    B-lynch suture in the management of uterine atony during caesarean section section

    Background: Third stage of labour is still the “sword of Damocle’s” hanging above an obstetrician , inspite  of  today’s advanced technologies and personal care .The importance in the management of this deadly stage lies in the anticipation of complications and being quick enough to treat them timely.  Hence in such scenarios, B-lynch suture is most popular method in treatment of uterine atony during caesarean section. The objective is to study and evaluate the cases in which the B-Lynch suture was used to treat the uterine atony during caesarean section.Methods: A prospective randomized study consisting of 50 women with high risk factors for atonic pph during caesarean section were included as study group patients were subjected to B-Lynch suture application when conventional drugs failed to control PPH. Blood loss was measured using a measuring jar. The fall in Hb% and the need for blood transfusion, and the need for hysterectomy as a last resort to save the life was evaluated.Results: The average blood loss was 1490 ml and the majority of patients had reduction in Hb% from 1.1-1.5 gm% (52%), 36% of the patients did not receive any blood transfusion after B-Lynch suture, and in 80% of cases hysterectomy was avoided.Conclusions: Our study shows cases treated with B-Lynch procedure showed that it is an effective method of containing pph.  It has the advantage of being applied easily and safely. It should be attempted when conservative management fails and before any radical surgery is considered

    Sorptivity of self-compacting concrete containing fly ash and silica fume

    This paper presents the surface water absorption of self-compacting concrete (SCC) containing fly ash and silica fume using sorptivity test. Ordinary Portland cement was partially replaced by various combinations of fly ash and silica fume. Test results show that the presence of fly ash and silica fume significantly reduce the surface water absorption of self-compacting concrete at a water-binder ratio of 0.38. When only fly ash is used to partially replace Ordinary Portland cement, a more noticeable reduction in sorptivity is found when the fly ash content is greater than 20%

    An Enhanced Analysis of Traffic Intelligence in Smart Cities Using Sustainable Deep Radial Function

    Smart cities have revolutionized urban living by incorporating sophisticated technologies to optimize various aspects of urban infrastructure, such as transportation systems. Effective traffic management is a crucial component of smart cities, as it has a direct impact on the quality of life of residents and tourists. Utilizing deep radial basis function (RBF) networks, this paper describes a novel strategy for enhancing traffic intelligence in smart cities. Traditional methods of traffic analysis frequently rely on simplistic models that are incapable of capturing the intricate patterns and dynamics of urban traffic systems. Deep learning techniques, such as deep RBF networks, have the potential to extract valuable insights from traffic data and enable more precise predictions and decisions. In this paper, we propose an RBF based method for enhancing smart city traffic intelligence. Deep RBF networks combine the adaptability and generalization capabilities of deep learning with the discriminative capability of radial basis functions. The proposed method can effectively learn intricate relationships and nonlinear patterns in traffic data by leveraging the hierarchical structure of deep neural networks. The deep RBF model can learn to predict traffic conditions, identify congestion patterns, and make informed recommendations for optimizing traffic management strategies by incorporating these rich and diverse data To evaluate the efficacy of our proposed method, extensive experiments and comparisons with real world traffic datasets from a smart city environment were conducted. In terms of prediction accuracy and efficiency, the results demonstrate that the deep RBF based approach outperforms conventional traffic analysis methods. Smart city traffic intelligence is enhanced by the model capacity to capture nonlinear relationships and manage large scale data sets.Comment: 25 pages, 6 figures, and 3 Table

    Knowledge and Attitude towards Risk Factors and Complications of Diabetes among Housekeeping Staff at a Tertiary Care Center in India

    Objective: This study aimed to assess the knowledge and attitude towards risk factors and complications of diabetes among housekeeping workers. Material and methods: After obtaining institutional ethics approval, a cross-sectional survey was conducted among housekeeping staff working in a tertiary care center with an age group ranging from 18 to 60 years to assess knowledge and attitude towards risk factors and complications of diabetes. The correlation between knowledge and attitude scores was determined using Karl Pearson correlation coefficient analysis. Results: A total of 287 housekeeping staff, with majority of females (73.9%) were included in the study. The average knowledge score was 16.24 ± 6.74, and the average attitude score was 7.22 ± 1.27. A weak correlation between knowledge and attitude scores was observed with a coefficient of 0.215 (p < 0.01). Multivariate linear regression analysis revealed that total knowledge and attitude scores and previously counseled people were statistically significant. Conclusions: The findings revealed that the overall level of knowledge about diabetes among housekeeping staff is poor, but the overall attitude is positive. There was a weak correlation between knowledge and attitude scores. Coordinated educational initiatives focusing on lower socioeconomic background, rural, and less educated individuals are needed to avoid diabetes and its complications

    Prioritization of candidate genes in "QTL-hotspot" region for drought tolerance in chickpea (Cicer arietinum L.)

    A combination of two approaches, namely QTL analysis and gene enrichment analysis were used to identify candidate genes in the "QTL-hotspot" region for drought tolerance present on the Ca4 pseudomolecule in chickpea. In the first approach, a high-density bin map was developed using 53,223 single nucleotide polymorphisms (SNPs) identified in the recombinant inbred line (RIL) population of ICC 4958 (drought tolerant) and ICC 1882 (drought sensitive) cross. QTL analysis using recombination bins as markers along with the phenotyping data for 17 drought tolerance related traits obtained over 1-5 seasons and 1-5 locations split the "QTL-hotspot" region into two subregions namely "QTL-hotspot_a" (15 genes) and "QTL-hotspot_b" (11 genes). In the second approach, gene enrichment analysis using significant marker trait associations based on SNPs from the Ca4 pseudomolecule with the above mentioned phenotyping data, and the candidate genes from the refined "QTL-hotspot" region showed enrichment for 23 genes. Twelve genes were found common in both approaches.     Chromatin-mediated translational control is essential for neural cell fate specification

    Neural cell fate specification is a multistep process in which stem cells undergo sequential changes in states, giving rise to particular lineages such as neurons and astrocytes. This process is accompanied by dynamic changes of chromatin and in transcription, thereby orchestrating lineage-specific gene expression programs. A pressing question is how these events are interconnected to sculpt cell fate. We show that altered chromatin due to loss of the chromatin remodeler Chd5 causes neural stem cell activation to occur ahead of time. This premature activation is accompanied by transcriptional derepression of ribosomal subunits, enhanced ribosome biogenesis, and increased translation. These untimely events deregulate cell fate decisions, culminating in the generation of excessive numbers of astrocytes at the expense of neurons. By monitoring the proneural factor Mash1, we further show that translational control is crucial for appropriate execution of cell fate specification, thereby providing new insight into the interplay between transcription and translation at the initial stages of neurogenesis

    Doubling grain Fe and Zn concentration in sorghum to combat the micronutrient malnutrition in sorghum eating populations

    Dietary induced micronutrient malnutrition (MNM) is one of the greatest global challenges of our times and India has largest number of malnourished people globally. Sorghum is among the major staples and a cheapest sources of micronutrients therefore, biofortification of sorghum is of high priority. From screening of more than 4000 accessions and breeding lines we identified promising donors for Fe and Zn and established the genetic control. Fe and Zn are quantitatively inherited. While grain Zn in predominantly under additive gene control, non-additive gene actions also has role in controlling grain Fe. To develop hybrids with high Fe and Zn both parents should have high Fe and Zn. We demonstrated the prediction of F1 hybrid performance based on mid-parental value for Fe and Zn. Both Fe and Zn are positively correlated (r=0.6 to 0.8) and simultaneous improvement for Fe and Zn is feasible. Using RIL population sorghum genetic map was constructed with 2,088 markers (1148 DArTs, 927 DArTSeqs and 13 SSRs) covering 1355.52 cM with an average marker interval of 0.6cM. Forty-seven QTLs (individual) and 7 QTLs (across) environments with small maineffect and 21 co-localized QTLs for Fe and Zn were identified

    Phosphorylation of cell cycle and apoptosis regulatory protein-1 by stress activated protein kinase P38γ is a novel mechanism of apoptosis signaling by genotoxic chemotherapy

    CARP-1, a perinuclear phospho-protein, regulates cell survival and apoptosis signaling induced by genotoxic drugs. However, kinase(s) phosphorylating CARP-1 and down-stream signal transduction events remain unclear. Here we find that CARP-1 Serine (S)626 and Threonine (T)627 substitution to Alanines (AA) inhibits genotoxic drug-induced apoptosis. CARP-1 T627 is followed by a Proline (P), and this TP motif is conserved in vertebrates. Based on these findings, we generated affinity-purified, anti-phospho-CARP-1 T627 rabbit polyclonal antibodies, and utilized them to elucidate chemotherapy-activated, CARP-1-dependent cell growth signaling mechanisms. Our kinase profiling studies revealed that MAPKs/SAPKs phosphorylated CARP-1 T627. We then UV cross-linked protein extracts from Adriamycin-treated HeLa cervical cancer cells with a CARP-1 (614–638) peptide, and conducted liquid chromatography-tandem mass spectrometry (LC-MS/MS) analyses of the peptide-bound protein complexes. This experiment revealed SAPK p38γ interaction with CARP-1 (614–638) peptide. Our studies further established that SAPK p38γ, but not other MAPKs, phosphorylates CARP-1 T627 in cancer cells treated with genotoxic drugs. Loss of p38γ abrogates CARP-1 T627 phosphorylation, and results in enhanced survival of breast cancer cells by genotoxic drugs. CARP-1 T627 phosphorylation was also noted in breast tumors from patients treated with radiation or endocrine therapies. We conclude that genotoxic drugs activate p38γ-dependent CARP-1 T627 phosphorylation to inhibit cell growth

    Distinct Roles of Brd2 and Brd4 in Potentiating the Transcriptional Program for Th17 Cell Differentiation

    The BET proteins are major transcriptional regulators and have emerged as new drug targets, but their functional distinction has remained elusive. In this study, we report that the BET family members Brd2 and Brd4 exert distinct genomic functions at genes whose transcription they co-regulate during mouse T-helper 17 (Th17) cell differentiation. Brd2 is associated with the chromatin insulator CTCF and the cohesin complex to support cis-regulatory enhancer assembly for gene transcriptional activation. In this context, Brd2 binds the transcription factor Stat3 in an acetylation-sensitive manner and facilitates Stat3 recruitment to active enhancers occupied with transcription factors Irf4 and Batf. In parallel, Brd4 temporally controls RNA polymerase II (Pol II) processivity during transcription elongation through cyclinT1/Cdk9 recruitment and Pol II Ser2 phosphorylation.
