    Probabilistic Analysis of a Composite Crew Module

    An approach for conducting reliability-based analysis (RBA) of a Composite Crew Module (CCM) is presented. The goal is to identify and quantify the benefits of probabilistic design methods for the CCM and future space vehicles. The coarse finite element model from a previous NASA Engineering and Safety Center (NESC) project is used as the baseline deterministic analysis model to evaluate the performance of the CCM using a strength-based failure index. The first step in the probabilistic analysis process is the determination of the uncertainty distributions for key parameters in the model. Analytical data from water landing simulations are used to develop an uncertainty distribution, but such data were unavailable for other load cases. The uncertainty distributions for the other load scale factors and the strength allowables are generated based on assumed coefficients of variation. Probability of first-ply failure is estimated using three methods: the first order reliability method (FORM), Monte Carlo simulation, and conditional sampling. Results for the three methods were consistent. The reliability is shown to be driven by first ply failure in one region of the CCM at the high altitude abort load set. The final predicted probability of failure is on the order of 10-11 due to the conservative nature of the factors of safety on the deterministic loads

    Systems Methodology and Framework for Problem Definition in Mobile Ad Hoc Networks

    Mobile Ad Hoc Networks are communication networks built up of a collection of mobile devices which can communicate through wireless connections. Mobile Ad Hoc Networks have many challenges such as routing, which is the task of directing data packets from a source node to a given destination. This task is particularly hard in Mobile Ad Hoc Networks: due to the mobility of the network elements and the lack of central control, robustness and adaptability in routing algorithms and work in a decentralized and self organizing way. Through the principles of systems architecting and Engineering; the problem statement in Mobile Ad Hoc Networks could be defined more specifically and accurately. The uncertainties and techniques for mitigating and even taking positive advantages of them can be achieved through a framework of uncertainties as in [1]. The systems methodology framework called Total Systems Intervention (TSI) described by Flood and Jackson [2] select a systems methodology for Mobile Ad Hoc Networks. The purpose of this paper is to show how TSI when integrated with a framework created to understand the risks and opportunities can help develop strategies to minimize the risks and to take advantage of the opportunities for facing challenges in Mobile Ad Hoc Networks

    Structural Analysis of the Right Rear Lug of American Airlines Flight 587

    A detailed finite element analysis of the right rear lug of the American Airlines Flight 587 - Airbus A300-600R was performed as part of the National Transportation Safety Board s failure investigation of the accident that occurred on November 12, 2001. The loads experienced by the right rear lug are evaluated using global models of the vertical tail, local models near the right rear lug, and a global-local analysis procedure. The right rear lug was analyzed using two modeling approaches. In the first approach, solid-shell type modeling is used, and in the second approach, layered-shell type modeling is used. The solid-shell and the layered-shell modeling approaches were used in progressive failure analyses (PFA) to determine the load, mode, and location of failure in the right rear lug under loading representative of an Airbus certification test conducted in 1985 (the 1985-certification test). Both analyses were in excellent agreement with each other on the predicted failure loads, failure mode, and location of failure. The solid-shell type modeling was then used to analyze both a subcomponent test conducted by Airbus in 2003 (the 2003-subcomponent test) and the accident condition. Excellent agreement was observed between the analyses and the observed failures in both cases. The moment, Mx (moment about the fuselage longitudinal axis), has significant effect on the failure load of the lugs. Higher absolute values of Mx give lower failure loads. The predicted load, mode, and location of the failure of the 1985- certification test, 2003-subcomponent test, and the accident condition are in very good agreement. This agreement suggests that the 1985-certification and 2003-subcomponent tests represent the accident condition accurately. The failure mode of the right rear lug for the 1985-certification test, 2003-subcomponent test, and the accident load case is identified as a cleavage-type failure. For the accident case, the predicted failure load for the right rear lug from the PFA is greater than 1.98 times the limit load of the lugs

    Computational identification of strain-, species- and genus-specific proteins

    BACKGROUND: The identification of unique proteins at different taxonomic levels has both scientific and practical value. Strain-, species- and genus-specific proteins can provide insight into the criteria that define an organism and its relationship with close relatives. Such proteins can also serve as taxon-specific diagnostic targets. DESCRIPTION: A pipeline using a combination of computational and manual analyses of BLAST results was developed to identify strain-, species-, and genus-specific proteins and to catalog the closest sequenced relative for each protein in a proteome. Proteins encoded by a given strain are preliminarily considered to be unique if BLAST, using a comprehensive protein database, fails to retrieve (with an e-value better than 0.001) any protein not encoded by the query strain, species or genus (for strain-, species- and genus-specific proteins respectively), or if BLAST, using the best hit as the query (reverse BLAST), does not retrieve the initial query protein. Results are manually inspected for homology if the initial query is retrieved in the reverse BLAST but is not the best hit. Sequences unlikely to retrieve homologs using the default BLOSUM62 matrix (usually short sequences) are re-tested using the PAM30 matrix, thereby increasing the number of retrieved homologs and increasing the stringency of the search for unique proteins. The above protocol was used to examine several food- and water-borne pathogens. We find that the reverse BLAST step filters out about 22% of proteins with homologs that would otherwise be considered unique at the genus and species levels. Analysis of the annotations of unique proteins reveals that many are remnants of prophage proteins, or may be involved in virulence. The data generated from this study can be accessed and further evaluated from the CUPID (Core and Unique Protein Identification) system web site (updated semi-annually) at . CONCLUSION: CUPID provides a set of proteins specific to a genus, species or a strain, and identifies the most closely related organism

    Mining users' significant driving routes with low-power sensors

    While there is significant work on sensing and recognition of significant places for users, little attention has been given to users' significant routes. Recognizing these routine journeys, opens doors to the development of novel applications, like personalized travel alerts, and enhancement of user's travel experience. However, the high energy consumption of traditional location sensing technologies, such as GPS or WiFi based localization, is a barrier to passive and ubiquitous route sensing through smartphones. In this paper, we present a passive route sensing framework that continuously monitors a vehicle user solely through a phone's gyroscope and accelerometer. This approach can differentiate and recognize various routes taken by the user by time warping angular speeds experienced by the phone while in transit and is independent of phone orientation and location within the vehicle, small detours and traffic conditions. We compare the route learning and recognition capabilities of this approach with GPS trajectory analysis and show that it achieves similar performance. Moreover, with an embedded co-processor, common to most new generation phones, it achieves energy savings of an order of magnitude over the GPS sensor.This research has been funded by the EPSRC Innovation and Knowledge Centre for Smart Infrastructure and Construction project (EP/K000314).This is the author accepted manuscript.     Overcoming Communication Delays in Distributed Frequency Regulation

DFR relies on iterative and distributed optimization algorithms to obtain an optimal feedback law for frequency regulation. But, it is, in general, hard to know beforehand how many iterations suffices to ensure stability. This paper develops a framework to determine a lower bound on the number of iterations required for two distributed optimization protocols. This allows prosumers to determine whether they can compute stabilizing control strategies within an acceptable time frame by taking communication delays into account.     Interaction of the NRF2 and p63 transcription factors promotes keratinocyte proliferation in the epidermis

    Epigenetic regulation of cell and tissue function requires the coordinated action of transcription factors. However, their combinatorial activities during regeneration remain largely unexplored. Here, we discover an unexpected interaction between the cytoprotective transcription factor NRF2 and p63-a key player in epithelial morphogenesis. Chromatin immunoprecipitation combined with sequencing and reporter assays identifies enhancers and promoters that are simultaneously activated by NRF2 and p63 in human keratinocytes. Modeling of p63 and NRF2 binding to nucleosomal DNA suggests their chromatin-assisted interaction. Pharmacological and genetic activation of NRF2 increases NRF2-p63 binding to enhancers and promotes keratinocyte proliferation, which involves the common NRF2-p63 target cyclin-dependent kinase 12. These results unravel a collaborative function of NRF2 and p63 in the control of epidermal renewal and suggest their combined activation as a strategy to promote repair of human skin and other stratified epithelia

    In vivo delivery of VEGF RNA and protein to increase osteogenesis and intraosseous angiogenesis

    Deficient bone vasculature is a key component in pathological conditions ranging from developmental skeletal abnormalities to impaired bone repair. Vascularisation is dependent upon vascular endothelial growth factor (VEGF), which drives both angiogenesis and osteogenesis. The aim of this study was to examine the efcacy of blood vessel and bone formation following transfection with VEGF RNA or delivery of recombinant human VEGF165 protein (rhVEGF165) across in vitro and in vivo model systems. To quantify blood vessels within bone, an innovative approach was developed using high-resolution X-ray computed tomography (XCT) to generate quantifable three-dimensional reconstructions. Application of rhVEGF165 enhanced osteogenesis, as evidenced by increased human osteoblast-like MG-63 cell proliferation in vitro and calvarial bone thickness following in vivo administration. In contrast, transfection with VEGF RNA triggered angiogenic efects by promoting VEGF protein secretion from MG-63VEGF165 cells in vitro, which resulted in signifcantly increased angiogenesis in the chorioallantoic (CAM) assay in ovo. Furthermore, direct transfection of bone with VEGF RNA in vivo increased intraosseous vascular branching. This study demonstrates the importance of continuous supply as opposed to a single high dose of VEGF on angiogenesis and osteogenesis and, illustrates the potential of XCT in delineating in 3D, blood vessel connectivity in bone

    A linear programming-based method for job shop scheduling

    We present a decomposition heuristic for a large class of job shop scheduling problems. This heuristic utilizes information from the linear programming formulation of the associated optimal timing problem to solve subproblems, can be used for any objective function whose associated optimal timing problem can be expressed as a linear program (LP), and is particularly effective for objectives that include a component that is a function of individual operation completion times. Using the proposed heuristic framework, we address job shop scheduling problems with a variety of objectives where intermediate holding costs need to be explicitly considered. In computational testing, we demonstrate the performance of our proposed solution approach
