    The cellular distribution of Na+/H+ exchanger regulatory factor 1 is determined by the PDZ-I domain and regulates the malignant progression of breast cancer

    The oncogenic role of ectopic expression of Na+/H+ exchanger regulatory factor 1 (NHERF1) was recently suggested. Here, we show that NHERF1 was upregulated in high grades compared with low grades. Increased NHERF1 expression was correlated with poor prognosis and poor survival. NHERF1 expression was higher in the nucleus of cancer cells than in contiguous non- mammary epithelial cells. A novel mutation, namely NHERF1 Y24S, was identified in human breast cancer tissues and shown to correspond to a conserved residue in the PDZ-I domain of NHERF1. Truncation and mutation of the PDZ-I domain of NHERF1 increased the nuclear distribution of the NHERF1 protein, and this redistribution was associated with the malignant phenotype of breast cancer cells, including growth, migration, and adhesion. The present results suggest a role for NHERF1 in the progression of breast cancer mediated by the nuclear distribution of the NHERF1 protein, as determined by the truncation or key site mutation of the PDZ-I domain

    A novel NHERF1 mutation in human breast cancer inactivates inhibition by NHERF1 protein in EGFR signaling

    Background: Na+/H+ exchanger regulatory factor 1 (NHERF1) has been reported to interact with many cancer-related proteins. We recently identified a novel NHERF1 mutation (E43G) in breast tumours. Materials and Methods: The candidates of NHERF1 mutation were identified in breast cancer tissues by polymerase chain reaction and DNA sequencing. Wild-type NHERF1 and E43G mutation were expressed in NHERF1-knockdown cells (MCF7ΔNHERF1) and low-NHERF1-expressing cells (SKMES-1). The effects of mutated NHERF1 on cell functions were examined using in vitro methods. Glutathione S-transferase pull-down assays and western blotting were performed to study the effects of NHERF1 mutation on its interaction with cancer-related proteins. Results: Compared to wild-type NHERF1, expression of the mutated NHERF1 failed to suppress malignant traits in cancer cells, attenuated interaction of NHERF1 protein with epidermal growth factor receptor (EGFR), and inactivated its inhibition of EGF-induced Akt and extracellular regulated protein kinases (ERK) activation. Conclusion: The results show the causal role of NHERF1 in the regulation of the EGFR pathway and the progression of breast cancer

    Modeling Dynamic Heterogeneous Graph and Node Importance for Future Citation Prediction

    Accurate citation count prediction of newly published papers could help editors and readers rapidly figure out the influential papers in the future. Though many approaches are proposed to predict a paper's future citation, most ignore the dynamic heterogeneous graph structure or node importance in academic networks. To cope with this problem, we propose a Dynamic heterogeneous Graph and Node Importance network (DGNI) learning framework, which fully leverages the dynamic heterogeneous graph and node importance information to predict future citation trends of newly published papers. First, a dynamic heterogeneous network embedding module is provided to capture the dynamic evolutionary trends of the whole academic network. Then, a node importance embedding module is proposed to capture the global consistency relationship to figure out each paper's node importance. Finally, the dynamic evolutionary trend embeddings and node importance embeddings calculated above are combined to jointly predict the future citation counts of each paper, by a log-normal distribution model according to multi-faced paper node representations. Extensive experiments on two large-scale datasets demonstrate that our model significantly improves all indicators compared to the SOTA models.Comment: Accepted by CIKM'202

    Development of an axial flux MEMS BLDC micromotor with increased efficiency and power density

    This paper presents a rigorous design and optimization of an axial flux microelectromechanical systems (MEMS) brushless dc (BLDC) micromotor with dual rotor improving both efficiency and power density with an external diameter of only around 10 mm. The stator is made of two layers of windings by MEMS technology. The rotor is developed by film permanent magnets assembled over the rotor yoke. The characteristics of the MEMS micromotor are analyzed and modeled through a 3-D magnetic equivalent circuit (MEC) taking the leakage flux and fringing effect into account. Such a model yields a relatively accurate prediction of the flux in the air gap, back electromotive force (EMF) and electromagnetic torque, whilst being computationally efficient. Based on 3-D MEC model the multi-objective firefly algorithm (MOFA) is developed for the optimal design of this special machine. Both 3-D finite element (FE) simulation and experiments are employed to validate the MEC model and MOFA optimization design

    Separate density and viscosity measurements of unknown liquid using quartz crystal microbalance

    Aqueous liquids have a wide range of applications in many fields. Basic physical properties like the density and the viscosity have great impacts on the functionalities of a given ionic liquid. For the millions kinds of existing liquids, only a few have been systematically measured with the density and the viscosity using traditional methods. However, these methods are limited to measure the density and the viscosity of an ionic liquid simultaneously especially in processing micro sample volumes. To meet this challenge, we present a new theoretical model and a novel method to separate density and viscosity measurements with single quartz crystal microbalance (QCM) in this work. The agreement of experimental results and theocratical calculations shows that the QCM is capable to measure the density and the viscosity of ionic liquid
