekb1 (TGF\textgreekb1)-induced epithelial--mesenchymal transition (EMT), the phenotypic switching of epithelial to fibroblast-like cells. Although alveolar epithelial type II (ATII) cells have been shown to undergo EMT, the precise mediators and mechanisms remain to be resolved. The objective of this study is to investigate the role of SNAI transcription factors in the process of EMT and in IPF.Methods: Using quantitative reverse transcription-PCR (RT-PCR), immunofluorescence, immunohistochemistry, western blotting, as well as gain- and loss-of-function studies and functional assays, the role of SNAI1 and SNAI2 in TGF\textgreekb1-induced EMT in ATII cells in vitro was assessed; and the expression of SNAI transcription factors was analysed in experimental and human IPF in vivo.Results: TGF\textgreekb1 treatment increased the expression and nuclear accumulation of SNAI1 and SNAI2, in concert with induction of EMT in ATII cells. SNAI overexpression was sufficient to induce EMT, and small interfering RNA (siRNA)-mediated SNAI depletion attenuated TGF\textgreekb1-induced ATII cell migration and EMT. SNAI expression was elevated in experimental and human IPF and localised to hyperplastic ATII cells in vivo.Conclusions: The results demonstrate that TGF\textgreekb1-induced EMT in ATII cells is essentially controlled by the expression and nuclear translocation of SNAI transcription factors. Increased SNAI1 and SNAI2 expression in experimental and human IPF in vivo suggests that SNAI-mediated EMT may contribute to the fibroblast pool in idiopathic pulmonary fibrosis

    Hyperfine, rotational and Zeeman structure of the lowest vibrational levels of the 87^{87}Rb2_2 \tripletex state

    We present the results of an experimental and theoretical study of the electronically excited \tripletex state of 87^{87}Rb2_2 molecules. The vibrational energies are measured for deeply bound states from the bottom up to v′=15v'=15 using laser spectroscopy of ultracold Rb2_2 Feshbach molecules. The spectrum of each vibrational state is dominated by a 47\,GHz splitting into a \cog and \clg component caused mainly by a strong second order spin-orbit interaction. Our spectroscopy fully resolves the rotational, hyperfine, and Zeeman structure of the spectrum. We are able to describe to first order this structure using a simplified effective Hamiltonian.Comment: 10 pages, 7 figures, 2 table

    Characterization of Trypanosoma brucei isolated from lymph nodes of rats

    Forms of Trypanosoma brucei isolated from lymph nodes of cyclically or intravenously infected rats, at different stages of infection, were morphologically compared with the corresponding bloodstream forms. Unlike the pleomorphic bloodstream trypanosome population, the lymph node trypanosome population was essentially monomorphic and resembled the slender forms of the bloodstream. Values for the morphometric parameters indicated a position between the slender and the stumpy forms. The number of trypanosomes/ml of blood showed the well-known periodic fluctuations while the number of trypanosomes/g of lymph node remained fairly constant. Serology revealed that lymph node trypanosome populations differed antigenically from the trypanosome populations isolated simultaneously from the bloodstrea

    The effects of donepezil in Alzheimer's disease - Results from a multinational trial

    Donepezil has been shown to be well tolerated and to improve cognition and global function in patients with mild to moderately severe Alzheimer's disease (AD). The current trial was undertaken to investigate further the efficacy and safety of donepezil, in a multinational setting, in patients with mild to moderately severe AD. This 30-week, placebo-controlled, parallel-group study consisted of a 24-week, double-blind treatment phase followed by a 6-week, single-blind, placebo washout. Eight hundred and eighteen patients with mild to moderately severe AD were randomly allocated to treatment with single, daily doses of 5 or 10 mg donepezil, or placebo. The two primary efficacy measures were: a cognitive performance test, the Alzheimer's Disease Assessment Scale-cognitive subscale (ADAS-cog) and a global evaluation, the Clinician's Interview-Based Impression of Change with caregiver input (CIBIC plus). Secondary outcome measures included the Sum of the Boxes of the Clinical Dementia Rating Scale (CDR-SB), a modified Interview for Deterioration in Daily living activities in Dementia (IDDD) and a patient-rated quality of life assessment. Statistically significant improvements in cognitive and global function were observed, as evaluated by ADAS-cog and CIBIC plus, respectively, in both the 5 and 10 mg/day donepezil groups, compared with placebo. Treatment-associated changes were also observed in functional skills, as shown by improved scores on the CDR-SB and the complex-tasks component of the IDDD. A dose-response effect was evident, with the 10 mg/day donepezil group demonstrating greater benefits in all outcome measures than the 5 mg/day group. Donepezil was well tolerated by this patient population and did not produce any clinically significant laboratory test abnormalities. The results of this study confirm that donepezil is effective and well tolerated in treating the symptoms of mild to moderately severe AD

    Repulsively bound atom pairs: Overview, Simulations and Links

    We review the basic physics of repulsively bound atom pairs in an optical lattice, which were recently observed in the laboratory, including the theory and the experimental implementation. We also briefly discuss related many-body numerical simulations, in which time-dependent Density Matrix Renormalisation Group (DMRG) methods are used to model the many-body physics of a collection of interacting pairs, and give a comparison of the single-particle quasimomentum distribution measured in the experiment and results from these simulations.     Atom-molecule dark states in a Bose-Einstein condensate

    We have created a dark quantum superposition state of a Rb Bose-Einstein condensate (BEC) and a degenerate gas of Rb2_2 ground state molecules in a specific ro-vibrational state using two-color photoassociation. As a signature for the decoupling of this coherent atom-molecule gas from the light field we observe a striking suppression of photoassociation loss. In our experiment the maximal molecule population in the dark state is limited to about 100 Rb2_2 molecules due to laser induced decay. The experimental findings can be well described by a simple three mode model.Comment: 4 pages, 6 figure

    Sequence differences between histones of procyclic Trypanosoma brucei brucei and higher eukaryotes

    Four histones, a, b, c, d from procyclic Trypanosoma brucei brucei, which show similarities with the amino acid composition of the core histones H3, H2A, H2B and H4, were isolated and cleaved with Endoproteinase Glu-C. The fragments were separated by FPLC reversed phase chromatography and a subset of the fragments (a5, a9, b6, c8, d3, d9, d11) was subjected to sequence analysis. A 54-71% identity was found in the sequences of the fragment c8 and the C-terminal half of H2B and of three fragments of protein d covering the N-terminal half as well as the C-terminal region of H4. The amino acid sequence of the fragment a9 showed a 57 and 54% identity with H3 sequences of Saccharomyces cerevisiae and Xenopus laevis. Neither the a5 nor the b6 sequence could be aligned with histone sequences of other eukaryotes. The significant differences of 21-48% between the T. b. brucei, histone sequences and those of calf thymus histones, which are more pronounced than the differences of Tetrahymena pyriformis and the higher eukaryote, resulted partially from replacements of amino acids with different properties and indicate specific patterns of histone-histone and/or histone-DNA contact sites in the nucleosome of T. b. brucei. These differences, together with the lack of a functional histone H1, may be sufficient to explain the lack of a salt-dependent formation of the nucleosome filament into the 30 nm fibre, which reflects alternative methods of organizing and processing the genetic information in the nucleus of the protozoan parasite and which may be of chemotherapeutic significanc
