    Estimation of quantitative genetic and stability parameters in maize under high and low N levels

    AB It is important to breed maize (Zea mays L) cultivars with high performance under variable N levels. We studied the effect of N levels and estimated quantitative genetic parameters for grain yield, quality, and other traits, and examined stability of performance for grain yield in diverse Chinese maize germplasm. From 2006 to 2008, each year 20 and in total 30 maize hybrids, including commercial hybrids currently grown in this region and other ex¬perimental hybrids as well as high-oil hybrids, were tested using nine environments (location-year combinations) in North China Plain. In each environment, two replicated trials were grown: one under high N application rate (HN, 225 kg N ha-1) and the other under low N application rate (LN, no N fertilization). Compared to HN, grain yield was significantly reduced (35.6%) under LN level, as well as kernel number per ear, 1000-kernel weight, plant and ear heights, and protein concentration. In the analysis over environments under each N level, genotypic variance was significant and heritability was high for all traits. In the analyses across N levels and environments, genotypic variance was significant for all traits and larger than the genotype × N and/or environment interaction variance components except for protein concentration. In stability analyses across N levels, hybrids differed for their linear response to environments, and some showed dissimilar response under HN and LN levels. Our results indicated that breeding maize adapted to variable N levels is feasible with the Chinese germplasm available in the summer breeding programs in North China Plain. Multi-environment tests are required to identify hybrids with high grain yield under variable N conditions, and examining yield stability separately under HN and LN would be useful

    Genetic diversity and linkage disequilibrium of two homologous genes to maize D8: sorghum SbD8 and pearl millet PgD8

    Yield and yield stability of sorghum [Sorghum bicolor (L.) Moench.] and pearl millet [Pennisetum glaucum (L.) R.Br.] are highly influenced by flowering time and photoperiodic sensitivity in the arid to semi-arid regions of West and Central Africa. Photoperiodic sensitivity is the key adaptation trait of local landraces because it assures flowering at the end of the rainy season, independent of variable dates of planting. Flowering time genes are mainly integrated into four pathways with close interaction among each other: Vernalization, autonomous, GA (gibberellic acid) and photoperiod. In the GA pathway, maize D8, wheat RHT1 and rice SLR have been identified as homologous genes to the Arabidopsis GAI, which is a negative regulator of GA response. We have identified two homologous genes to D8: Sorghum SbD8 and pearl millet PgD8. These genes were expressed in the root and leaves of sorghum and pearl millet as revealed by EST database search and reverse transcription PCR, respectively. The genetic diversity of SbD8 was considerably lower than that of PgD8. The extent of linkage disequilibrium in PgD8 is lower than that of maize D8. SbD8 and PgD8 polymorphisms might be appropriate for dissection of photoperiod sensitivity using association mapping approaches

    Optimum allocation of resources for QTL detection using a nested association mapping strategy in maize

    In quantitative trait locus (QTL) mapping studies, it is mandatory that the available financial resources are spent in such a way that the power for detection of QTL is maximized. The objective of this study was to optimize for three different fixed budgets the power of QTL detection 1 − β* in recombinant inbred line (RIL) populations derived from a nested design by varying (1) the genetic complexity of the trait, (2) the costs for developing, genotyping, and phenotyping RILs, (3) the total number of RILs, and (4) the number of environments and replications per environment used for phenotyping. Our computer simulations were based on empirical data of 653 single nucleotide polymorphism markers of 26 diverse maize inbred lines which were selected on the basis of 100 simple sequence repeat markers out of a worldwide sample of 260 maize inbreds to capture the maximum genetic diversity. For the standard scenario of costs, the optimum number of test environments (Eopt) ranged across the examined total budgets from 7 to 19 in the scenarios with 25 QTL. In comparison, the Eopt values observed for the scenarios with 50 and 100 QTL were slightly higher. Our finding of differences in 1 − β* estimates between experiments with optimally and sub-optimally allocated resources illustrated the potential to improve the power for QTL detection without increasing the total resources necessary for a QTL mapping experiment. Furthermore, the results of our study indicated that also in studies using the latest genomics tools to dissect quantitative traits, it is required to evaluate the individuals of the mapping population in a high number of environments with a high number of replications per environment

    Selection Strategy for Sorghum Targeting Phosphorus-limited Environments in West Africa: Analysis of Multi-environment Experiments

    Although sorghum [Sorghum bicolor (L.) Moench] in West Africa (WA) is generally cultivated with limited or no fertilization on soils of low phosphorus availability, no assessments of the genetic variation among WA sorghum varieties for adaptation to low soil P are known. We assessed grain yields of 70 diverse sorghum genotypes under −P (no P fertilization) and +P conditions at two locations in Mali over 5 yr. Genetic variation for grain yield under −P conditions and the feasibility and necessity of sorghum varietal testing for grain yield under −P conditions were evaluated. Delayed heading dates (0–9.8 d) and reductions of grain yield (2–59%) and plant height (13–107 cm) were observed in −P relative to the +P trials. High estimates of genetic variance and broad-sense heritabilities were found for grain yield across both −P (h2 = 0.93) and +P (h2 = 0.92) environments. The genetic correlation for grain yield performance between −P and +P conditions was high (rG = 0.89), suggesting that WA sorghum varieties generally possess good adaptation to low-P conditions. However, genotype × phosphorus crossover interaction was observed between some of the highest yielding genotypes from the −P and +P selected sets, with the variety IS 15401 showing specific adaptation to −P. Direct selection for grain yield in −P conditions was predicted to be 12% more efficient than indirect selection in +P conditions. Thus, selection under −P conditions should be useful for sorghum improvement in WA

    Marker-Assisted Breeding of Improved Maternal Haploid Inducers in Maize for the Tropical/Subtropical Regions

    For efficient production of doubled haploid (DH) lines in maize, maternal haploid inducer lines with high haploid induction rate (HIR) and good adaptation to the target environments is an important requirement. In this study, we present second-generation Tropically Adapted Inducer Lines (2GTAILs), developed using marker assisted selection (MAS) for qhir1, a QTL with a significant positive effect on HIR from the crosses between elite tropical maize inbreds and first generation Tropically Adapted Inducers Lines (TAILs). Evaluation of 2GTAILs for HIR and agronomic performance in the tropical and subtropical environments indicated superior performance of 2GTAILs over the TAILs for both HIR and agronomic performance, including plant vigor, delayed flowering, grain yield, and resistance to ear rots. One of the new inducers 2GTAIL006 showed an average HIR of 13.1% which is 48.9% higher than the average HIR of the TAILs. Several other 2GTAILs also showed higher HIR compared to the TAILs. While employing MAS for qhir1 QTL, we observed significant influence of the non-inducer parent on the positive effect of qhir1 QTL on HIR. The non-inducer parents that resulted in highest mean HIR in the early generation qhir1+ families also gave rise to highest numbers of candidate inducers, some of which showed transgressive segregation for HIR. The mean HIR of early generation qhir1+ families involving different non-inducer parents can potentially indicate recipient non-inducer parents that can result in progenies with high HIR. Our study also indicated that the HIR associated traits (endosperm abortion rate, embryo abortion rate, and proportion of haploid plants among the inducer plants) can be used to differentiate inducers vs. non-inducers but are not suitable for differentiating inducers with varying levels of haploid induction rates. We propose here an efficient methodology for developing haploid inducer lines combining MAS for qhir1 with HIR associated traits

    Improved Heterosis Prediction by Combining Information on DNA- and Metabolic Markers

    Background: Hybrids represent a cornerstone in the success story of breeding programs. The fundamental principle underlying this success is the phenomenon of hybrid vigour, or heterosis. It describes an advantage of the offspring as compared to the two parental lines with respect to parameters such as growth and resistance against abiotic or biotic stress. Dominance, overdominance or epistasis based models are commonly used explanations. Conclusion/Significance: The heterosis level is clearly a function of the combination of the parents used for offspring production. This results in a major challenge for plant breeders, as usually several thousand combinations of parents have to be tested for identifying the best combinations. Thus, any approach to reliably predict heterosis levels based on properties of the parental lines would be highly beneficial for plant breeding. Methodology/Principal Findings: Recently, genetic data have been used to predict heterosis. Here we show that a combination of parental genetic and metabolic markers, identified via feature selection and minimum-description-length based regression methods, significantly improves the prediction of biomass heterosis in resulting offspring. These findings will help furthering our understanding of the molecular basis of heterosis, revealing, for instance, the presence of nonlinear genotype-phenotype relationships. In addition, we describe a possible approach for accelerated selection in plant breeding

    Effects of general practitioner training and family support services on the care of home-dwelling dementia patients - Results of a controlled cluster-randomized study

    <p>Abstract</p> <p>Background</p> <p>More than 90% of dementia patients are cared for by their general practitioners, who are decisively involved in the diagnosis, therapy and recommendation of support services. <it>Objective: </it>To test whether special training of general practitioners alters the care of dementia patients through their systematic recommendation of caregiver counseling and support groups.</p> <p>Method</p> <p>129 general practitioners enrolled 390 dementia patients and their informal caregivers in a prospective, three-arm cluster-randomized 2-year study. Arm A constituted usual care, in Arm B and C support groups and caregiver counseling (in Arm B one year after baseline, in Arm C at baseline) were recommended by the general practitioners. The general practitioners received arm-specific training. Diagnostic and therapeutic behavior of physicians was recorded at baseline. Informal caregivers were questioned in follow-up after 2 years about the utilization of support services.</p> <p>Results</p> <p>The diagnostic behavior of the general practitioners conforms to relevant guidelines. The procedure in newly-diagnosed patients does not differ from previously diagnosed patients with the exception of the rate of referral to a specialist. About one-third of the newly-diagnosed dementia patients are given an anti-dementia drug. The utilization of support groups and counseling increased five- and fourfold, respectively. Utilization of other support services remained low (< 10%), with the exception of home nursing and institutional short-term nursing.</p> <p>Conclusion</p> <p>Trained general practitioners usually act in conformity with guidelines with respect to diagnosing dementia, and partly in conformity with the guidelines with respect to recommended drug therapy. Recommendations of support services for informal caregivers by the general practitioner are successful. They result in a marked increase in the utilization rate for the recommended services compared to offers which are not recommended by the general practitioner.</p> <p>Trial registration</p> <p>ISRCTN68329593</p

    Empirical Comparison of Simple Sequence Repeats and Single Nucleotide Polymorphisms in Assessment of Maize Diversity and Relatedness

    While Simple Sequence Repeats (SSRs) are extremely useful genetic markers, recent advances in technology have produced a shift toward use of single nucleotide polymorphisms (SNPs). The different mutational properties of these two classes of markers result in differences in heterozygosities and allele frequencies that may have implications for their use in assessing relatedness and evaluation of genetic diversity. We compared analyses based on 89 SSRs (primarily dinucleotide repeats) to analyses based on 847 SNPs in individuals from the same 259 inbred maize lines, which had been chosen to represent the diversity available among current and historic lines used in breeding. The SSRs performed better at clustering germplasm into populations than did a set of 847 SNPs or 554 SNP haplotypes, and SSRs provided more resolution in measuring genetic distance based on allele-sharing. Except for closely related pairs of individuals, measures of distance based on SSRs were only weakly correlated with measures of distance based on SNPs. Our results suggest that 1) large numbers of SNP loci will be required to replace highly polymorphic SSRs in studies of diversity and relatedness and 2) relatedness among highly-diverged maize lines is difficult to measure accurately regardless of the marker system
