    Antioxidative and antibacterial activities of phenolic compounds from Ficus sur Forssk. and Ficus sycomorus L. (Moraceae) : potential for sickle cell disease treatment in Burkina Faso

    Ficus sur and Ficus sycomorus, two medicinal species used in Burkinabe traditional medicine for the treatment of sickle cell disease were investigated. The determination of total phenolic and tannins contents, using the method of Folin-Ciocalteu (FCR), indicate values significantly (p <0.05) higher in extracts of Ficus sycomorus compared to those of Ficus sur, with respective values of 336.80 ± 0.80 mg TAE/g of extract and 203.74 ± 0.9 mg TAE/g of extract against 247.00 ± 0.52 mg TAE/g of extract and 120.8 ± 0.83 mg TAE/g of extract. Similarly, using the method of 1,1-diphenyl-2-picrylhydrazyl (DPPH), the results of the analysis indicate that extracts of Ficus sycomorus produce the highest antiradical activity with IC50 value of 9.60 ± 0.02 μg/mL against 31.83 ± 0.55 μg/ mL for Ficus sur. The IC50 value of quercetin, used as reference antioxidant, was of 4.6 ± 0.08 μg/mL. The latex of Ficus sycomorus presented the lowest minimum inhibitory concentrations (MIC) against Staphylococcus aureus and Escherichia coli, with respective values of 0.13 mg/mL and 0.25 mg/mL. The difference in phenolic content could explain the difference in biological activity between the two Ficus species.Keywords: drepanocitosis, radical scavenging, tannins, flavonoids, medicinal plant

    Indigenous use and bio-efficacy of medicinal plants in the Rasuwa District, Central Nepal

    <p>Abstract</p> <p>Background</p> <p>By revealing historical and present plant use, ethnobotany contributes to drug discovery and socioeconomic development. Nepal is a natural storehouse of medicinal plants. Although several ethnobotanical studies were conducted in the country, many areas remain unexplored. Furthermore, few studies have compared indigenous plant use with reported phytochemical and pharmacological properties.</p> <p>Methods</p> <p>Ethnopharmacological data was collected in the Rasuwa district of Central Nepal by conducting interviews and focus group discussions with local people. The informant consensus factor (F<sub>IC</sub>) was calculated in order to estimate use variability of medicinal plants. Bio-efficacy was assessed by comparing indigenous plant use with phytochemical and pharmacological properties determined from a review of the available literature. Criteria were used to identify high priority medicinal plant species.</p> <p>Results</p> <p>A total of 60 medicinal formulations from 56 plant species were documented. Medicinal plants were used to treat various diseases and disorders, with the highest number of species being used for gastro-intestinal problems, followed by fever and headache. Herbs were the primary source of medicinal plants (57% of the species), followed by trees (23%). The average F<sub>IC</sub> value for all ailment categories was 0.82, indicating a high level of informant agreement compared to similar studies conducted elsewhere. High F<sub>IC </sub>values were obtained for ophthalmological problems, tooth ache, kidney problems, and menstrual disorders, indicating that the species traditionally used to treat these ailments are worth searching for bioactive compounds: <it>Astilbe rivularis</it>, <it>Berberis asiatica</it>, <it>Hippophae salicifolia, Juniperus recurva</it>, and <it>Swertia multicaulis</it>. A 90% correspondence was found between local plant use and reported plant chemical composition and pharmacological properties for the 30 species for which information was available. Sixteen medicinal plants were ranked as priority species, 13 of which having also been prioritized in a country-wide governmental classification.</p> <p>Conclusions</p> <p>The <it>Tamang </it>people possess rich ethnopharmacological knowledge. This study allowed to identify many high value and high priority medicinal plant species, indicating high potential for economic development through sustainable collection and trade.</p

    Thiopurine withdrawal during sustained clinical remission in inflammatory bowel disease:relapse and recapture rates, with predictive factors in 237 patients

    Background: Thiopurines (azathioprine and mercaptopurine) remain integral to most medical strategies for maintaining remission in Crohn's disease (CD) and ulcerative colitis (UC). Indefinite use of these drugs is tempered by long-term risks. While clinical relapse is noted frequently following drug withdrawal, there are few published data on predictive factors. Aim: To investigate the success of planned thiopurine withdrawal in patients in sustained clinical remission to identify rates and predictors of relapse. Methods This was a multicentre retrospective cohort study from 11 centres across the UK. Patients included had a definitive diagnosis of IBD, continuous thiopurine use &#8805;3 years and withdrawal when in sustained clinical remission. All patients had a minimum of 12 months follow-up post drug withdrawal. Primary and secondary end points were relapse at 12 and 24 months respectively. Results: 237 patients were included in the study (129 CD; 108 UC). Median duration of thiopurine use prior to withdrawal was 6.0 years (interquartile range 4.4–8.4). At follow-up, moderate/severe relapse was observed in 23% CD and 12% UC patients at 12 months, 39% CD and 26% UC at 24 months. Relapse rate at 12 months was significantly higher in CD than UC (P = 0.035). Elevated CRP at withdrawal was associated with higher relapse rates at 12 months for CD (P = 0.005), while an elevated white cell count was predictive at 12 months for UC (P = 0.007). Conclusion Thiopurine withdrawal in the context of sustained remission is associated with a 1-year moderate-to-severe relapse rate of 23% in Crohn's disease and 12% in ulcerative colitis

    Ethnobotanical investigations on local oilseed species in Burkina Faso revealed that oils from tree oilseeds are frequently used for food, cosmetics and traditional medicine by local people. To test the bio-preservative capacity, the effect of the oilseed extracts on the stabilization of Shea butter was evaluated. Levels of bioactive phyto-components were evaluated through several methods. Some aspects of the quality of oilcakes were evaluated based on their inhibitory capacity on the endogenous digestive enzymes of cattle. For fat stabilizing quality, the extract from Lannea microcarpa Engl. &amp; K. Krause has showed the largest increase (91.47%) in the Rancimat induction time (TIR) of Shea butter. Using DPPH and FRAP methods for prevention or scavenging of free radicals, the results evidenced the highest activity with extracts of L. microcarpa. With respect to the feed quality of the oilcakes, the IC50 obtained with α-amylase and trypsin have showed that the ethanolic extracts of Pentadesma butyracea Sabine (1824) is less toxic for cattle. These results suggest that phenolic compounds extracted from these native oilseeds, especially those of Lannea microcarpa, can be used as biological antioxidants for bio-preservation of edible fats. In addition, the by-products of the oil-extraction can be valorized as nontoxic and protein-rich oilcakes for livestock feed

    Ethnobotanical and Phytochemical Profiling of Medicinal Plants from Burkina Faso Used to Increase Physical Performance

    Background: Some ergogenic medicinal plants are used in exercise and sport in Africa in order to increase sport performance. However, data on their composition and their possible impacts on health are limited. This study was initiated to provide ethnobotanical data on plants traditionally used to optimize physical performance and to perform a qualitative characterization of their main chemical groups. Methods: Ethnobotanical surveys in two communes (Dedougou and Nouna), of the region of Boucle du Mouhoun, Burkina Faso and phytochemical analyses of the most interesting plants were conducted. Results: A total of 50 respondents including traditional hunters dozo, farmers, healers, herbalists, marabouts, etc., were interviewed. Fifty-two species used in the optimization of exercise and sports have been identified. The most cited species were Cassia sieberiana, Tamarindus indica, Annona senegalensis, Gardenia sokotensis, Securidaca longepedunculata, and Ficus tonningii. These plants are known to prevent muscle and skeletal disorders, aches and pains, and mental disorders. The study identified several types of plants including those displaying stimulation, anxiolytic, sedative, adaptogenic, or erythropoietic activities. Phytochemical screening revealed the presence of phenolic compounds, alkaloids, terpenes, and steroids, which are similar molecules families of those of doping molecules. Additionally, TLC screening allowed the characterization of numerous terpene and flavonoid compounds including rutin. Conclusions: The possible structural similarity of the characterized chemical groups of these species with those of doping families raise concerns about the consequences of their consumption. However, the identification of the active molecules of these species remains to be performed in order to predict the real risks associated with their consumption

    Antibacterial activity against β- lactamase producing Methicillin and Ampicillin-resistants <it>Staphylococcus aureus</it>: fractional Inhibitory Concentration Index (FICI) determination

    <p>Abstract</p> <p>Background</p> <p>The present study reports the antibacterial capacity of alkaloid compounds in combination with Methicillin and Ampicillin-resistants bacteria isolated from clinical samples. The resistance of different bacteria strains to the current antibacterial agents, their toxicity and the cost of the treatment have led to the development of natural products against the bacteria resistant infections when applied in combination with conventional antimicrobial drugs.</p> <p>Method</p> <p>The antibacterial assays in this study were performed by using inhibition zone diameters, MIC, MBC methods, the time-kill assay and the Fractional Inhibitory Concentration Index (FICI) determination. On the whole, fifteen Gram-positive bacterial strains (MRSA/ARSA) were used. Negative control was prepared using discs impregnated with 10 % DMSO in water and commercially available Methicillin and Ampicillin from Alkom Laboratories LTD were used as positive reference standards for all bacterial strains.</p> <p>Results</p> <p>We noticed that the highest activities were founded with the combination of alkaloid compounds and conventional antibiotics against all bacteria strains. Then, results showed that after 7 h exposition there was no viable microorganism in the initial inoculums.</p> <p>Conclusion</p> <p>The results of this study showed that alkaloid compounds in combination with conventional antibiotics (Methicillin, Ampicillin) exhibited antimicrobial effects against microorganisms tested. These results validate the ethno-botanical use of <it>Cienfuegosia digitata</it> Cav. (Malvaceae) in Burkina Faso. Moreover, this study demonstrates the potential of this herbaceous as a source of antibacterial agent that could be effectively used for future health care purposes.</p
