    Gene expression profiling of rat livers with Yin-deficiency-heat syndrome

    AbstractObjectiveTo explore the nature of ”Yin internal heat caused by Yin-deficiency,” in terms of the theory of Traditional Chinese Medicine, by studying energy metabolism in rats with Yin-deficiency-heat syndrome and analyzing the gene expression profile of their livers.MethodsA Yin-deficiency-heat syndrome model was induced in rats using three Chinese medicinal herbs. Glycogen and triglycerides in blood plasma, and the enzyme activity of ATP in livers were measured colorimetrically. Triiodothyronine (T3), thyroxine (T4), and thyroid stimulating hormone levels in blood plasma were also measured with enzyme linked immunosorbent assay. The gene expression profile of livers was detected with gene chip analysis. Differentially expressed genes were screened out and classified according to Gene Ontology. The accuracy of results were examined with reverse transcription-polymerase chain reaction.ResultsCompared with the control group, body weight (P<0.05) and hepatic glycogen (P<0.05) were significantly lower in the Yin-deficiency-heat syndrome group. Moreover, toe temperature (P< 0.01) and triglyceride (P<0.05), Na+−K+−ATPase (P< 0.01), Mg2+−ATPase (P<0.01), T3 (P<0.05), and T4 (P< 0.01) levels were significantly higher. There were 99 differentially expressed genes in livers from the Yin-deficiency-heat syndrome group. Genes were mainly related to sterol synthesis (Pc=0.0392), defense response (Pc=0.0448), and sterol metabolism (Pc=0.0533).ConclusionAbnormal expression genes in rats with Yin-deficiency-heat syndrome prompted the synthesis and metabolism of cholesterol, increased energy consumption, and reduced defense response. This gene expression might be the molecular mechanism underlying “internal heat caused by Yin-deficiency” in the rats with Yin-deficiency-heat syndrome

    RESEARCH ON THE MOTION RESPONSE OF AQUACULTURE SHIP AND TANK SLOSHING UNDER ROLLING RESONANCE

    The double-row and double-chamfered aquaculture tank is a special tank structure of the aquaculture ship. The tank sloshing of this structure is coupled with the hull motion, which has an important impact on the safety of the hull motion. In the present study, research on the tank sloshing and hull motion response of aquaculture ships was conducted based on the model seakeeping and tank sloshing tests in regular waves. The test results were compared with the numerical simulation results of solid loading without sloshing. The results showed that the numerical simulation of the pitch motion was consistent with the amplitude-frequency response curve of the experimental results. Under certain transverse wave conditions, a large discrepancy existed between the amplitude-frequency response curve of the heave motion by the numerical simulation and the test results, and the roll motion differed most from the experimental result. Severe roll resonance occurred when the wave length-ship length ratio was 0.6. The roll motion amplitude was increased by 183.2%. Therefore, compared with aquaculture ships without sloshing, the sloshing of the tank has little effect on the pitch but has a great impact on the roll and heave motions, with the most significant effect on the roll motion

    Exploring Chinese herbal medicine for ischemic stroke: insights into microglia and signaling pathways

    Ischemic stroke is a prevalent clinical condition affecting the central nervous system, characterized by a high mortality and disability rate. Its incidence is progressively rising, particularly among younger individuals, posing a significant threat to human well-being. The activation and polarization of microglia, leading to pro-inflammatory and anti-inflammatory responses, are widely recognized as pivotal factors in the pathogenesis of cerebral ischemia and reperfusion injury. Traditional Chinese herbal medicines (TCHMs) boasts a rich historical background, notable efficacy, and minimal adverse effects. It exerts its effects by modulating microglia activation and polarization, suppressing inflammatory responses, and ameliorating nerve injury through the mediation of microglia and various associated pathways (such as NF-κB signaling pathway, Toll-like signaling pathway, Notch signaling pathway, AMPK signaling pathway, MAPK signaling pathway, among others). Consequently, this article focuses on microglia as a therapeutic target, reviewing relevant pathway of literature on TCHMs to mitigate neuroinflammation and mediate IS injury, while also exploring research on drug delivery of TCHMs. The ultimate goal is to provide new insights that can contribute to the clinical management of IS using TCHMs

    Effects of High-Stakes Accountability Testing on Teacher Motivation

    There is always a driving force behind our actions in any given situation. In 2000, NCLB instituted accountability testing with strict requirements for all schools in the U.S., hoping to motivate teachers to reach higher levels of instruction by using sanctions or rewards against teachers when standards were not met. However, Deci and Ryan¿s (2000) SDT explains that fostering intrinsic motivation and providing autonomy support for an individual results in more effective outcomes and increased satisfaction. Private and public school teachers in PA were surveyed using the Work Climate Questionnaire and the Work Task Motivation Scale for Teachers to examine differences in self-reported motivation and perceived autonomy support in order to investigate if the use of high-stakes accountability testing (HST) is related to decreased teacher motivation. The results from ANOVAs and correlations indicate differences in self-reported motivation levels of teachers in public and private schools, as well as aid ratio, grade taught, number of years in teaching, and the time the participant responded (before or after the PSSA), as well as perceived autonomy support, suggesting that the presence of HST may influence motivation in teachers

    Orf virus DNA vaccines expressing ORFV 011 and ORFV 059 chimeric protein enhances immunogenicity

    Background: ORFV attenuated live vaccines have been the main prophylactic measure against contagious ecthyma in sheep and goats in the last decades, which play an important role in preventing the outbreak of the disease. However, the available vaccines do not induce lasting immunity in sheep and goats. On the other hand, variation in the terminal genome of Orf virus vaccine strains during cell culture adaptation may affect the efficacy of a vaccine. Currently, there are no more effective antiviral treatments available for contagious ecthyma. Results: We constructed three eukaryotic expression vectors pcDNA3.1-ORFV011, pcDNA3.1-ORFV059 and pcDNA3.1-ORFV011/ORFV059 and tested their immunogenicity in mouse model. High level expression of the recombinant proteins ORFV011, ORFV059 and ORFV011/ORFV059 was confirmed by western blotting analysis and indirect fluorescence antibody (IFA) tests. The ORFV-specific antibody titers and serum IgG1/IgG2a titers, the proliferation of lymphocytes and ORFV-specific cytokines (IL-2, IL-4, IL-6, IFN-gamma, and TNF-alpha) were examined to evaluate the immune responses of the vaccinated mice. We found that mice inoculated with pcDNA3.1-ORFV 011/ORFV059 had significantly stronger immunological responses than those inoculated with pcDNA3.1-ORFV011, pcDNA3.1-ORFV059, or pcDNA3.1-ORFV011 plus pcDNA3.1-ORFV059. Compared to other vaccine plasmids immunized groups, pcDNA3.1-ORFV011/ORFV059 immunized group enhances immunogenicity.     NEFA-Sensitive Orai1 Expression in Regulation of De Novo Lipogenesis

    Background/Aims: Non-esterified fatty acids (NEFAs) are important inducers of inflammatory responses and hepatic lipid accumulation, which lead to non-alcoholic fatty liver disease (NAFLD). High plasma NEFA is found in NAFLD patients, and associated with metabolic syndrome and type-2 diabetes. NFκB is known to upregulate Orai1, the Ca2+ channel responsible for store-operated Ca2+ entry. The present study explored the role of NEFA-sensitive NFκB-dependent Orai1 expression in the regulation of lipid synthesis. Methods: BRL-3A rat liver hepatocyte lines were studied in the absence and presence of NEFA. Transcript and protein expression levels of factors involved in lipid synthesis were quantified by quantitative polymerase chain reaction (qPCR) and western blot analyses. Fatty acids were measured by immunofluorescence. Results: NEFA significantly increased, as indicated by the expression of sterol regulatory element-binding protein 1 (SREBP-1c), fatty acid synthase (FAS), acetyl-CoA carboxylase α (ACC1), Orai1, and NFκB p65 by qPCR and western blot analyses. These effects were reversed by the Orai1 inhibitor, 2-aminoethoxydiphenyl borate, and the NFκB inhibitor, wogonin. Furthermore, SREBP-1c, FAS, ACC1, and Orai1 were significantly decreased by Orai1 silencing. Conclusions: Taken together, these results demonstrated that NEFA-sensitive NFκB-dependent Orai1 expression regulates de novo lipogenesis

    Interdigitated Back‐Contacted Carbon Nanotube–Silicon Solar Cells

    Carbon/silicon heterojunctions provide a new perspective for silicon solar cells and in particular those made from carbon nanotubes (CNTs) have already achieved industrial-level power conversion efficiency and device size when using organic passivation and a back-junction design. However, the current state of the art device geometry for silicon photovoltaics is the interdigitated back contact (IBC) cell and this has yet to be demonstrated for CNT/Si solar cells due to the complexity of fabricating the required patterns. Herein, IBC-CNT solar cells are demonstrated via the simple spin coating of a conductive hole-selective passivating film and the evaporation of buried silicon oxide/magnesium electron-selective contacts for both polarities. The CNT coverage area fraction (fCNT) and the gap between the two polarities are optimized to minimize electrical shading loss and ensure high photocarrier collection. Large-area (4.76 cm2) highly efficient (17.53%) IBC-CNT solar cells with a Voc of 651 mV and Jsc of 40.56 mA cm−2 are demonstrated and are prepared with one alignment step for the CNT/Si contact, and photolithographic-free and room-temperature processes. These performance parameters are among the best for solution-processed dopant-free IBC schemes and indicate the feasibility of using low-dimensional carbon materials in IBC solar cells

    Effect of Environmental Planning on Elderly Individual Quality of Life in Severe Cold Regions: A Case Study in Northeastern China

    With the development of urbanization and the ageing population, the improvement of the urban environment and the quality of life (QOL) of the elderly in cities with a cold climate have become critical issues to be addressed. However, only a few studies have focused on this aspect. According to a review of the literature, the contents of the built environment (BE) in severe cold regions are defined as thirteen key factors of four categories (density, environmental aesthetics, outdoor environment, and accessibility) and the QOL of old people consists of three aspects (residential, health, and social satisfaction). This study explores how BE variables are associated with the QOL of older adults by using ordered logit and gologit2 models. The data consist of the results of 1945 questionnaires from field surveys in 11 different residential areas, across two cities in northeastern China. The results show: (1) Walkability in winter, distance to a public park of 400&ndash;1000 m, outdoor shelters and seating, less than five buses available, and a mixture of evergreen and deciduous trees are five of the most important variables of BE that have a significant positive impact on QOL compared with other climatic regions; (2) &ldquo;Pocket parks&rdquo; and pedestrian walkway safety are appropriate approaches to improve wellbeing under local economic conditions. For public transportation, metro and rail transit systems are encouraged, and some rules are needed to reduce the number of buses in harsh weather conditions; (3) Compared with spatial distribution in other climates, the scope of a 15 min city should be less than 1km in severe cold areas. From the findings, we conclude that there are six possible pattern languages to improve the urban environment, and they can provide information for further study on environmental planning in severe cold regions
