    Dysregulation of microtubule stability impairs morphofunctional connectivity in primary neuronal networks

    Functionally related neurons assemble into connected networks that process and transmit electrochemical information. To do this in a coordinated manner, the number and strength of synaptic connections is tightly regulated. Synapse function relies on the microtubule (MT) cytoskeleton, the dynamics of which are in turn controlled by a plethora of MT-associated proteins, including the MT-stabilizing protein Tau. Although mutations in the Tau-encodingMAPT gene underlie a set of neurodegenerative disorders, termed tauopathies, the exact contribution of MT dynamics and the perturbation thereof to neuronal network connectivity has not yet been scrutinized. Therefore, we investigated the impact of targeted perturbations of MT stability on morphological (e.g., neurite- and synapse density) and functional (e.g., synchronous calcium bursting) correlates of connectivity in networks of primary hippocampal neurons. We found that treatment with MT-stabilizing or -destabilizing compounds impaired morphofunctional connectivity in a reversible manner. We also discovered that overexpression of MAPT induced significant connectivity defects, which were accompanied by alterations in MT dynamics and increased resistance to pharmacological MT depolymerization. Overexpression of a MAPT variant harboring the P301L point mutation in the MT-binding domain did far less, directly linking neuronal connectivity with Tau's MT binding affinity. Our results show that MT stability is a vulnerable node in tauopathies and that its precise pharmacological tuning may positively affect neuronal network connectivity. However, a critical balance in MT turnover causes it to be a difficult therapeutic target with a narrow operating window

    Perkembangan ilmu pengetahuan dan teknologi membawa pengaruh akan meningkatnya kebutuhan akan daya listrik, namun ketersediaan sumber energi konvensional (fossil) semakin menipis yang tentunya akan berdampak pada tingkat ketahanan listrik. Oleh karenanya diperlukan pembangkit-pembangkit tersebar berskala kecil (microgrid). Pembangkit tersebar ini diupayakan bersumber pada energi terbarukan dengan meminimalkan pemakaian dari sumber energi konvensional serta digunakan energy storage untuk power balance. Oleh karena adanya microgrid ini maka penting untuk menentukan besarnya pembangkitan daya listrik yang optimal dari masing-masing pembangkit dan kapasitas optimal energy storage sehingga kebutuhan daya listrik dapat dipenuhi dengan biaya yang optimal. Pada penelitian ini dilakukan analisis mengenai economic dispatch di dalam pengoperasian sistem kelistrikan microgrid dengan energy storage. Algoritma APSO digunakan untuk memecahkan masalah minimalisasi total biaya sistem. Simulasi komputer menggunakan Matlab dilakukan untuk menunjukkan efektivitas dari metodologi yang diusulkan dan dampak dari harga dan sistem penyimpanan pada economic dispatch. Hasil simulasi menunjukkan bahwa penggunaan metode APSO untuk ED pada sistem kelistrikan microgrid memiliki performa kecepatan konvergensi yang lebih baik dibanding metode PSO dan dengan pemanfaatan energy storage pada sistem memberikan dampak penghematan biaya operasi

    Magnetized Accretion-Ejection Structures: 2.5D MHD simulations of continuous Ideal Jet launching from resistive accretion disks

    We present numerical magnetohydrodynamic (MHD) simulations of a magnetized accretion disk launching trans-Alfvenic jets. These simulations, performed in a 2.5 dimensional time-dependent polytropic resistive MHD framework, model a resistive accretion disk threaded by an initial vertical magnetic field. The resistivity is only important inside the disk, and is prescribed as eta = alpha_m V_AH exp(-2Z^2/H^2), where V_A stands for Alfven speed, H is the disk scale height and the coefficient alpha_m is smaller than unity. By performing the simulations over several tens of dynamical disk timescales, we show that the launching of a collimated outflow occurs self-consistently and the ejection of matter is continuous and quasi-stationary. These are the first ever simulations of resistive accretion disks launching non-transient ideal MHD jets. Roughly 15% of accreted mass is persistently ejected. This outflow is safely characterized as a jet since the flow becomes super-fastmagnetosonic, well-collimated and reaches a quasi-stationary state. We present a complete illustration and explanation of the `accretion-ejection' mechanism that leads to jet formation from a magnetized accretion disk. In particular, the magnetic torque inside the disk brakes the matter azimuthally and allows for accretion, while it is responsible for an effective magneto-centrifugal acceleration in the jet. As such, the magnetic field channels the disk angular momentum and powers the jet acceleration and collimation. The jet originates from the inner disk region where equipartition between thermal and magnetic forces is achieved.     General-relativistic Resistive Magnetohydrodynamics with Robust Primitive-variable Recovery for Accretion Disk Simulations

    Recent advances in black hole astrophysics, particularly the first visual evidence of a supermassive black hole at the center of the galaxy M87 by the Event Horizon Telescope, and the detection of an orbiting "hot spot" nearby the event horizon of Sgr A* in the Galactic center by the Gravity Collaboration, require the development of novel numerical methods to understand the underlying plasma microphysics. Non-thermal emission related to such hot spots is conjectured to originate from plasmoids that form due to magnetic reconnection in thin current layers in the innermost accretion zone. Resistivity plays a crucial role in current sheet formation, magnetic reconnection, and plasmoid growth in black hole accretion disks and jets. We included resistivity in the three-dimensional general-relativistic magnetohydrodynamics (GRMHD) code BHAC and present the implementation of an implicit–explicit scheme to treat the stiff resistive source terms of the GRMHD equations. The algorithm is tested in combination with adaptive mesh refinement to resolve the resistive scales and a constrained transport method to keep the magnetic field solenoidal. Several novel methods for primitive-variable recovery, a key part in relativistic magnetohydrodynamics codes, are presented and compared for accuracy, robustness, and efficiency. We propose a new inversion strategy that allows for resistive-GRMHD simulations of low gas-to-magnetic pressure ratio and highly magnetized regimes as applicable for black hole accretion disks, jets, and neutron-star magnetospheres.     A global framework for action to improve the primary care response to chronic non-communicable diseases: a solution to a neglected problem.

    BACKGROUND: Although in developing countries the burden of morbidity and mortality due to infectious diseases has often overshadowed that due to chronic non-communicable diseases (NCDs), there is evidence now of a shift of attention to NCDs. DISCUSSION: Decreasing the chronic NCD burden requires a two-pronged approach: implementation of the multisectoral policies aimed at decreasing population-level risks for NCDs, and effective and affordable delivery of primary care interventions for patients with chronic NCDs. The primary care response to common NCDs is often unstructured and inadequate. We therefore propose a programmatic, standardized approach to the delivery of primary care interventions for patients with NCDs, with a focus on hypertension, diabetes mellitus, chronic airflow obstruction, and obesity. The benefits of this approach will extend to patients with related conditions, e.g. those with chronic kidney disease caused by hypertension or diabetes. This framework for a "public health approach" is informed by experience of scaling up interventions for chronic infectious diseases (tuberculosis and HIV). The lessons learned from progress in rolling out these interventions include the importance of gaining political commitment, developing a robust strategy, delivering standardised interventions, and ensuring rigorous monitoring and evaluation of progress towards defined targets. The goal of the framework is to reduce the burden of morbidity, disability and premature mortality related to NCDs through a primary care strategy which has three elements: 1) identify and address modifiable risk factors, 2) screen for common NCDs and 3) and diagnose, treat and follow-up patients with common NCDs using standard protocols. The proposed framework for NCDs borrows the same elements as those developed for tuberculosis control, comprising a goal, strategy and targets for NCD control, a package of interventions for quality care, key operations for national implementation of these interventions (political commitment, case-finding among people attending primary care services, standardised diagnostic and treatment protocols, regular drug supply, and systematic monitoring and evaluation), and indicators to measure progress towards increasing the impact of primary care interventions on chronic NCDs. The framework needs evaluation, then adaptation in different settings. SUMMARY: A framework for a programmatic "public health approach" has the potential to improve on the current unstructured approach to primary care of people with chronic NCDs. Research to establish the cost, value and feasibility of implementing the framework will pave the way for international support to extend the benefit of this approach to the millions of people worldwide with chronic NCDs

    Progressive tau aggregation does not alter functional brain network connectivity in seeded hTau.P301L mice

    Progressive accumulation of hyperphosphorylated tau is a hallmark of various neurodegenerative disorders including Alzheimer's disease. However, to date, the functional effects of tau pathology on brain network connectivity remain poorly understood. To directly interrogate the impact of tau pathology on functional brain connectivity, we conducted a longitudinal experiment in which we monitored a fibril-seeded hTau.P301L mouse model using correlative whole-brain microscopy and resting-state functional MRI. Despite a progressive aggravation of tau pathology across the brain, the major resting-state networks appeared unaffected up to 15 weeks after seeding. Targeted analyses also showed that the connectivity of regions with high levels of hyperphosphorylated tau was comparable to that observed in controls. In line with the ostensible retention of connectivity, no behavioural changes were detected between seeded and control hTau.P301L mice as determined by three different paradigms. Our data indicate that seeded tau pathology, with accumulation of tau aggregates throughout different regions of the brain, does not alter functional connectivity or behaviour in this mouse model. Additional correlative functional studies on different mouse models should help determine whether this is a generalizable trait of tauopathies
