Abnormal myofiber morphology and limb dysfunction in claudication
Background Peripheral artery disease (PAD), which affects an estimated 27 million people in Europe and North America, is caused by atherosclerotic plaques that limit blood flow to the legs. Chronic, repeated ischemia in the lower leg muscles of PAD patients is associated with loss of normal myofiber morphology and myofiber degradation. In this study, we tested the hypothesis that myofiber morphometrics of PAD calf muscle are significantly different from normal calf muscle and correlate with reduced calf muscle strength and walking performance. Methods Gastrocnemius biopsies were collected from 154 PAD patients (Fontaine stage II) and 85 control subjects. Morphometric parameters of gastrocnemius fibers were determined and evaluated for associations with walking distances and calf muscle strength. Results Compared with control myofibers, PAD myofiber cross-sectional area, major and minor axes, equivalent diameter, perimeter, solidity, and density were significantly decreased (P \u3c 0.005), whereas roundness was significantly increased (P \u3c 0.005). Myofiber morphometric parameters correlated with walking distances and calf muscle strength. Multiple regression analyses demonstrated myofiber cross-sectional area, roundness, and solidity as the best predictors of calf muscle strength and 6-min walking distance, whereas cross-sectional area was the main predictor of maximum walking distance. Conclusions Myofiber morphometrics of PAD gastrocnemius differ significantly from those of control muscle and predict calf muscle strength and walking distances of the PAD patients. Morphometric parameters of gastrocnemius myofibers may serve as objective criteria for diagnosis, staging, and treatment of PAD

Systemic multicompartmental effects of the gut microbiome on mouse metabolic phenotypes
To characterize the impact of gut microbiota on host metabolism, we investigated the multicompartmental metabolic profiles of a conventional mouse strain (C3H/HeJ) (n=5) and its germ-free (GF) equivalent (n=5). We confirm that the microbiome strongly impacts on the metabolism of bile acids through the enterohepatic cycle and gut metabolism (higher levels of phosphocholine and glycine in GF liver and marked higher levels of bile acids in three gut compartments). Furthermore we demonstrate that (1) well-defined metabolic differences exist in all examined compartments between the metabotypes of GF and conventional mice: bacterial co-metabolic products such as hippurate (urine) and 5-aminovalerate (colon epithelium) were found at reduced concentrations, whereas raffinose was only detected in GF colonic profiles. (2) The microbiome also influences kidney homeostasis with elevated levels of key cell volume regulators (betaine, choline, myo-inositol and so on) observed in GF kidneys. (3) Gut microbiota modulate metabotype expression at both local (gut) and global (biofluids, kidney, liver) system levels and hence influence the responses to a variety of dietary modulation and drug exposures relevant to personalized health-care investigations

Search for B+ -> D*+ pi0 decay
We report on a search for the doubly Cabibbo suppressed decay B+ -> D*+ pi0,
based on a data sample of 657 million BBbar pairs collected at the Upsilon(4S)
resonance with the Belle detector at the KEKB asymmetric energy e+ e- collider.
We find no significant signal and set an upper limit of Br(B+ -> D*+ pi0) < 3.6
x 10^-6 at the 90% confidence level. This limit can be used to constrain the
ratio between suppressed and favored B -> D* pi decay amplitudes, r < 0.051, at

Search for CP Violation in the Decays
We have searched for CP violation in the decays
where denotes a neutral pseudo-scalar meson which is either a ,
, or using KEKB asymmetric-energy collision data
corresponding to an integrated luminosity of 791 fb collected with the
Belle detector. No evidence of significant CP violation is observed. We report
the most precise CP asymmetry measurement in the decay to date: . We also report the first measurements of
CP asymmetries in the decays and : and , respectively
Study of the decays B->D_s1(2536)+ anti-D(*)
We report a study of the decays B -> D_s1(2536)+ anti-D(*), where anti-D(*)
is anti-D0, D- or D*-, using a sample of 657 x 10^6 B anti-B pairs collected at
the Upsilon(4S) resonance with the Belle detector at the KEKB asymmetric-energy
e+e- collider. The branching fractions of the decays B+ -> D_s1(2536)+ anti-D0,
B0 -> D_s1(2536)+ D- and B0 -> D_s1(2536)+ D*- multiplied by that of
D_s1(2536)+ -> (D*0K+ + D*+K0) are found to be (3.97+-0.85+-0.56) x 10^-4,

assuming a simple Cabibbo-suppression
factor of . The small branching fraction could be attributed
to a suppression due to the large momentum of the baryonic decay products,
which has been observed in other charmed baryonic two-body B decays. trend
