    Finite-Difference Time-Domain Simulation for Three-dimensional Polarized Light Imaging

    Three-dimensional Polarized Light Imaging (3D-PLI) is a promising technique to reconstruct the nerve fiber architecture of human post-mortem brains from birefringence measurements of histological brain sections with micrometer resolution. To better understand how the reconstructed fiber orientations are related to the underlying fiber structure, numerical simulations are employed. Here, we present two complementary simulation approaches that reproduce the entire 3D-PLI analysis: First, we give a short review on a simulation approach that uses the Jones matrix calculus to model the birefringent myelin sheaths. Afterwards, we introduce a more sophisticated simulation tool: a 3D Maxwell solver based on a Finite-Difference Time-Domain algorithm that simulates the propagation of the electromagnetic light wave through the brain tissue. We demonstrate that the Maxwell solver is a valuable tool to better understand the interaction of polarized light with brain tissue and to enhance the accuracy of the fiber orientations extracted by 3D-PLI.Comment: 13 pages, 5 figure

    Detailed analysis of Balmer lines in cool dwarf stars

    An analysis of H alpha and H beta spectra in a sample of 30 cool dwarf and subgiant stars is presented using MARCS model atmospheres based on the most recent calculations of the line opacities. A detailed quantitative comparison of the solar flux spectra with model spectra shows that Balmer line profile shapes, and therefore the temperature structure in the line formation region, are best represented under the mixing length theory by any combination of a low mixing-length parameter alpha and a low convective structure parameter y. A slightly lower effective temperature is obtained for the sun than the accepted value, which we attribute to errors in models and line opacities. The programme stars span temperatures from 4800 to 7100 K and include a small number of population II stars. Effective temperatures have been derived using a quantitative fitting method with a detailed error analysis. Our temperatures find good agreement with those from the Infrared Flux Method (IRFM) near solar metallicity but show differences at low metallicity where the two available IRFM determinations themselves are in disagreement. Comparison with recent temperature determinations using Balmer lines by Fuhrmann (1998, 2000), who employed a different description of the wing absorption due to self-broadening, does not show the large differences predicted by Barklem et al. (2000). In fact, perhaps fortuitously, reasonable agreement is found near solar metallicity, while we find significantly cooler temperatures for low metallicity stars of around solar temperature.Comment: 17 pages, 9 figures, to appear in A&

    Empirically Constrained Color-Temperature Relations. II. uvby

    (Abriged) A new grid of theoretical color indices for the Stromgren uvby photometric system has been derived from MARCS model atmospheres and SSG synthetic spectra for cool dwarf and giant stars. At warmer temperatures this grid has been supplemented with the synthetic uvby colors from recent Kurucz atmospheric models without overshooting. Our transformations appear to reproduce the observed colors of extremely metal-poor turnoff and giant stars (i.e., [Fe/H]<-2). Due to a number of assumptions made in the synthetic color calculations, however, our color-temperature relations for cool stars fail to provide a suitable match to the uvby photometry of both cluster and field stars having [Fe/H]>-2. To overcome this problem, the theoretical indices at intermediate and high metallicities have been corrected using a set of color calibrations based on field stars having accurate IRFM temperature estimates and spectroscopic [Fe/H] values. Encouragingly, isochrones that employ the transformations derived in this study are able to reproduce the observed CMDs (involving u-v, v-b, and b-y colors) for a number of open and globular clusters (including M92, M67, the Hyades, and 47Tuc) rather well. Moreover, our interpretations of such data are very similar, if not identical, with those given by VandenBerg & Clem (2003, AJ, 126, 778) from a consideration of BV(RI)c observations for the same clusters. In the present investigation, we have also analyzed the observed Stromgren photometry for the classic Population II subdwarfs, compared our "final" (b-y)-Teff relationship with those derived empirically in a number of recent studies, and examined in some detail the dependence of the m1 index on [Fe/H].Comment: 70 pages, 26 figures.     Enhancing glycan stability via site-selective fluorination: modulating substrate orientation by molecular design

    Single site OH → F substitution at the termini of maltotetraose leads to significantly improved hydrolytic stability towards α-amylase and α-glucosidase relative to the natural compound. To explore the effect of molecular editing, selectively modified oligosaccharides were prepared via a convergent α-selective strategy. Incubation experiments in purified α-amylase and α-glucosidase, and in human and murine blood serum, provide insight into the influence of fluorine on the hydrolytic stability of these clinically important scaffolds. Enhancements of ca. 1 order of magnitude result from these subtle single point mutations. Modification at the monosaccharide furthest from the probable enzymatic cleavage termini leads to the greatest improvement in stability. In the case of α-amylase, docking studies revealed that retentive C2-fluorination at the reducing end inverts the orientation in which the substrate is bound. A co-crystal structure of human α-amylase revealed maltose units bound at the active-site. In view of the evolving popularity of C(sp3)–F bioisosteres in medicinal chemistry, and the importance of maltodextrins in bacterial imaging, this discovery begins to reconcile the information-rich nature of carbohydrates with their intrinsic hydrolytic vulnerabilities

    We present new B, V and I-band photometry for a sample of 43 local subdwarfs with HIPPARCOS parallax errors < 13%, in the metallicity range -1.0 < [Fe/H] < -0.3, which we use to perform main sequence (MS) fitting to the Galactic globular cluster 47 Tuc. This sample is many times larger than those used in previous MS-fitting studies and also enables us to fit in two colour planes, V/(B-V) and V/(V-I). With this enlarged subdwarf sample we investigate whether the current discrepancy in empirical distance estimates for 47 Tuc, arising from recent MS-fitting and white dwarf fitting results, is due to inaccuracies in the MS-fitting method. Comparison of published photometries for 47 Tuc has revealed systematic offsets which mean that the (BV)(B-V) main-line used in previous studies may be too blue by ~ 0.02 mag. We have derived main-lines in V/(B-V) and V/(V-I) from the data of Kaluzny et al. (1998), which we have recalibrated from the `secondary' standards in 47 Tuc of Stetson (2000). Using an assumed cluster reddening of E(B-V)=0.04, our best-fit apparent distance modulus is (m-M)V=13.37 (+0.10)(-0.11) in both colour planes, which implies a cluster age of 11.0 +/-1.4 Gyr and leads to a dereddened distance modulus of (m-M)0=13.25 (+0.06)(-0.07). Comparison with previous work shows that our apparent distance modulus is ~0.2 mag smaller than those derived in previous MS-fitting studies. The difference is accounted for by our preferred cluster reddening and the recalibration of the cluster photometry, which has made the main-line redder by an average of 0.02 mag in (B-V). Independent support for our MS-fitting distance comes from consideration of the Red Clump in the cluster, from which we derive a dereddened distance modulus of (m-M)0=13.31 +/-0.05, which is in agreement with the MS-fitting result.     Rare brain biopsy findings in a first ADEM-like event of pediatric MS: histopathologic, neuroradiologic and clinical features

    Pediatric MS tends to present more often with an acute onset and a polysymptomatic form of the disease, possibly with encephalopathy and large tumefactive lesions similar to those observed in some cases of acute disseminated encephalomyelitis (ADEM), which makes it more difficult to differentiate between an explosive and severe onset of MS vs. ADEM. An ADEM-like first demyelinating event can be the first attack of pediatric MS, but international consensus definitions require two or more non-ADEM demyelinating events for diagnosis of MS. In our patient KIDMUS MRI criteria for MS (Mikaeloff et al. J Pediatr 144:246–252, 2004a; Mikaeloff et al. Brain 127:1942–1947, 2004b) were negative at first attack, but Barkhof criteria for lesion dissemination in space in adults (Barkhof et al. 120:2059–2069, 1997), Callen modified MS-criteria and Callen MS-ADEM criteria for children (Callen et al. Neurology 72:961–967, 2009a; Callen et al. Neurology 72:968–973, 2009b) were positive suggesting pediatric MS. As the clinical course was devastating with non-responsiveness upon high-dose immune modulatory therapy and due to the absence of an alternative diagnosis other than demyelinating disease brain biopsy was performed. Brain biopsy studies or autopsy case reports of fulminant pediatric MS patients are extremely rare. Histopathology revealed an inflammatory demyelinating CNS process with confluent demyelination, indicating the likelihood of a relapsing disease course compatible with an acute to subacute demyelinating inflammatory disease. This pattern was corresponding to the early active multiple sclerosis subtype I of Lucchinetti et al. (Ann Neurol 47(6):707–717, 2000)
