Infant lung function tests as endpoints in the ISIS multicenter clinical trial in cystic fibrosis
BACKGROUND:
The Infant Study of Inhaled Saline (ISIS) in CF was the first multicenter clinical trial to utilize infant pulmonary function tests (iPFTs) as an endpoint.
METHODS:
Secondary analysis of ISIS data was conducted in order to assess feasibility of iPFT measures and their associations with respiratory symptoms. Standard deviations were calculated to aid in power calculations for future clinical trials.
RESULTS:
Seventy-three participants enrolled, 70 returned for the final visit; 62 (89%) and 45 (64%) had acceptable paired functional residual capacity (FRC) and raised volume measurements, respectively. Mean baseline FEV0.5, FEF75 and FRC z-scores were 0.3 (SD: 1.2), -0.2 (SD: 2.0), and 1.8 (SD: 2.0).
CONCLUSIONS:
iPFTs are not appropriate primary endpoints for multicenter clinical trials due to challenges of obtaining acceptable data and near-normal average raised volume measurements. Raised volume measures have potential to serve as secondary endpoints in future clinical CF trials

Talk Like an Electrician: Student Dialogue Mimicking Behavior in an Intelligent Tutoring System
Abstract. Students entering a new field must learn to speak the specialized language of that field. Previous research using automated measures of word overlap has found that students who modify their language to align more closely to a tutor's language show larger overall learning gains. We present an alternative approach that assesses syntactic as well as lexical alignment in a corpus of human-computer tutorial dialogue. We found distinctive patterns differentiating high and low achieving students. Our high achievers were most likely to mimic their own earlier statements and rarely made mistakes when mimicking the tutor. Low achievers were less likely to reuse their own successful sentence structures, and were more likely to make mistakes when trying to mimic the tutor. We argue that certain types of mimicking should be encouraged in tutorial dialogue systems, an important future research direction
MAGESTIC: Magnetically Enabled Structures Using Interacting Coils
In our NIAC Phase I study, awarded September 2011, the MIT Space Systems Lab (MIT SSL) began investigating a new structural and mechanical technique aimed at reducing the mass and increasing the stowed-to-deployed ratio of spacecraft systems. This technique uses the magnetic fields from current passing through coils of high temperature superconductors (HTSs) to support spacecraft structures and deploy them to operational configurations from their positions as stowed inside a launch vehicle fairing. These electromagnetic coils are tethered or hinged together in such a way that their motion in some directions or around some axes is constrained, as in Figure 1. Our Phase II study,awarded in Fall 2012, continued this work on electromagnetic structures, with an added focus on developing a new thermal system, investigating additional, non-structural electromagnet functions, and creating a maturation roadmap and plan for addressing barriers to feasibility of the technology. We now call the project MAGESTIC, or Magnetically Enabled STructures using Interacting Coils
Preliminary Characterization and Analysis of the Designs and Research-Manufacturing Approaches
This report summarizes the results of Phase I of a study entitled, Low-Cost Manufacturing Of Multilayer Ceramic Fuel Cells. The work was carried out by a group called the Multilayer Fuel Cell Alliance (MLFCA) led by NexTech Materials and including Adaptive Materials, Advanced Materials Technologies (AMT), Cobb & Co., Edison Materials Technology Center, Iowa State University, Gas Technology Institute (GTI), Northwestern University, Oak Ridge National Laboratory (ORNL), Ohio State University, University of Missouri-Rolla (UMR), and Wright-Patterson Air Force Base. The objective of the program is to develop advanced manufacturing technologies for making solid oxide fuel cell components that are more economical and reliable for a variety of applications. In the Phase I effort, five approaches were considered: two based on NexTech's planar approach using anode and cathode supported variations, one based on UMR's ultra-thin electrolyte approach, and two based on AMI's co-extrusion technology. Based on a detailed manufacturing cost analysis, all of the approaches are projected to result in a significantly reduced production cost. Projected costs range from 179/kW for planar designs. Development risks were assessed for each approach and it was determined that the NexTech and UMR approaches carried the least risk for successful development. Using advanced manufacturing methods and a proprietary high power density design, the team estimated that production costs could be reduced to $94/kW
Advancing translational research with the Semantic Web
<p>Abstract</p> <p>Background</p> <p>A fundamental goal of the U.S. National Institute of Health (NIH) "Roadmap" is to strengthen <it>Translational Research</it>, defined as the movement of discoveries in basic research to application at the clinical level. A significant barrier to translational research is the lack of uniformly structured data across related biomedical domains. The Semantic Web is an extension of the current Web that enables navigation and meaningful use of digital resources by automatic processes. It is based on common formats that support aggregation and integration of data drawn from diverse sources. A variety of technologies have been built on this foundation that, together, support identifying, representing, and reasoning across a wide range of biomedical data. The Semantic Web Health Care and Life Sciences Interest Group (HCLSIG), set up within the framework of the World Wide Web Consortium, was launched to explore the application of these technologies in a variety of areas. Subgroups focus on making biomedical data available in RDF, working with biomedical ontologies, prototyping clinical decision support systems, working on drug safety and efficacy communication, and supporting disease researchers navigating and annotating the large amount of potentially relevant literature.</p> <p>Results</p> <p>We present a scenario that shows the value of the information environment the Semantic Web can support for aiding neuroscience researchers. We then report on several projects by members of the HCLSIG, in the process illustrating the range of Semantic Web technologies that have applications in areas of biomedicine.</p> <p>Conclusion</p> <p>Semantic Web technologies present both promise and challenges. Current tools and standards are already adequate to implement components of the bench-to-bedside vision. On the other hand, these technologies are young. Gaps in standards and implementations still exist and adoption is limited by typical problems with early technology, such as the need for a critical mass of practitioners and installed base, and growing pains as the technology is scaled up. Still, the potential of interoperable knowledge sources for biomedicine, at the scale of the World Wide Web, merits continued work.</p
Aggregate risk of cardiovascular disease among adolescents perinatally infected with the human immunodeficiency virus
BACKGROUND: Perinatally HIV-infected adolescents may be susceptible to aggregate atherosclerotic cardiovascular disease risk, as measured by the Pathobiological Determinants of Atherosclerosis in Youth (PDAY) coronary arteries and abdominal aorta risk scores, as a result of prolonged exposure to HIV and antiretroviral therapy.
METHODS AND RESULTS: Coronary arteries and abdominal aorta PDAY scores were calculated for 165 perinatally HIV-infected adolescents, using a weighted combination of modifiable risk factors: dyslipidemia, cigarette smoking, hypertension, obesity, and hyperglycemia. Demographic and HIV-specific predictors of scores ≥1 were identified, and trends in scores over time were assessed. Forty-eight percent and 24% of the perinatally HIV-infected adolescents had coronary arteries and abdominal aorta scores ≥1, representing increased cardiovascular disease risk factor burden. Significant predictors of coronary arteries scores ≥1 included male sex, history of an AIDS-defining condition, longer duration of use of a ritonavir-boosted protease inhibitor, and no prior use of tenofovir. Significant predictors of abdominal aorta scores ≥1 included suppressed viral load, history of an AIDS-defining condition, and longer duration of boosted protease inhibitor use. No significant changes in coronary arteries and abdominal aorta risk scores were observed over the 4-year study period.
CONCLUSIONS: A substantial proportion of perinatally HIV-infected youth have high PDAY scores, reflecting increased aggregate atherosclerotic cardiovascular disease risk factor burden. High scores were predicted by HIV disease severity and boosted protease inhibitor use. PDAY scores may be useful in identifying high-risk youth who may benefit from early lifestyle or clinical interventions
