    Duodenal intraepithelial lymphocytes of children with cow milk allergy preferentially bind the glycan-binding protein galectin-3

    A breakdown in intestinal homeostasis results in inflammatory bowel diseases including coeliac disease and allergy. Galectins, evolutionarily conserved beta-galactoside-binding proteins, can modulate immune-epithelial cell interactions by influencing immune cell fate and cytokine secretion. In this study we investigated the glycosylation signature, as well as the regulated expression of galectin-1 and -3 in human duodenal samples of allergic and non-allergic children. Whereas galectin-1 was predominantly localized in the epithelial compartment (epithelial cells and intraepithelial lymphocytes) and the underlying lamina propria (T cells, macrophages and plasma cells), galectin-3 was mainly expressed by crypt epithelial cells and macrophages in the lamina propria. Remarkably, expression of these galectins was not significantly altered in allergic versus non-allergic patients. Investigation of the glycophenotype of the duodenal inflammatory microenvironment revealed substantial alpha2-6-linked sialic acid bound to galactose in lamina propria plasma cells, macrophages and intraepithelial lymphocytes and significant levels of asialo core 1 O-glycans in CD68+ macrophages and enterocytes. Galectin-1 preferentially bound to neutrophils, plasma cells and enterocytes, while galectin-3 binding sites were mainly distributed on macrophages and intraepithelial lymphocytes. Notably, galectin-3, but not galectin-1 binding, was substantially increased in intraepithelial gut lymphocytes of allergic patients compared to non-allergic subjects, suggesting a potential role of galectin-3-glycan interactions in shaping epithelial-immune cell connections during allergic inflammatory processes.Fil: Mercer, Natalia.     Inferring the parallax of Westerlund 1 from Gaia DR2

    Westerlund 1 (Wd1) is potentially the largest star cluster in the Galaxy. That designation critically depends upon the distance to the cluster, yet the cluster is highly obscured, making luminosity-based distance estimates difficult. Using {\it Gaia} Data Release 2 (DR2) parallaxes and Bayesian inference, we infer a parallax of 0.35−0.06+0.070.35^{+0.07}_{-0.06} mas corresponding to a distance of 2.6−0.4+0.62.6^{+0.6}_{-0.4} kpc. To leverage the combined statistics of all stars in the direction of Wd1, we derive the Bayesian model for a cluster of stars hidden among Galactic field stars; this model includes the parallax zero-point. Previous estimates for the distance to Wd1 ranged from 1.0 to 5.5 kpc, although values around 5 kpc have usually been adopted. The {\it Gaia} DR2 parallaxes reduce the uncertainty from a factor of 3 to 18\% and rules out the most often quoted value of 5 kpc with 99\% confidence. This new distance allows for more accurate mass and age determinations for the stars in Wd1. For example, the previously inferred initial mass at the main-sequence turn-off was around 40 M⊙_{\odot}; the new {\it Gaia} DR2 distance shifts this down to about 22 M⊙_{\odot}. This has important implications for our understanding of the late stages of stellar evolution, including the initial mass of the magnetar and the LBV in Wd1. Similarly, the new distance suggests that the total cluster mass is about four times lower than previously calculated.Comment: 14 pages, 10 figure

    The Nature of Nearby Counterparts to Intermediate Redshift Luminous Compact Blue Galaxies I. Optical/H I Properties and Dynamical Masses

    We present single-dish H I spectra obtained with the Green Bank Telescope, along with optical photometric properties from the Sloan Digital Sky Survey, of 20 nearby (D < 70 Mpc) Luminous Compact Blue Galaxies (LCBGs). These ~L*, blue, high surface brightness, starbursting galaxies were selected with the same criteria used to define LCBGs at higher redshifts. We find these galaxies are gas-rich, with M(HI) ranging from 5*10^8 to 8*10^9 M_sun, and M(HI)/L_B ranging from 0.2 to 2 M_sun/L_sun, consistent with a variety of morphological types of galaxies. We find the dynamical masses (measured within R_25) span a wide range, from 3*10^9 to 1*10^11 M_sun. However, at least half have dynamical mass-to-light ratios smaller than nearby galaxies of all Hubble types, as found for LCBGs at intermediate redshifts. By comparing line widths and effective radii with local galaxy populations, we find that LCBGs are consistent with the dynamical mass properties of Magellanic (low luminosity) spirals, and the more massive irregulars and dwarf ellipticals, such as NGC 205.Comment: 33 pages, 8 figures, accepted by Ap

    We investigate the dynamics of a continuous atom laser based on the merging of independently formed atomic condensates. In a first attempt to understand the dynamics of the system, we consider two independent elongated Bose-Einstein condensates which approach each other and focus on intermediate inter-trap distances so that a two-mode model is well justified. In the framework of a mean-field theory, we discuss the quasi steady-state population of the traps as well as the energy distribution of the outcoupled atoms.Comment: 21 pages, 9 figure, to appear in J.     Radiochemical Separation and Measurement by Mass Spectrometry with Magnetic Sector with Inductively Coupled Plasma source (ICP-SFMS) of Plutonium Isotopes in Soil Samples

    The aim of this work is twofold: to optimize the radiochemical separation of Plutonium (Pu) from soil samples, and to measure the Pu concentration. Soil samples were prepared using acid digestion assisted by microwaves; then, Pu purification was carried out with Pu AG1X8 resin. Pu isotopes were measured using Mass Spectrometry with Magnetic Sector with Inductively Coupled Plasma source (ICP-SFMS). In order to reduce the interference due to the presence of 238UH+ in the samples a desolvation system (Apex) was used. The limit of detection (LOD) of Pu was determined. The efficiency of Pu recovery from soil samples varies from 70 to 93%

    West Nile virus infection in the golden hamster (Mesocricetus auratus): a model for West Nile encephalitis.

    This report describes a new hamster model for West Nile (WN) virus encephalitis. Following intraperitoneal inoculation of a New York isolate of WN virus, hamsters had moderate viremia of 5 to 6 days in duration, followed by the development of humoral antibodies. Encephalitic symptoms began 6 days after infection; about half the animals died between the seventh and 14th days. The appearance of viral antigen in the brain and neuronal degeneration also began on the sixth day. WN virus was cultured from the brains of convalescent hamsters up to 53 days after initial infection, suggesting that persistent virus infection occurs. Hamsters offer an inexpensive model for studying the pathogenesis and treatment of WN virus encephalitis

    Symbiotic Stars in OGLE Data I. Large Magellanic Cloud Systems

    Symbiotic stars are long-orbital-period interacting-binaries characterized by extended emission over the whole electromagnetic range and by complex photometric and spectroscopic variability. In this paper, the first of a series, we present OGLE light curves of all the confirmed symbiotic stars in the Large Magellanic Cloud, with one exception. By careful visual inspection and combined time-series analysis techniques, we investigate for the first time in a systematic way the photometric properties of these astrophysical objects, trying in particular to distinguish the nature of the cool component (e.g., Semi-Regular Variable vs. OGLE Small-Amplitude Red Giant), to provide its first-order pulsational ephemerides, and to link all this information with the physical parameters of the binary system as a whole. Among the most interesting results, there is the discovery of a 20-year-long steady fading of Sanduleak's star, a peculiar symbiotic star known to produce the largest stellar jet ever discovered. We discuss by means of direct examples the crucial need for long-term multi-band observations to get a real understanding of symbiotic and other interacting binary stars. We eventually introduce BOMBOLO, a multi-band simultaneous imager for the SOAR 4m Telescope, whose design and construction we are currently leading.Comment: 16 pages, 4 Tables, 12 Figures.
