    Twenty years of geomagnetic field observations at Mario Zucchelli Station (Antarctica)

    During the 1986-87 austral summer a geomagnetic observatory was installed at Terra Nova Bay. During the first years both geomagnetic field time variation monitoring and absolute measurements were carried out only during summer. Since 1991 variometer measurements are automatically performed during the whole year, while absolute measurements are still performed only during summer. In spite of this, interesting observations were obtained during the life (quite long for Antarctica) of the geomagnetic observatory. In particular in this paper some of the most relevant results are briefly presented: studies about secular variation, daily variation (and its dependence from solar cycle and seasons) and geomagnetic higher frequency variations, such as geomagnetic pulsations

    Para-infectious brain injury in COVID-19 persists at follow-up despite attenuated cytokine and autoantibody responses

    To understand neurological complications of COVID-19 better both acutely and for recovery, we measured markers of brain injury, inflammatory mediators, and autoantibodies in 203 hospitalised participants; 111 with acute sera (1–11 days post-admission) and 92 convalescent sera (56 with COVID-19-associated neurological diagnoses). Here we show that compared to 60 uninfected controls, tTau, GFAP, NfL, and UCH-L1 are increased with COVID-19 infection at acute timepoints and NfL and GFAP are significantly higher in participants with neurological complications. Inflammatory mediators (IL-6, IL-12p40, HGF, M-CSF, CCL2, and IL-1RA) are associated with both altered consciousness and markers of brain injury. Autoantibodies are more common in COVID-19 than controls and some (including against MYL7, UCH-L1, and GRIN3B) are more frequent with altered consciousness. Additionally, convalescent participants with neurological complications show elevated GFAP and NfL, unrelated to attenuated systemic inflammatory mediators and to autoantibody responses. Overall, neurological complications of COVID-19 are associated with evidence of neuroglial injury in both acute and late disease and these correlate with dysregulated innate and adaptive immune responses acutely

    Preservation of fresh coconut gratings by hurdle technique

    Hurdle technology employs the intelligent combination of different preservation factors to achieve mild but reliable preservation effects. Fresh coconut gratings treated with different combinations of hurdles such as acidulants, humectants, preservatives, antioxidants, a mild heat treatment and packaging were evaluated for changes in physico-chemical and sensory properties. It is possible to preserve the grating for four weeks in laminated polythene packages at 5°C in combination with 3% sodium chloride, 0.3% citric acid, 0.009% sodium citrate, 0.02% butvlated hydroxvanisole.C

    Adhering to the educational reforms implemented on national level on the recommendation of the presidential task force, The Faculty of Management Studies and Commerce (FMSC), University of Sri Jayewardenepura (USJ) was restructured in the year 2000. This effort comprised of the achievement of objectives to face the challenges of the new millennium, specially to address the gap created due to the paradigm shift from labor-intensive industry to knowledge based technology. How did the faculty members of FMSC evaluate the implementation phase of the restructuring program and its study effectiveness were studied in this attempt. Structured questionnaires were administered to fifty-four senior lecturers that have taught in both systems and the response rate was 66%. There was 50% response rate for each system for the question on the suitability of new and old systems for the present Sri Lankan universities with respect to teaching, learning, assessments and student-teacher relationships. This follows the finding that most of the objectives of restructuring are being achieved at a moderate level. Accordingly students are benefited through rendering the maximum service of the expert teacher in respective areas of specialization, dissemination of the same level of knowledge and availability of different specialization areas. Establishment of service departments to develop a research culture at FMSC has been achieved at a moderate level but lack of freedom to get involved in such activities hinders the above trend. Teaching in the old system is preferred by 60% of the respondents along with a better position on student learning. Student evaluations too are more successful in the old system than in the new system. Findings strongly support the view that student-teacher relationships in with the new system are at a stake.     Laparoscopic Bilateral Adrenalectomy in a Young Female Patient with Recurrent Cushing’s Disease

    Introduction. Synchronous bilateral adrenalectomy is undertaken less often due to numerous perioperative challenges and rare circumstances of patients needing this procedure. Bilateral adrenalectomy is an important second-line option for patients with persistent or recurrent hypercortisolism following transsphenoidal surgery for Cushing’s disease. Here, we present a challenging case of synchronous laparoscopic bilateral adrenalectomy for a young female patient with recurrent Cushing’s disease and fertility wishes. Case Presentation. A 21-year-old recently married patient who was diagnosed with Cushing’s disease with a pituitary microadenoma had undergone two attempts of transsphenoidal excision of the pituitary tumour. Follow-up evaluation showed an unresectable residual tumour with invasion of the intracavernous part of the left internal carotid artery. As the patient had the hypothalamic-pituitary-ovarian axis intact with strong fertility wishes, she was offered bilateral adrenalectomy instead of radiotherapy. She was prepared for the surgery with close perioperative support from the endocrinology and anaesthesia teams. Intravenous hydrocortisone infusion was started at the induction of anaesthesia. Transperitoneal approach was used with the patient positioned in left and right lateral positions for right and left glands, respectively. A meticulous surgical technique was used for the identification of adrenal veins to clip them before division followed by handling of the glands. The patient had minimal haemodynamic disturbances during surgery. Intraoperative blood loss was less than 100 ml, and operative time was 220 minutes. She had a gradual recovery following postoperative respiratory distress due to basal atelectasis and consolidation. Cortisol levels were less than 20 nmol/L postoperatively, suggesting successful surgical intervention. Two months after surgery, she continued on maintenance therapy of oral hydrocortisone and fludrocortisone and was encouraged to go ahead with pregnancy. Conclusion. Although bilateral adrenalectomy is considered a high-risk procedure, these risks can be mitigated and performed safely while maintaining close multidisciplinary perioperative support

    Combining BET and HDAC inhibitors synergistically induces apoptosis of melanoma and suppresses AKT and YAP signaling

    Histone acetylation marks have an important role in controlling gene expression and are removed by histone deacetylases (HDACs). These marks are read by bromodomain and extra-terminal (BET) proteins and novel inhibitiors of these proteins are currently in clinical development. Inhibitors of HDAC and BET proteins have individually been shown to cause apoptosis and reduce growth of melanoma cells. Here we show that combining the HDAC inhibitor LBH589 and BET inhibitor I-BET151 synergistically induce apoptosis of melanoma cells but not of melanocytes. Induction of apoptosis proceeded through the mitochondrial pathway, was caspase dependent and involved upregulation of the BH3 pro-apoptotic protein BIM. Analysis of signal pathways in melanoma cell lines resistant to BRAF inhibitors revealed that treatment with the combination strongly downregulated anti-apoptotic proteins and proteins in the AKT and Hippo/YAP signaling pathways. Xenograft studies showed that the combination of inhibitors was more effective than single drug treatment and confirmed upregulation of BIM and downregulation of XIAP as seen in vitro. These results support the combination of these two classes of epigenetic regulators in treatment of melanoma including those resistant to BRAF inhibitors
