    Complement factor H and the hemolytic uremic syndrome

    Immune recognition is coupled to powerful proinflammatory effector pathways that must be tightly regulated. The ancient alternative pathway of complement activation is one such proinflammatory pathway. Genetic susceptibility factors have been identified in both regulators and activating components of the alternative pathway that are associated with thrombotic microangiopathies, glomerulonephritides, and chronic conditions featuring debris deposition. These observations indicate that excessive alternative pathway activation promotes thrombosis in the microvasculature and tissue damage during debris accumulation. Intriguingly, distinct genetic changes in factor H (FH), a key regulator of the alternative pathway, are associated with hemolytic uremic syndrome (HUS), membranoproliferative glomerulonephritis (dense deposit disease), or age-related macular degeneration (AMD). A mouse model of HUS designed to mirror human mutations in FH has now been developed, providing new understanding of the molecular pathogenesis of complement-related endothelial disorders

    Survival of Organic Materials in Hypervelocity Impacts of Ice on Sand, Ice, and Water in the Laboratory

    The survival of organic molecules in shock impact events has been investigated in the laboratory. A frozen mixture of anthracene and stearic acid, solvated in dimethylsulfoxide (DMSO), was fired in a two-stage light gas gun at speeds of ?2 and ?4?km s?1 at targets that included water ice, water, and sand. This involved shock pressures in the range of 2–12 GPa. It was found that the projectile materials were present in elevated quantities in the targets after impact and in some cases in the crater ejecta as well. For DMSO impacting water at 1.9?km s?1 and 45° incidence, we quantify the surviving fraction after impact as 0.44±0.05. This demonstrates successful transfer of organic compounds from projectile to target in high-speed impacts. The range of impact speeds used covers that involved in impacts of terrestrial meteorites on the Moon, as well as impacts in the outer Solar System on icy bodies such as Pluto. The results provide laboratory evidence that suggests that exogenous delivery of complex organic molecules from icy impactors is a viable source of such material on target bodies

    Self-collected gargle fluids and nasopharyngeal swabs as a strategy for molecular diagnostics of respiratory viruses

    Diagnosis of respiratory viruses traditionally relies on deep oropharynx or nasopharynx swabs collected by healthcare workers (HCW). However, outpatients must make an appointment, and the procedure can cause discomfort in patients. Self-collecting has the potential as a strategy to improve participants’ willingness to participate in diagnostics, surveillance, or studies. We compared self-collected gargle fluids and nasopharyngeal swabs as a strategy for molecular diagnostics of respiratory viruses and compared the average cycle threshold (Ct)-values with those of samples collected by HCW. The study was conducted among technicians of the Laboratory of Clinical Microbiology and Infectious Diseases, Zwolle, the Netherlands, and their family members, between April 2019 and March 2020. It included a questionnaire regarding the severity and date of first symptoms and an assessment of the sampling experience. The primary outcome was the mean Ct of positive PCRs. Similar mean Ct values were obtained using self- or HCW-collected swabs. In addition, gargle fluids and self-swabbed specimens had comparable detection rates of respiratory viruses. Notably, most participants preferred gargling over self-swabbing. Interestingly, but not surprisingly, the time between the onset of symptoms and sampling was shorter in PCR-positive compared to PCR-negative participants. Though this study was abrogated by the SARS-CoV-2 pandemic, the results indicate that both self-swabs and gargle fluids are acceptable for diagnosing common respiratory viruses in the outpatient population, including influenza virus, rhinovirus, adenovirus, SARS-CoV-2 and endemic human coronaviruses. Gargling could be considered an alternative sampling strategy and may enhance willingness to participate in screenings or diagnostics for respiratory viruses

    Understanding the Gap between Website Value and Consumer Shopping Orientation: An Application of Task-Technology Fit Theory to Online shopping Values

    Value judgments, such as functional, emotional, utilitarian, and hedonic values have received considerable attention in recent years. However, research on these interrelationships has been somewhat limited in online shopping, particularly in regards to the gap between website values versus consumer shopping orientations. The purpose of this research is to propose and empirically test a conceptual framework that incorporates the interrelationships of website values, shopping satisfaction, and repurchase intention into framework and validate them in a B2C online shopping context. Specifically, we examine the moderating role of consumer shopping orientations in the impact of website values on shopping satisfaction. Our results suggest that the impact of website values on shopping satisfaction is negatively moderated by consumer shopping orientations. Therefore, functional and emotional values are probably necessary but are an insufficient precondition for cultivating shopping satisfaction. Moreover, consumer hedonic orientation plays a slightly higher influence on shopping satisfaction than utilitarian orientation. Understanding these factors can provide direction for planning website features as well as consumer desired values that will increase shopping satisfaction and repurchase intention

    Assessment of pesticide residues in tomatoes and watermelons (fruits) from markets in Dar es Salaam, Tanzania

    This study investigated the levels of pesticide residues in selected fruits from major markets in Dar es Salaam city. Samples of tomatoes and watermelons were analysed for eighteen organochlorine, organophosphorus and pyrethroid pesticide residues. Extraction was performed using acetone followed by dichloromethane: cyclohexane mixture and the extracts were cleanedup using florisil. The compounds were determined by gas chromatography–mass spectrometry (GC–MS). Pesticides and metabolites were detected in 95.8% of the samples. The compounds detected included chlorpyrifos, α-endosulfan, β-endosulfan and cypermethrin and their highest concentrations were 3810 ± 50, 370 ± 20, 120 ± 6 and 50 ± 4 ng/g, respectively. Others were p,p'- DDD, o,p'-DDD, p,p'-DDE and α-HCH with highest concentrations varying from 1 ± 0.5 to 20 ± 1.2 ng/g. There were no significant variations in concentrations of the pesticide residues between the fruits and among the sampling sites, indicating similarities in contamination patterns. The concentrations of the contaminants were above the maximum residue limits (MRLs) in 41.7% to 50% of the tomatoes and watermelons indicating risks and concerns for public health. The Tanzanian agrochemicals and food regulatory agencies (e.g. TPRI, TFDA and TBS) and the government in general should ensure strict applications of laws that regulate pesticides in the country and develop effective educational programmes for farmers to apply good agricultural practices such as reducing the use of pesticides, applying appropriate pesticides and doses, and restrict the spray before harvesting.     Radiating dipoles in photonic crystals

    The radiation dynamics of a dipole antenna embedded in a Photonic Crystal are modeled by an initially excited harmonic oscillator coupled to a non--Markovian bath of harmonic oscillators representing the colored electromagnetic vacuum within the crystal. Realistic coupling constants based on the natural modes of the Photonic Crystal, i.e., Bloch waves and their associated dispersion relation, are derived. For simple model systems, well-known results such as decay times and emission spectra are reproduced. This approach enables direct incorporation of realistic band structure computations into studies of radiative emission from atoms and molecules within photonic crystals. We therefore provide a predictive and interpretative tool for experiments in both the microwave and optical regimes.Comment: Phys. Rev. E, accepte
